电针胃俞募穴对单纯性肥胖大鼠下丘脑和血清神经肽Y的影响

摘    要:目的 观察电针胃俞募穴对单纯性肥胖大鼠脂代谢及神经肽Y(neuropeptide Y,NPY)水平的影响。方法 选用清洁级SD雄性大鼠80只,随机选10只为空白组,采取普通饲料喂养,其余大鼠采用高脂饲料饲养。6周后,筛选40只单纯性肥胖大鼠,将其随机分为模型组、中脘组(电针中脘)、胃俞组(电针胃俞)、胃俞募穴组(电针中脘配胃俞),持续电针3周。计算Lee指数,采用全自动生化分析仪检测血清三酰甘油(triglyceride,TG)、总胆固醇(total cholesterol,TC)和低密度脂蛋白胆固醇(low-density lipoprotein- cholesterol,LDL-C)水平,采用放射免疫法检测下丘脑和血清NPY含量。结果 模型组大鼠Lee指数,血清TG、TC、LDL-C水平,以及下丘脑和血清NPY含量均较空白组显著升高(P<0.01)。两因素析因设计的方差分析结果显示,针刺中脘和针刺胃俞对降低Lee指数,血清TG、TC、LDL-C水平,及下丘脑和血清NPY含量的主效应均具有统计学意义(P<0.01);针刺中脘和针刺胃俞对降低Lee指数、血清TG及下丘脑和血清NPY的交互作用均具有统计学意义(P<0.05,或P<0.01)。结论 单独电针中脘和胃俞均具有显著的减肥降脂作用,其机制与降低下丘脑和血清NPY有关,俞募配穴对减肥及降低血清TG和下丘脑、血清NPY具有显著的协同作用。

Effect of Electroacupuncture at Gastric Back-Shu and Front-Mu Points on Neuropeptide Y in Hypothalamus and Serum in Rats with Simple Obesity

Abstract:Objective To investigate the effect of electroacupuncture at gastric Back-Shu and Front-Mu points on the lipid metabolism and neuropeptide Y (NPY) level in rats with simple obesity.Methods Eighty clean male Sprague-Dawley rats were selected in the study. Ten rats were selected from the 80 rats as blank group to be fed a normal diet, and other rats were fed a high-fat diet. Six weeks later, 40 rats with simple obesity were screened out and randomly divided into model group, Zhongwan group (electroacupuncture at Zhongwan), Weishu group (electroacupuncture at Weishu), and gastric Back-Shu and Front-Mu points group (electroacupuncture at Zhongwan and Weishu points); electroacupuncture was performed for 3 weeks. Lee index was calculated; serum levels of triglyceride (TG), total cholesterol (TC), and low-density lipoprotein-cholesterol (LDL-C) were measured by an automatic biochemical analyzer; the content of NPY in the hypothalamus and serum was determined by radioimmunoassay. Results The model group had significantly increased Lee index, serum levels of TG, TC, and LDL-C, and content of NPY in the hypothalamus and serum (P<0.01). The results of two-way factorial design ANOVA showed that electroacupuncture at Zhongwan and electroacupuncture at Weishu showed significant main effects in reducing Lee index, serum levels of TG, TC, and LDL-C, and the content of NPY in the hypothalamus and serum (P<0.01); electroacupuncture at Zhongwan and electroacupuncture at Weishu had significant interaction in reducing Lee index, serum TG level, and the content of NPY in the hypothalamus and serum (P<0.05 or P <0.01). Conclusion Electroacupuncture at Zhongwan and electroacupuncture at Weishu have significant slimming and antihyperlipidemic effects, which is related to decreased NPY levels in the hypothalamus and serum. Acupuncture using a combination of Back-Shu and Front-Mu points shows a synergistic effect in reducing the body weight, serum TG level, and the content of NPY in the hypothalamus and serum.
