抵当陷胸汤对糖尿病大鼠肝脏TGF-β1/Smad3信号通路的影响

摘    要:目的观察抵当陷胸汤通过调节TGF-β1/Smad3信号通路干预糖尿病大鼠肝纤维化病变的部分机制。方法根据数字随机表法将34只SD大鼠分为空白组(10只)、模型组(8只)、抵当陷胸汤组(8只)和ALT-711组(8只),按55mg/kg剂量腹腔单次注射链脲佐菌素溶液制备糖尿病模型,予以相应的药物每天按相应剂量经胃灌药,于第8周末,麻醉后取材。采用Western blot法和实时荧光定量PCR法分别检测大鼠肝脏转化生长因子-β1(transforming growth factor-β1,TGF-β1)及其信号蛋白Smad3蛋白及其mRNA表达水平。结果与空白组比较,模型组大鼠肝组织TGF-β1、Smad3蛋白及mRNA表达水平均明显上调(P<0.05),p-Smad3蛋白表达水平明显上调(P<0.05);与模型组比较,抵当陷胸汤组和ALT-711组大鼠肝脏组织TGF-β1、Smad3蛋白及mRNA表达水平均明显下调(P<0.05),p-Smad3蛋白表达水平明显下调(P<0.05);抵当陷胸汤组和ALT-711组大鼠肝脏组织TGF-β1、Smad3、p-Smad3蛋白及TGF-β1、Smad3mRNA表达水平比较,差异均无统计学意义(P>0.05)。结论抵当陷胸汤通过抑制TGF-β1/Smad3信号通路激活干预糖尿病肝纤维化病变。

Effect of Didang Xianxiong Decoction on the TGFβ1/Smad3 Signaling Pathway in the Liver of Diabetic Rats

Abstract:Objective To investigate the effect of Didang Xianxiong Decoction on the TGFβ1/Smad3 signaling pathway in the liver of diabetic rats.Methods A total of 34 Sprague-Dawley rats were randomly divided into blank group with 10 rats,model group with 8 rats,Didang Xianxiong Decoction group with 8 rats,and ALT-711 group with 8 rats,using a random number table.Single intraperitoneal injection of streptozotocin at a dose of 55 mg/kg was given to establish a model of diabetes.Corresponding drugs were given by gavage every day,and samples were collected after anesthesia at the end of week 8.Western blot and quantitative real-time PCR were used to measure the protein and mRNA expression of transforming growth factor-β1(TGF-β1)and its signaling protein Smad3 in the liver.Results Compared with the blank control group,the model group had significantly upregulated protein and mRNA expression of TGF-β1 and Smad3(P<0.05)as well as protein expression of p-Smad3(P<0.05).Compared with the model group,the Didang Xianxiong Decoction group and the ALT-711 group had significantly downregulated protein and mRNA expression of TGF-β1 and Smad3(P<0.05)as well as protein expression of p-Smad3(P<0.05).There were no significant differences in the protein expression of TGF-β1,Smad3,and p-Smad3 and the mRNA expression of TGF-β1 and Smad3 between the Didang Xianxiong Decoction group and the ALT-711 group(P>0.05).Conclusion Didang Xianxiong Decoction exerts an interventional effect on liver fibrosis in diabetes by inhibiting the TGF-β1/Smad3 signaling pathway.
