益气养阴方抗血管生成作用及对肿瘤细胞VEGF及MMP2表达的影响

目的观察益气养阴方抗血管生成作用及对肿瘤细胞血管内皮生长因子(vascular endothelialgrowth factor,VEGF)和基质金属蛋白酶(matrix metalloproteinase,MMP)2表达的效应。方法制备接种肿瘤细胞的绒毛尿囊膜(chorioallantoic membrane,CAM)模型,采用血清药理学方法制备益气养阴方含药血清。观察含药血清对肿瘤细胞诱导CAM血管生成的影响,并与9.0 g/L氯化钠注射液(normal saline,NS)血清组对照。另外,用U14子宫颈癌细胞悬液经肌肉接种小鼠,观察小鼠实体瘤质量、抑瘤率、肺部转移率、肺部转移瘤生长情况。应用免疫组织化学及其组织形态学定量分析方法对VEGF和MMP2在肿瘤细胞中的表达进行研究。结果益气养阴方含药血清可抑制肿瘤细胞诱导CAM血管生成,与NS组比较差异具有显著性。益气养阴方组肺部转移结节较模型组明显减少,VEGF和MMP2的表达均明显低于对照组,差异有显著性。结论益气养阴方对肿瘤血管形成有明显抑制作用,降低VEGF和MMP2的表达可能是其抑制肿瘤血管形成的主要机制之一。
