桃红四物汤对产后血瘀证大鼠血清溶血磷脂酸的影响

摘    要:目的 探讨桃红四物汤对产后血瘀证大鼠血清溶血磷脂酸(lysophosphatidic acid,LPA)水平的影响。方法 采用米非司酮(8.3 mg/kg)和米索前列醇(0.1 mg/kg)灌胃早孕鼠,复制血瘀证大鼠模型,将模型复制成功的大鼠随机分为模型组、益母草颗粒组(4.3 g/kg)及桃红四物汤高(18.0 g/kg)、中(9.0 g/kg)、低(4.5 g/kg)剂量组,并设立正常对照组;测定血液流变学指标以判定桃红四物汤治疗血瘀证的疗效,采用酶联免疫吸附试验测定血清LPA含量。结果 模型组全血黏度、血浆黏度、血清LPA均较正常对照组显著增高(P<0.05,或P<0.01);桃红四物汤中剂量组及益母草颗粒组大鼠全血低切变率黏度和高切变率黏度显著降低(P<0.05),各药物组大鼠血浆黏度显著降低(P<0.05,或P<0.01);桃红四物汤中剂量组及益母草颗粒组大鼠LPA水平显著降低(P<0.05)。结论 桃红四物汤祛瘀作用显著,其治疗作用可能与降低血清LPA的水平有关。

Effect of Taohong Siwu Decoction on Serum Level of Lysophosphatidic Acid in Rats with Postpartum Blood Stasis Syndrome

Abstract:Objective To investigate the effect of Taohong Siwu Decoction (TSD) on the serum level of lysophosphatidic acid (LPA) in rats with postpartum blood stasis syndrome (BSS). Methods The rats in early pregnancy were administered mifepristone (8.3 mg/kg) and misoprostol (0.1 mg/kg) by gavage to induce a rat model of BSS. These rats were randomly divided into model group, Herba Leonuri (4.3 g/kg) group, high-dose TSD (18.0 g/kg) group, middle-dose TSD (9.0 g/kg) group, low-dose TSD (4.5 g/kg) group, and normal control group. The hemorheological indices were measured to determine the therapeutic efficacy of TSD in the treatment of BSS. The serum level of LPA was measured by enzyme-linked immunosorbent assay. Results Compared with the normal control group, the model group showed significantly increased blood viscosity, plasma viscosity, and serum level of LPA (P<0.05 or P<0.01). Compared with the model group, the middle-dose TSD group and Herba Leonuri group showed significantly decreased blood viscosity at low shear rate and blood viscosity at high shear rate (P<0.05); all medication groups showed significantly decreased plasma viscosity (P<0.05 or P<0.01); the middle-dose TSD group and Herba Leonuri group showed significantly decreased serum level of LPA (P<0.05). Conclusion TSD has marked stasis-removing effect, which may be related to its ability to decrease serum level of LPA.
