维药迪娜口服液对急性肝损伤小鼠的保护作用

摘    要:目的 研究迪娜口服液对四氯化碳致小鼠急性肝损伤的保护作用。方法 取小鼠60只,随机分成正常组,模型组,联苯双酯组,迪娜口服液小、中、大剂量组。连续给药6 d后,采用0.15%四氯化碳腹腔注射(10 ml/kg)复制急性肝损伤模型。禁食不禁水18 h后,测定各组小鼠血清丙氨酸氨基转移酶(alanine aminotransferase,ALT)、天冬氨酸氨基转氨酶(aspartate transaminase,AST)和碱性磷酸酶(alkaline phosphatase,ALP)水平,观察各组小鼠肝脏组织形态变化。结果 与模型组比较,各给药组小鼠血清中ALT和AST水平均显著降低,其中迪娜口服液中、大剂量组ALT水平显著低于联苯双酯组和迪娜口服液小剂量组 (P<0.01)。各组小鼠血清ALP水平比较,差异均无统计学意义(P>0.05)。联苯双酯组和迪娜口服液小、中、大剂量组肝小叶结构破坏程度和水肿变性程度较模型组明显减轻,肝索排列较整齐。结论 迪娜口服液对四氯化碳致小鼠急性肝损伤具有明显的保护作用。

Protective Effect of Traditional Uighur Medicine Dina Oral Liquid in Mice with Acute Liver Injury

Abstract:Objective To investigate the protective effect of Dina oral liquid in mice with acute liver injury caused by carbon tetrachloride (CCl4). Methods Sixty mice were randomly divided into normal group, model group, bifendate group, and low-, middle-, and high-dose Dina oral liquid groups. After 6 d of treatment, all mice except those in the normal group were given 0.15% CCl4 (10 ml/kg) by intraperitoneal injection to induce a model of acute liver injury. After 18 h of fasting (without water deprivation), serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP) were determined, and the histomorphological changes in the liver were evaluated. Results Compared with the model group, all treated groups showed significant decreases in serum ALT and AST levels, and the middle- and high-dose Dina oral liquid groups had significantly lower serum ALT levels than the bifendate group and low-dose Dina oral liquid group ( P <0.01). There were no significant differences in serum ALP level between all groups (P>0.05). The bifendate group and low-, middle-, and high-dose Dina oral liquid groups showed less damage of lobular architecture and hydropic degeneration than the model group and had ordered arrangement of hepatic cords. Conclusion Dina oral liquid has a protective effect in mice with acute liver injury caused by CCl4.
