豆类丝核菌次级代谢产物对小白鼠胎儿的致畸作用

摘    要:将妊娠的雌鼠随机分为5组,分别灌胃给予生理盐水、10 mg/kg环磷酰铵(CP)及高、中、低剂量豆类丝核菌次级代谢产物(苦马豆素含量分别为56.62、14.15、3.54 mg/kg),临产前剖腹取出胎鼠,将1/2胎鼠用茜素红染色做骨骼畸形观察,另1/2胎鼠用Bouins染色做内脏畸形检查,研究豆类丝核菌次级代谢产物对小鼠胎儿的致畸作用。结果显示,3个剂量组孕鼠增重、外观畸形率、骨骼畸形率、内脏畸形率与空白对照组均差异不显著(P>0.05),而与CP阳性对照组均差异极显著(P<0.01)。表明,在此剂量和条件下,豆类丝核菌次级代谢产物对小鼠无明显母体毒性,对胎鼠无发育毒性和致畸作用。

Abstract:To investigate the teratogenic effects of the secondary metabolites of Rhizotonia legumini-cola on mouse embryos,the gestationed mice were divided randomly into normal saline group,positive control group(10 mg/kg cyclophosphamide),high dosage group(containing SW 56.62 mg/kg),medium dosage group(containing SW 14.15 mg/kg) and low dosage group(containing SW 3.54 mg/kg).Before mouse accouchement,the fetus were taken out by the cesarotomy.Analyzing the developmental indexes of fetus,the appearance teratismus was observed.Half of the fetus were stained by alizarin red to observe skeletal deformity,and the other half of the fetus was dyed by Bouins staining solution to observe internal organ teratismus.No significant difference was observed between the three groups of the secondary metabolites of Rhizotonia leguminicola and the physiological saline group in body weight of gestationed mice,the rate of appearance teratismus,the rate of skeletal deformity and the rate of internal organ teratismus(P>0.05),but significant difference was observed between the 3 groups and the positive control group(P<0.01).The secondary metabolites of Rhizotonia leguminicola had no maternal toxicity in mice,and no developmental toxicity and teratogenic effects on the mice fetus under the experimental conditions.
