改良AS-PCR引物对ABO基因分型的影响

摘    要:目的通过改进AS-PCR引物,提高ABO基因型分型正确率。方法把引物P13′末端第5位碱基由G改为C,其分型图谱与改进前进行对比分析。结果改进AS-PCR引物后,OO型非特异产物减少,避免了将OO型误判为AO型。结论引物P13′末端第5位碱基由G改为C后,可有效防止OO型被误判为AO型。

Abstract:OBJECTIVE: To improve the correct rate of ABO genotyping by meliorating AS-PCR primer. METHODS: The primer P1 was changed into primer P1' by substituting the fifth base G for C of 3' end and the ABO genotyping results of primer P1 and P1' was compared and analysed. RESULTS: The non-specific product of OO typing is reducing and the wrong genotyping of OO and AO was avoided by meliorating AS-PCR primer. CONCLUSION: The rates of wrong ABO genotyping results could be effectively reduced by using altering primer P1'.
