甘青虎耳草黄酮对H22肝癌小鼠抗肿瘤作用的研究

摘    要:为探究甘青虎耳草黄酮抑制小鼠H22肝癌原位移植瘤的作用。给小鼠肝接种H22细胞建立肝癌原位移植瘤模型,并随机分组:模型组、环磷酰胺组、甘青虎耳草黄酮剂量组(低、中、高剂量组)和对照组。治疗期间,观察各组小鼠生命体征和生存时间;停药次日,测定抑瘤率、小鼠血清谷丙转氨酶(ALT)、天门冬氨酸氨基转移酶(AST)、血清白蛋白(ALB)、白蛋白/球蛋白比值(A/G)、碱性磷酸酶(ALP)、γ-谷氨酰转移酶(GGT)和岩藻糖苷酶(AFU)水平以及肝组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量。结果显示,各剂量组ALT和AST低于模型组(P<0.05),ALB及A/G比值高于模型组(P<0.05),剂量组ALP、GGT、AFU活性明显低于模型组(P<0.05),其中,甘青虎耳草黄酮各剂量组MDA含量显著低于模型组(P<0.05),SOD、GSH-Px活性显著高于模型组。上述结果表明,甘青虎耳草黄酮能够显著抑制原位移植瘤生长,具有显著抑制肝癌的作用。

Abstract:The inhibited effect of the flavonoids obtained from Saxifraga tangutica Engl.(S.taugutica flavonoids)on the transplanted H22 liver tumor in mice was explored.The tumor model groop was established in vivo by gavaging the mice H22 cell and grouping randomly.The tumor-bearing mice were divided into control group,cyclophosphamide group,and small,medium and large S.taugutica flavonoid dose groups.The flavonoids were gavaged for the does groops.Vital signs and survival time were investigated during the administration period.The indexes were examined at the next day of drug discontinuance,which includ the tumor inhibition rate,the levels of the serum alanine aminotransferase(ALT),the aspartate aminotransferase(AST),the serum albumin(ALB),the alkaline phosphatase(ALP),the fucosidase(AFU),and the gamma-glutamine Acyltransferase(GGT),the ratio of albumin to globulin(A/G),the activities of the liver tissue erythrocuprein(SOD)and the glutathione peroxidase(GSH-Px),as well as the content of malonaldehyde(MDA).For all of the dose groups,the results showd that the levels of ALT and AST,the activity of ALP,GGT and AFU were significantly lower than the model one(P<0.05);however,the level of ALB and the ratio of A/G were higher than the model one(P<0.05).For the different S.taugutica flavonoids dose groups,the MDA content was significantly lower than the model one(P<0.05);the activities of the SOD and GSH-Px were significantly higher than the model one.In conclusion,the growth of the transplanted tumor was inhibited significantly by the S.taugutica flavonoids,indicating that the S.taugutica flavonoids displays an excellent antitumor effect in vivo.
