To establish a database for DXS8378, HPRTB, DXS7423, DXS7132, DXS10134, DXS10074, DXS10101, DXS10135 loci in an Italian population sample from Brescia (Northern Italy) and from the Sardinia island, 131 unrelated individuals were typed. DNA was amplified in a multiplex reaction with subsequent automatic detection using capillary electrophoresis. The obtained data give a contribution to the definition of Italian population STR allele frequencies for the eight analysed loci.  相似文献

The investigation of the X-linked DNA markers are well established in the forensic routine case work. We studied an Ewe population sample from Ghana. The eight X-chromosomal STRs DXS10135, DXS8378, DXS7132, DXS10074, HPRTB, DXS10101, DXS10134 and DXS7423 were analyzed in 182 Ewe individuals (108 females and 74 males) from the region of Sogakofe (Ghana). Allele frequencies and statistical parameter as well as comparison with known data from Germans and with data from an Amharic population (Ethiopia) are presented.  相似文献

目的调查居住在广州的非洲籍人群的15个STR基因座多态性。方法应用Sinofiler TM荧光标记检测系统对122个无关个体样本进行分型检测,使用Powerstats V12、Genopop 3.4、Mega 3.4等软件进行统计计算与群体间遗传距离比较。结果居住在广州的非洲籍人群15个STR基因座的累积个体识别率为1-5.80147×10-18、累积非父排除率为1-8.6019×10-8,居住在广州的非洲籍人群与广州汉族人群遗传距离最远,与巴哈马群岛非裔混合群体间的遗传距离最近。结论该系统可满足广州地区非洲籍人群个体识别与亲权鉴定的需要,同时与广州汉族人群存在的较大统计学差异,在进行人群区分时具有重要意义。  相似文献

Allele frequencies, together with some parameters of forensic interest were estimated for nine STRs included in the AmpF/STR Profiler kit (CSF1PO, D3S1358, D5S818, D7S820, D13S317, FGA, TH01, TPOX and vWA) in a sample of 215 unrelated individuals from Cartagena (Colombia). For all loci, no significant deviations from Hardy-Weinberg equilibrium were observed. Comparative analysis results between our data and those from other Colombian and African population samples revealed significant differences, except with two Colombian Caribbean Coast sub-regions.  相似文献

Eight X-chromosomal short tandem repeat (X-STR) markers were analyzed in 258 unrelated Japanese (144 males and 114 females) using Mentype® Argus X-8 PCR Amplification Kit (Biotype AG) which contains DXS7132, DXS7423, DXS8378, DXS10074, DXS10101, DXS10134, DXS10135 and HPRTB. The DXS10135 locus proved to be highly polymorphic marker (PIC: 0.945) and the DXS7423 showed the lowest value (PIC: 0.453). The exact test for genotype distribution showed no significant deviation from the Hardy-Weinberg equilibrium.  相似文献

With the aim to establish a Y-STR haplotype database, a total of 554 males from seven Western Mediterranean populations were genotyped for the 12 Y-chromosome STR loci (minimal haplotype extended by loci DYS437, DYS438 and DYS439) included in the Powerplex Y System (Promega).Among the 554 males analyzed, 443 different haplotypes were obtained, of which 372 were unique. The other haplotypes were shared by two to seven men. The two most frequent haplotypes were both found in seven Ibiza men. A remarkable haplotype diversity of 0.9988 ± 0.0002 was observed.The haplotypes were searched against the Y-haplotype reference database (YHRD) and 149 haplotypes matched to at least one YHRD sample.  相似文献

目的调查459例北京汉族无关个体21个常染色体非CODIS的STR基因座遗传多态性并评价其应用价值。方法用AGCU21+1荧光标记复合扩增系统对459例无关个体的21个STR基因座(D6S474、D12SATA63、D22S1045、D10S1248、D1S1677、D11S4463、D1S1627、D3S4529、D2S441、D6S1017、D4S2408、D19S433、D17S1301、D1GATA113、D18S853、D20S482、D14S1434、D9S1122、D2S1776、D10S1435、D5S2500)进行检验。得到STR分型后,用相关软件进行统计分析并计算法医学应用参数。结果获得21个STR基因座的频率分布;相关参数为:H值从0.5894-0.8038,PD值从0.7898-0.9265,PE3值从0.3618-0.6029,PE2值从0.2031-0.4256,PIC值从0.5638到0.7640。结论联合应用Identifiler系统和AGCU21+1系统,有利于亲子关系的认定及对可疑突变的判断。  相似文献

Seventeen autosomal STR loci (D8S1179, D21S11, D7S820, CSF1PO, D3S1358, TH01, D13S317, D16S539, D2S1338, D19S433, VWA, TPOX, D18S51, D5S818, FGA, Penta E and Penta D) and 16 Y-STR haplotype loci (DYS19, DYS385, DYS389I, DYS398II, DYS390, DYS391, DYS392, DYS393, DYS437, DYS438, DYS439, DYS448, DYS456, DYS458, DYS635 and GATA H4.1) were analyzed in the sample of 200 unrelated Croatians. The agreement with HWE was confirmed for all autosomal STR loci. The combined power of discrimination (PD) and the combined power of exclusion (PE) for the 17 autosomal STR loci were 0.999999999999999999682299331476 and 0.99999995, respectively. Penta E proved to be the most informative autosomal STR locus. Among 200 Croatian males, 197 Y-STR haplotypes were identified and haplotype diversity was estimated at 0.9998 ± 0.0005.  相似文献

Genetic population data for five X-STR (DXS6854, DXS7424, DXS101, DXS6808 and DXS7132) were obtained from Bauru population (São Paulo, Brazil). No deviations from the Hardy-Weinberg equilibrium were observed, with the exception of DXS101. The combined powers of discrimination in males and females were 0.99897253 and 0.99999120, respectively. These high values show the potential of this system in human identification and paternity testing.  相似文献

The X-linked short tandem repeats (STR) markers have proven to be very useful tools for paternity testing when the disputed child is female. The aim of this study was to describe the polymorphism of three X-chromosomal STR loci (DXS6797, DXS6800 and HPRTB) in an Antioquian (Colombian) population sample. PCR products were separated in 4% acrylamide-bis-acrylamide denaturing gels followed by silver staining. Allele size determination and genotyping were performed according to recommendations of the DNA Commission of the International Society of Forensic Genetic using the allelic ladder manufactured at home and based on DNA controls including K562 and 9947A (Promega). Gene frequencies were calculated using ARLEQUIN version 3.11. Population genetic data were obtained by analyzing 127-400 unrelated males and 135 unrelated females from Antioquian (Colombian) population. Distribution of the allele frequencies of these systems for Antioquia population is similar to the European populations.  相似文献

In this study, Japanese and Han Chinese individuals (n = 32, each) were genotyped for 261 autosomal STRs, and allele frequencies were calculated for each locus in each population. The average number of alleles for all loci in Japanese and Han Chinese populations was 6.65 and 6.56, respectively. The tests for deviations from HWE performed using an exact test showed that the number of STRs (P > 0.05) in Japanese and Han Chinese populations was 236 and 241, respectively. Calculation of forensic parameters showed heterozygosity, and the exclusion means in the Japanese population were 0.7185 and 4813 and those in the Han Chinese population were 0.7308 and 0.5008. In addition, population genetic analyses, such as principal component analysis and factorial correspondence analysis, were performed and a differential formula with likelihood ratios was applied for various number of STR loci based on the effectiveness of differentiation between the two populations. Accordingly, this study suggests that statistical differentiation between genetically close populations, such as the Japanese and Han Chinese populations, is possible if approximately 40–50 effective STR loci are analyzed.  相似文献

The most efficient markers to solve filiation cases are the STRs, including complex cases that require the analysis of a greater number of markers. In this study samples from 123 unrelated individuals from the department of Santander (northeast Colombia) were typed for 23 autosomal STRs included in VeriFiler express kit (Thermo Fisher Scientific),and their allele frequencies and parameters of forensic relevance were determined. Results demonstrate independence within and between the loci analyzed, and the accumulate power of exclusion for the full set of markers was high (99.9996%), as well as the match probability, which was 1 in 8.77E + 29. Therefore, this northeast Colombian population database can be used in forensic to estimate the frequency of the genetic profile using of a multiple locus including in this DNA kit.  相似文献

Eight X-chromosome STR were investigated in two groups of individuals from two Italian regions, Umbria and Sardinia. These two regions have a very different history. Umbria, situated in the centre of Italy and without defensive natural barriers, in the course of the centuries has been object of many invasions and appropriations by foreign people. On the contrary, Sardinia, an island in the centre of the Mediterranean sea, has endured a minor number of foreign invasions. The ChrX markers were amplified in a pentaplex (DXS6789, DXS7423, DXS6807, DXS101, DXS8377) and a triplex (DXS10074, DXS10075, DXS10079) that represents a haplotype. The DNA was extracted from 200 blood samples using the QIAmp DNA Minikit (Qiagen™). Amplification products were detected on an ABI PRISM 310 Genetic Analyzer (Applied Biosystems), using the same dye labels, run conditions, standard (GeneScan 500 Liz) and matrix file of AmpFlSTRIdentifiler. Statistical analyses for all the loci and intergroup comparisons between the two regional groups of individuals were performed.  相似文献
