A new individualization marker of semen: deoxyribonuclease I (DNase I) polymorphism.

We describe a method for obtaining specific and reproducible deoxyribonuclease I (DNase I) typing from liquid semen. Isoelectric focusing of the enzymes on polyacrylamide gel (IEF-PAGE, pH 3.5-5) was accomplished using a 0.5-mm thick gel. The separated isozymes were visualized by a new activity staining method, dried agarose film-overlay (DAFO). Pretreatment of semen samples with neuraminidase markedly enhanced the isozyme-band resolution and sensitivity. The method was simple and reliable, with high resolution and sensitivity. The DNase I types in semen samples were correlated with the types found in corresponding blood and urine samples. DNase I typing could therefore provide an additional discriminant characteristic in the forensic examination of semen.     

A novel multiplex PCR system consisting of Y-STRs DYS441, DYS442, DYS443, DYS444, and DYS445

We have developed a new sensitive multiplex PCR system consisting of five male-specific and polymorphic tetranucleotide STRs--DYS441 (GDB: 10013873), DYS442 (GDB: 10030304), DYS443 (GDB: 10807127), DYS444 (GDB: 10807128), and DYS445 (GDB: 10807129) on the Y chromosome. Fifty pg DNA per 10 microL reaction volume was required for the correct typing of five STRs. Using this system, the five Y-STRs were correctly typed from blood and semen stains that had been stored for several years at room temperature.     

`Civil society' in Japanese politics: Implications for contemporary political research

Abstract. This article presents a definitional and historical summary of 'civil society' as a western concept, and then traces how it was used in Japanese political theory and practice. 'Civil society' discourse became a familiar term amongst post–war Japanese historians and social scientists in the period 1945–1970, and thus preceded the recent international proliferation of 'civil society' literature in the 'west' from the 1970s onwards. 'Civil society' discourse was politicised in practice in Japan in the 1950s and 1960s, principally in opposition movements wanting to encourage political participation by ordinary citizens. While some 'civil society' discourse in Japan was based on an idealisation of 'western' ideas and practice, it is also the case that the individualism and democratisation implied in 'civil society' discourse has been very differently understood in 'western' countries and very unevenly instituted in their political practice. A simple East–West frame makes significant similarities between Japanese ideas and practice and 'western' concepts and politics disappear, and additionally causes significant differences within the 'west' regarding individualism and democratisation to become invisible.     

Adolescents’ Daily Romantic Experiences and Negative Mood: A Dyadic,Intensive Longitudinal Study

Romantic relationships, although increasingly normative during adolescence, also present unique developmental challenges that can portend psychological difficulties. Underlying these difficulties may be the degree to which daily romantic transactions potentiate fluctuations in negative mood. The present study examined associations between adolescents’ daily romantic relationship experiences and their same-day negative affective states (i.e., fluctuations in high-arousal, aversive mood). Using a dyadic ecological momentary assessment (EMA) design, this study followed an ethnically and socioeconomically diverse sample of 98 adolescent romantic couples twice weekly for 12 weeks (n?=?196 individuals; M_age?=?16.74 years, SD?=?0.90; 45% Latina/o, 45% White; 55% receiving free or reduced meals). The results indicated that various daily romantic experiences (e.g., conflict, feelings about the relationship) predicted greater same-day negative affect. Beyond the effects of these romantic experiences, adolescent couples were also synchronized in their fluctuating negative affective states, evidencing the presence of emotional contagion. Overall, the findings indicate the salience of romantic relationships in the everyday lives of adolescents.

GC polymorphism detected in human urine by isoelectric focusing and immunoblotting

Genetic polymorphism of GC (vitamin D-binding protein) in human urine was revealed by isoelectric focusing and immunoblotting on thin-layer polyacrylamide gels containing 2 M urea. Urine samples from 530 unrelated Japanese from the Fukui district, being only 1-2 ml of original urine, were examined, and correct GC typing was achieved by comparison with the results of direct grouping using plasma. Six common and twelve rare phenotypes were observed. The frequencies of the genes were 0.473 for GC*1F, 0.241 for GC*1S, 0.254 for GC*2, and 0.032 for the total of six rare alleles.     

Transferrin subtyping of bloodstains and semen using isoelectric focusing and immunoblotting

Transferrin (TF) subtyping was carried out on bloodstains that had been made on cotton sheeting and stored under a variety of conditions ranging from −20°C to +37°C. The time limit of detection was longer than 54 weeks after dry storage under each condition. Moreover the correlation between isoprotein types of the TF in blood and semen samples from the same individual was determined in 103 men. All three TF common types and two rare types in all semen samples correlated with the type found in the corresponding blood sample. A combination of isoelectric focusing separation and immuno-enzyme-linked detection may prove to be very useful for forensic TF subtyping.

A simple method of DNA extraction and STR typing from urine samples using a commercially available DNA/RNA extraction kit

We devised a simple DNA extraction procedure suitable for STR typing of urine sample. Use of a commercially available DNA/RNA extraction kit equipped with a silica-gel-based membrane made it possible to omit the recovery of urinary nucleated cells by sedimentation before the extraction. Successful genotyping of the TH01, HumTPO and multiplex STRs was achieved using aliquots of urine as small as 100 microL. Furthermore, application of this DNA extraction procedure to frozen urine samples provided STR allele results comparable to results obtained from fresh samples. Therefore, this extraction procedure is considered to be effective for STR typing of urine samples in both the frozen and aqueous state. Furthermore, addition of sodium azide to fresh urine samples prolonged their storage duration even at room temperature.
