摘    要:目的 探究云南不明原因猝死案件事发当地隐花青鹅膏的毒性及毒理机制.方法 采集事发地的隐花青鹅膏,经基因测序鉴定种属后制成干粉和熬煮物,SD大鼠灌胃染毒,提血清测定肝、肾功能以及心肌酶谱的生化指标.灌胃染毒7次后,取大鼠心肌、肝脏和肾脏组织进行HE染色和透射电镜观察.最后运用SPSS 21.0统计学软件分析数据.结果 高...

Abstract:Objective To investigate the toxicity of Amanita manginiana in Yunnan sudden unexplained death area through SD rats.Methods The Amanita lucidum from the site of the incident was collected.After the species were identified by gene sequencing,it was made to dry powder and boiled product.SD rats were gavaged with poison,and serum was collected to determine the biochemical indicators of liver and kidney function and myocardial enzyme spectrum.After intragastric administration for 7 times,the rat myocardium,liver and kidney tissues were taken for HE staining and transmission electron microscopy observation.Finally,SPSS 21.0 statistical software was used to analyze the data.Results The rats in the two experimental groups with high-dose intragastric administration showed symptoms of decreased activity,listlessness,and loss of appetite after three times of intragastric administrations.After intragastric administration,uric acid(Ua),Aspartate aminotransferase(AST)and alanine aminotransferase(ALT)were significantly increased in all experimental groups(P<0.05);lactate dehydrogenase(LDH),α-hydroxybutyrate dehydrogenase(α-HBDH),creatine kinase(CK)and creatine kinase isoenzyme(CK-MB)were significantly increased in the two high-dose experimental groups(P<0.05).HE staining showed that myocardial fiber rupture and renal tubular epithelial cell edema were found in all experimental groups.Hepatocyte edema was obvious in boiled food group,steatosis and necrosis of hepatocytes were obvious in dry powder group,and myocardium,hepatocytes and renal tubules obviously changed in in high dose group as well.Results from transmission electron microscopy indicated that the mitochondria of liver cells and rough endoplasmic reticulum in each experimental group were damaged,and the mitochondria of myocardial cells were damaged as well.The dry powder group was also accompanied by an increase in lysosomes and lipid droplets.Meanwhile,some myocardial fibrils were broken.Conclusion The Amanita manginiana in YNSUD area has obvious hepatotoxicity and myocardial toxicity to SD rats.Boiling can not detoxify it,and eating has certain safety risks.Its myocardial toxicity may be one of the factors causing the occurrence of YNSUD.
