Abstract: | Objective:To study functional changes in the activity of respiratory chain enzyme of mitochondrion isolated from the liver of aged rats and the effects of 941 Recipe(941R,a traditional medicine formula)on it.Method:The liver mitochondrion were isolated from rats aged 4 months(young group),24 months(senescent group)and 941 group respectively by differential centrifugation;the determination of mitochondria population in rat hepatic cell was obtained quantitatively from measurement of mitochondria protein by a bruit method;NADH oxides (NADHOX),succinct oxides(SUCOX),and the content of cytochrome a,b,c,and c 1(Cyt a,b,c and c 1)were evaluated by the differential spectra(dithionite-reduced minus ferricyanide-oxidized).Result:In comparison with the young group,in senescent rats there was a decline in mitochondria population;The activities of these oxidizes,however,were significantly increased,and the content of cytochrome,especially Cyt b,c 1,was markedly increased;the parameters cited above in 941R group were significantly elevated over those in young and senescent rats.Conclusion:These results suggest that the enhancement of respiratory enzymatic activities of the liver mitochondrion from aged rats appears to be some compensatory mechanism by witch increase workload of the remaining intact population of mitochondrion,and 941R more efficiently improves the compensatory mechanism. |
