| 摘 要： | 为获取金黄色葡萄球菌8型荚膜多糖(CP8),采用高压破碎菌体释放荚膜多糖,酶处理去除核酸和蛋白,再通过离子交换层析进一步纯化得到CP8荚膜多糖,并对荚膜多糖的纯度和反应原性进行了检测。结果显示,得到的荚膜多糖纯度为63.83%,净产量为6.23 mg/L。用免疫琼脂双扩散试验检测,结果纯化的CP8仅与CP8抗血清反应,反应原性在pH2.5~11.5范围内稳定。表明,获得的CP8荚膜多糖具有较高的纯度和良好的反应原性,可用于制备载体蛋白-荚膜多糖完全抗原,研发有效的预防金黄色葡萄球菌疫苗。
