DNA and fingerprint identifications are now accepted as an integral part of the investigation of a wide range of criminal offences from burglary and auto crime to serious and major crime. Despite this, there is still much variation between U.K. police forces in the recovery of fingerprint and DNA material from crime scenes. Analysis of burglary and auto crime data for Northamptonshire, U.K., during a 3-year period has enabled an examination of the relationship between the deprivation of the neighborhood in which the crime was committed and the level of service provided by Crime Scene Examiners. The results indicate that the time spent examining a crime scene for forensic evidence is not affected by the deprivation of the neighborhood. Further, there is no statistical significance between deprivation and the recovery of fingerprints from the crime scene. The relationship between deprivation and DNA recovery is, however, statistically significant with DNA being recovered more frequently from less deprived neighborhoods. 相似文献

Abstract: Collection and interpretation of forensic intelligence (primarily through DNA and fingerprint identifications) is an integral part of the investigation of criminal offenses ranging from burglary and vehicle crime to major crime. The forensic contribution depends not only on the successful recovery of material, but also the ability to identify potential offenders and apply this intelligence to solve the crime. This study examines burglary and vehicle crimes investigated by Northamptonshire Police (U.K.) by analyzing relationships between deprivation of a crime location and the recovery and identification of DNA and fingerprint material. The results show that, for stolen vehicles, although significantly more forensic material (both DNA and fingerprints) is recovered and identified in more deprived neighborhoods, this does not lead to a corresponding increase in solved cases. These findings are considered in relation to previous studies, which have advocated the prioritization of resources at crime scenes most likely to yield forensic material. 相似文献

The primary aim of any DNA Database is to link individuals to unsolved offenses and unsolved offenses to each other via DNA profiling. This aim has been successfully realised during the operation of the New Zealand (NZ) DNA Databank over the past five years. The DNA Intelligence Project (DIP), a collaborative project involving NZ forensic and law enforcement agencies, interrogated the forensic case data held on the NZ DNA databank and collated it into a functional intelligence database. This database has been used to identify significant trends which direct Police and forensic personnel towards the most appropriate use of DNA technology. Intelligence is being provided in areas such as the level of usage of DNA techniques in criminal investigation, the relative success of crime scene samples and the geographical distribution of crimes. The DIP has broadened the dimensions of the information offered through the NZ DNA Databank and has furthered the understanding and investigative capability of both Police and forensic scientists. The outcomes of this research fit soundly with the current policies of 'intelligence led policing', which are being adopted by Police jurisdictions locally and overseas. 相似文献

Previous research has shown that as crime scene location deprivation increases (lower socioeconomic status), the recovery of forensic material, principally DNA and fingerprints, also increases. However, this increase does not result in more crimes being solved by forensic means. In this study, we analyze stolen vehicle data and find a statistically significant positive association between deprivation and the amount of forensic material that matched either the victim or an associate of the victim on a criminal database. The nature of this association was investigated further by inspecting recovered stolen vehicles to establish whether the condition of a stolen vehicle and the tidiness of its interior influenced the recovery of forensic material that was attributed to the victim or an associate. Contradictory results suggest that other factors may contribute to understanding the association between the recovery of victim- or associate-attributable forensic material and crime scene location deprivation. 相似文献

Preliminary tests for the detection of stains at crime scenes aim to focus the police work making them more efficient in the combat of criminality. The application of the luminol chemiluminescence reaction (3-aminoftalhidrazida) in presumptive tests for the detection of bloodstains is known for more than 40 years in forensic science. This reaction is based on the emission of light through the chemical reaction of luminol mixed with hydrogen peroxide and a hydroxide in the presence of a catalytic molecule (iron from the hemoglobin) (Laux [1]).This work evaluates the luminol interference and its effect on subsequent serological and DNA testing. Samples prepared with blood and different concentrations of luminol solution containing luminol, peroxide of hydrogen and sodium carbonate, were analyzed. Additionally, samples of serial dilutions of standard DNA mixed with luminol solution were also analyzed. Although presumptive tests with luminol do not establish the characterization and identification of stains at crime scenes, preliminary results indicated that it is suitable for the detection of invisible bloodstains for forensic analysis, with few detrimental effects on the serological tests and subsequent DNA recovery and typing. 相似文献

The transfer, persistence, prevalence, and recovery of DNA (DNA-TPPR) can be highly relevant in forensic investigations to evaluate the presence and/or actions of a person of interest (POI). Whilst the DNA-TPPR-related research has increased significantly over the last decade, there is a lack of data on companion animals and their relationship to human DNA transfer. Given the commonality of cats and dogs in households around the world, companion animals as receptors and vectors for DNA transfer can be highly relevant in cases involving animals as victims of a criminal offense, or cases requiring activity level evaluations. Samples were collected from an external area on the right side of 20 cats to determine the prevalence and sources of human DNA on this area. Preliminary data shows that human DNA is present on household cats, its source is mainly from household inhabitants. Further studies are required to elucidate the means and level of transfer of human DNA to and from cats and other household animals. This knowledge can be relevant to sample targeting in specific case circumstances and/or when considering possible means of the presence of a person’s DNA at the crime scene location. 相似文献

Papaver somniferum, commonly known as opium poppy, is the source of natural opiates, which are used as analgesics or as precursors in the creation of semi-synthetic opioids such as heroin. An increase in opioid addiction in the United States has resulted in high rates of illicit opioid use and overdoses. It has recently been shown that P. somniferum DNA suitable for genetic analysis can be recovered from heroin samples. The development of a comprehensive genetic individualization tool for opium poppy could serve to link cases and strengthen programs such as the Drug Enforcement Administration’s (DEA) Heroin Signature Program, which seeks to combat rising opioid use.The purpose of this study was to develop a quantitative real-time PCR (qPCR) method for the quantification of opium poppy DNA, compare three commercial DNA extraction kits for their ability to isolate DNA from poppy seeds, and evaluate nineteen opium poppy short tandem repeat (STR) markers for their use in a forensic identification panel. Such a panel could be used for individualizing samples and determining the geographic origin in heroin or poppy seed tea cases. The qPCR method was proven to be reproducible and reliable, specific for P. somniferum, and sensitive enough for forensic case-type samples. Of the three kits tested, the nexttec™ one-step DNA Isolation Kit for Plants was the optimal method and facilitated rapid extraction of DNA from poppy seeds. The majority of evaluated STR primer sets were unreliable or had low discriminatory power, limiting their use for individualization of poppy samples. A six-locus STR multiplex was developed and evaluated according to Scientific Working Group on DNA Analysis Methods (SWGDAM) and International Society of Forensic Genetics (ISFG) guidelines, including the use of a sequenced allelic ladder. The multiplex was found to have low discriminatory power, with greater than two-thirds of samples analyzed having just two different genotypes. The multiplex was determined to be unsuitable for individualization; however, a genotype map was developed as a proof of concept that these markers may be useful for determining the biogeographical origin of samples. Searching the poppy genome for new STR markers and developing new primer sets may be necessary for the creation of a powerful genetic tool for the individualization of P. somniferum. 相似文献

Numerous crimes (including murder), all having a common denominator, occurred in Germany and Austria between 1993 and 2009. All of these cases presented with identical female DNA traces being found at the crime scene. The crimes committed differed markedly, as did the suspects involved, which were of varied origin. Many of these cases could be solved. However, none of the suspects could identify an involved female. Fourteen of these cases (including one murder) occurred in Upper Austria.A special task force of the Austrian police, together with the Institute of Legal Medicine in Salzburg, began systematically searching for errors in the investigative process after the cases became more and more incoherent and nebulous. In the end, the DNA trace evidence was shown to be contaminated. A woman involved in the manufacture of the cotton swabs turned out to be the source of the female DNA profile.Following this, several products of other manufacturers were tested for contamination with DNA. It was noted that cotton swabs which had been sterilised with radiation were often contaminated. As a result, it is recommended that the manufacturing process, as well as the products themselves used in collection of DNA trace evidence, should be re-evaluated with the emphasis on preventing contamination. 相似文献

The analysis of trace DNA is a crucial component in forensic applications. Biological materials containing low-level DNA collected at crime scenes, such as fingerprints, can be valuable as evidence. Automatic detection of biological samples has been largely embraced in forensic applications to meet the increasing throughput requirements. However, the amount of DNA automatically retrieved from trace evidence often tends to be small and unstable, ultimately resulting in poor detection of DNA profiles. Thus, in this work, we introduced a robust DNA extraction and purification platform named Bionewtech® BN3200 (Bionewtech®, Shanghai, China) with the goal of constructing a rapid automatic detection system for trace DNA. The establishment of automatic detection system for trace DNA mainly encompassed two parts: assessing the sensitivity of automatic extraction platform and screening the optimal short tandem repeat (STR) typing kit. The sensitivity of Bionewtech® BN3200 platform based on Ultra-sensitive DNA Extraction kit was initially estimated, demonstrating that this extraction platform might contain large potential in the trace DNA extraction. For the amplification part, three sets of commercial multiplex STR typing kits were selected as candidates, and the amplified products were further genotyped on the Applied Biosystems 3500xl Genetic Analyzer. After comparation, SiFa™ 23 Plex Kit was determined as the most suitable amplification system for trace DNA. Eventually, the newly exploited trace DNA detection system was successfully implemented in the detection of fingerprints derived from glass surfaces with the five-seconds contact time. As a result, the DNA recovered from the fingerprints fluctuated approximately from 57.60 pg to 18.05 ng, in addition, over 70% of the total STR loci were detected in 75% of the fingerprint samples. 相似文献

Incorporating a simulated crime scene into one’s pedagogy in forensic science undergraduate courses allows students the opportunity to experience a realistic scenario while demonstrating their knowledge and enhancing their critical thinking skills. The purpose of this paper is to examine an active learning approach to using simulated crime scenes to provide an impactful learning experience for students. While potentially challenging for the instructor, constructing a crime scene scenario can provide students with hands-on practical experience while helping to dispel forensic science misconceptions. Through many years of creating crime scenes, best practices for using such high impact activities in forensic science courses are described in detail, including preparatory exercises that culminate in the final crime scene scenario, considerations in preparing and constructing a crime scene activity, and supervising and assessing students once the activity begins. Three major challenges to implementing active learning exercises such as simulated crime scenes in undergraduate forensic science programs include high workload for instructors, limited supplies, and lack of suitable facilities. Workload solutions include instructors and departments considering the cost and benefit of course releases to improve curriculum and student enrollment. Supplies and facilities solutions involve innovation in reuse and repurposing of supplies, and instructor flexibility in using classroom and outdoor spaces. 相似文献

选择合适的 STR基因座 ,建立中国“罪犯 DNA数据库”模式库。以 2 2 11名汉族、15 0名维吾尔族、10 4名回族群体为分析对象 ,提取罪犯血样 DNA,用复合 PCR和四色荧光技术检测了 D3S135 8、v WA、FGA、D8S1179、D2 1S11、D18S5 1、D5 S818、D13S317、D16 S5 39、TH0 1、TPOX、CSF1PO、D7S82 0等 13个 STR基因座及性别 Amelogenin基因座。在 13个 STR基因座中 ,除 TH0 1、TPOX基因座外 ,其余各基因座的个体鉴别能力 (DP)值均接近 0 .9,杂合度(H)均大于 0 .7,排除概率 (PE)大都在 0 .5以上 ,其中 FGA、D8S1179、D2 1S11和 D18S5 1基因座 DP≥ 0 .95 ,H≥ 0 .85 ,PE≥ 0 .6 5 ,表明它们在法医学上极有应用价值。TH0 1和 TPOX在多态性方面较差 (H分别为 0 .6 430和 0 .6 2 96 ,PE分别为0 .40 46和 0 .370 1) ,但也符合应用于法医学的要求。13个基因座的平均偶合率为 5× 10 - 1 5 ~ 1.2× l0 - 1 4 ,适合作为中国人群的遗传学标志 ,用于建立中国“罪犯 DNA数据库” 相似文献

The transfer of DNA from hands to objects by holding or touching has been examined in the past. The main purpose of this study was to examine the variation in the amount of DNA transferred from hands to glass, fabric and wood. The study involved 300 volunteers (100 for glass, 100 for fabric and 100 for wood) 50% of which were male and 50% female. The volunteers held the material for 60 s. The DNA was recovered from the objects using a minitape lift, quantified using the Quantifiler kit assay, extracted using a ‘Qiagen® QIAamp DNA mini kit’ and amplified using the AmpFlSTR® SGM Plus™ Amplification Kit at 28 cycles. The results show that using ANOVA there was a significant difference (F = 8.2, p < 0.05) between the three object types in the amount of DNA recovered. In terms of DNA transfer and recovery, wood gave the best yield, followed by fabric and then glass. The likelihood of success of obtaining a profile indicative of the holder was approximately 9% for glass samples, 23% for fabric and 36% for wood. There was no significant difference between the amount of DNA transferred by male or female volunteers. In this study good shedder status, as defined by obtaining useful profiles of 6 or more alleles, is estimated at approximately 22% of the population. The phenomenon of secondary transfer was observed when mixed DNA profiles were obtained but the incidence was low at approximately 10% of the total number of samples. DNA profiles corresponding to more than one person were found on objects which had been touched by only one volunteer. Although secondary transfer is possible the profiles obtained from touched objects are more likely to be as a result of primary transfer rather than a secondary source. 相似文献

Abstract: DNA material is now collected routinely from crime scenes for a wide range of offenses and its timely processing is acknowledged as a key element to its success in solving crime. An analysis of the processing of approximately 1500 samples of DNA material recovered from the property crime offenses of residential burglary, commercial burglary, and theft of motor vehicle in Northamptonshire, U.K. during 2006 identified saliva and cigarette ends as the main sources of DNA recovered (approximately 63% of samples) with blood, cellular DNA, and chewing gum accounting for the remainder. The conversion of these DNA samples into DNA profiles and then into matches with offender profiles held on the U.K. National DNA database is considered in terms of the ease with which Crime Scene Examiners can recover DNA rich samples of different sources, the location of the DNA at the crime scene, and its mobility. A logistical regression of the DNA material recovered has revealed a number of predictors, other than timeliness, that greatly influence its conversion into a DNA profile. The most significant predictor was found to be Crime Scene Examiner accreditation with offense type and DNA sample condition also being relevant. A similar logistical regression of DNA samples profiled that produced a match with an offender on the U.K. National DNA database showed no significance with any of the predictors considered. 相似文献

The development of a nucleic acid extraction method based on magnetic separation has opened up possibilities forl automation of DNA extraction. The BioRobot M48 is one of robotic stations applicable to automated DNA extraction in forensics. However, each new method should be thoroughly validated before application to routine casework. Our aim was to compare the effectiveness of the currently utilized organic/Microcon 100 based extraction procedure and magnetic extraction with BioRobot M48. The DNA concentration of DNA extracts obtained from different kinds of typical forensic material was evaluated followed by amplification with the SGM Plus or Identifiler kit and capillary electrophoresis using ABI 3100 Avant. We can conclude that BioRobot M48 is a very effective instrument for DNA extraction from most specimens and can be successfully applied in forensic laboratories. 相似文献

Due to recent improvements in forensic DNA testing kit sensitivity,there has been an increased demand in the criminal justice community to revisit past convictions or cold cases.Some of these cases have little biological evidence other than touch DNA in the form of archived latent fingerprint lift cards.In this study,a previously developed optimised workflow for this sample type was tested on aged fingerprints to determine if improved short tandem repeat(STR)profiles could be obtained.Two-year-old samples processed with the optimised workflow produced an average of approximately five more STR alleles per profile over the traditional method.The optimised workflow also produced detectable alleles in samples aged out to 28 years.Of the methods tested,the optimised workflow resulted in the most informative profiles from evidence samples more representative of the forensic need.This workflow is recommended for use with archived latent fingerprint samples,regardless of the archival time. 相似文献

The issue of DNA transfer is becoming increasingly important in crime scene situations, as DNA analytical techniques now detect tiny amounts. Whereas primary and secondary DNA transfers have been well studied, subsequent transfer steps have received much less focus. This study aimed to measure the detectability of a DNA source after multiple transfer events. Transfer of wet blood gave a full genetic profile well beyond the secondary transfer events on both cotton and glass substrates. Dry blood gave a full profile well beyond the secondary transfer events on glass only, but to a lesser extent than wet blood. Touch DNA only produced a full profile on the primary substrate on both cotton and glass, and detectable quantities beyond the secondary transfer event on glass only. Our results will contribute to a better understanding of the tertiary and subsequent transfer of DNA, which will allow for improved evaluation of the likelihood of alternative scenarios explaining why an individual's DNA was found at a crime scene. 相似文献
