通督调神针灸法对局灶性脑缺血再灌注模型大鼠皮质区CD34、CD133及血清VEGFR-2表达的影响

摘    要:目的 探究通督调神针灸法对局灶性脑缺血再灌注模型(middle cerebral artery occlusion model/reperfusion model,MCAO/R)大鼠脑保护及血管新生的作用。方法 将150只SD大鼠随机分为假手术组、通督调神针灸组、常规针刺组、通心络药物组和模型对照组,每组30只。通督调神针灸组取百会穴艾灸,大椎穴刺络放血;常规针刺组选取风池、曲池、合谷、足三里、三阴交针刺治疗;通心络药物组予以通心络胶囊,每日每次1.0 g/kg灌胃。假手术组、模型对照组不予治疗;其余各组每日治疗1次,共28 d。治疗第7、14、21、28天处死大鼠立即取脑,脑组织石蜡包埋后切片,免疫组织化学法检测缺血皮质区CD34、CD133阳性表达,采用ELISA法检测VEGFR-2的表达。结果 MCAO/R大鼠缺血皮质区CD34、CD133及血清VEGFR-2表达量在第7天较高,随后逐渐降低;通督调神针灸法、常规针刺及通心络药物均可不同程度促进MCAO/R模型大鼠缺血皮质区CD34、CD133及血清VEGFR-2表达(P<0.05),且通督调神针灸法促进MCAO/R模型大鼠缺血皮质区CD34、CD133及血清VEGFR-2表达水平高于常规针刺及通心络药物(P<0.05)。结论 通督调神针灸法能有效促进MCAO/R大鼠缺血皮质区CD34、CD133及血清VEGFR-2表达。

Effect of Tongdu Tiaoshen Acupuncture on the Cortical Expression of CD34 and CD133 and Serum Expression of VEGFR-2 in A Rat Model of Middle Cerebral Artery Occlusion/Reperfusion

Abstract:Objective To investigate the effect of Tongdu Tiaoshen acupuncture on brain protection and angiogenesis in a rat model of middle cerebral artery occlusion/reperfusion (MCAO/R). Methods One hundred and fifty Sprague-Dawley rats were equally and randomly divided into sham operation group, Tongdu Tiaoshen acupuncture group, conventional acupuncture group, drug treatment group, and model control group. The Tongdu Tiaoshen acupuncture group received moxibustion treatment at Baihui (DU20) and bloodletting puncture at Dazhui (DU14). The conventional acupuncture group received punctures at Fengchi (GB20), Quchi (LI11), Hegu (LI4), Tsusanli (ST36), and Sanyinjiao (SP6). The drug treatment group received Tongxinluo Capsule by gavage at a daily dose of 1.0 g/kg. The sham operation group and the model control group received no treatment. The other groups received treatment once a day for a total of 28 days. The rats were sacrificed at 7, 14, 21, and 28 days and the brain tissues were collected. The expression of CD34 and CD133 in the ischemic cortex was detected on paraffin-embedded brain tissue sections by immunohistochemistry. The expression of VEGFR-2 was determined by enzyme-linked immunosorbent assay. Results In the MCAO/R rats, the expression of CD34 and CD133 in the ischemic cortex and serum expression of VEGFR-2 were the highest at 7 days and gradually decreased afterwards. Tongdu Tiaoshen acupuncture, conventional acupuncture, and Tongxinluo Capsule significantly elevated the expression of CD34 and CD133 in the ischemic cortex and serum expression of VEGFR-2 in the MCAO/R rats (all P<0.05). Moreover, Tongdu Tiaoshen acupuncture had significantly better efficacy than conventional acupuncture and Tongxinluo Capsule (all P<0.05). Conclusion Tongdu Tiaoshen acupuncture is an effective approach for promoting the expression of CD34 and CD133 in the ischemic cortex and serum expression of VEGFR-2 in the MCAO/R rats.
