This study evaluated the compatibility of the most common enhancement methods and lifting techniques with DNA profiling. Emphasis is placed on modern lifting techniques (i.e., gelatin lifters and Isomark?) and historical fingerprint lifts for which limited research has been previously conducted. A total of 180 fingerprints were deposited on a glass surface, enhanced, lifted, and processed for DNA typing. DNA could be extracted and profiled for all the powders and lifts tested and from both groomed fingerprints and natural prints with no significant difference in the percentage of profile recovered. DNA profiles could also be obtained from historical fingerprint lifts (79.2% of 72 lifts) with one or more alleles detected. These results demonstrate the compatibility between different powder/lift combinations and DNA profiling therefore augmenting the evidential value of fingerprints in forensic casework.  相似文献

Abstract: A study was conducted to investigate whether natural magnetite (Fe3O4), which is an abundant mineral in Thailand, could be used as a magnetic powder in the detection of latent fingerprints. Because of the presence of impurities, powdered magnetite is only weakly attracted by a magnet and cannot be used as a magnetic fingerprint powder by itself. Mixing a small amount of magnetite powder with nickel powder greatly enhances the magnetic attraction. A mixture of magnetite powder and nickel powder in a mass ratio of approximately 1:100 was found to be suitable for use as a magnetic fingerprint powder. Fingerprints developed using the magnetite/nickel mixture on nonporous surfaces were found to exhibit good adherence and clarity. Using an automated fingerprint identification system, the number of minutiae detected in fingerprints developed by using the prepared powder on nonporous surfaces was found to be comparable to those detected in fingerprints developed by using a commercial black magnetic powder. The cost is lowered by more than 60%.  相似文献

Two seizures of counterfeit 100 US dollar bills related to the same indicative number were submitted for processing of latent fingerprints. On one group of notes, identifiable fingerprints could be detected by the routine application of amino acid reagents. In the second case, this technique gave no results, even on deliberately deposited prints. Fingerprints could be revealed, however, by cyanoacrylate fuming followed by magnetic powder. Comprehensive paper analysis showed that banknotes from both seizures differed remarkably by chemical composition as well as paper macroscopic properties. The difference in surface free energy (related to surface tension) of the banknotes in the two groups seemed to be the major factor responsible for the great variance in fingerprint detectability.  相似文献

This study evaluated the effectiveness of using liquid latex as a pre-treatment for fingerprint recovery from the exterior surfaces of vehicles in summer. The sample of this study was 540 sebaceous latent fingerprints deposited on the lower body of three vehicles. Thirty control and thirty experimental fingerprints were deposited on each vehicle, and the experiment was repeated three times. The three vehicles were driven daily for either 2, 3, or 4 weeks after the deposition of fingerprints. After the vehicles reached their designated debris accumulation duration, the latent fingerprints in the control groups were developed with black fingerprint powder. Liquid latex was applied onto the fingerprints in the experimental groups, and they were subsequently developed with black fingerprint powder. A chi-sure test indicated that there was a significant difference in fingerprints recovery performance between two methods (X2 = 4.903, d.f. = 1, p = 0.027). An odds ratio test indicated the control method increases the probability of fingerprint recovery by 1.54 times. A Fisher's exact test was used to evaluate the quality of fingerprints recovered from both methods and it indicated that there is no significant difference in quality using the two methods (p = 0.058). This study indicated that the traditional fingerprint powder method performed better for fingerprint recovery from exterior surfaces of vehicles in summer.  相似文献

To understand how time and temperature could affect latent fingerprints, wooden pallets were used to construct five units that each housed ten 60-watt incandescent light bulbs. Fingerprints were deposited on the top, middle, and base of the globes. The bulbs were powered on (except the control bulb in each unit) for 18, 48, 72, 120, 168, 336, 504, and 672 h. Fingerprints recovered from the bulbs by tape lift after black powder processing were given a quality score. A thermal imaging camera determined temperatures on three areas of the bulbs. Fingerprints on the top of the globe (156.3°C) had the lowest quality score, and fingerprints on the middle of the globe (112.6°C) had the highest quality score. The mean quality scores of the three temperature classes were within one standard deviation of one another, meaning there is no way to estimate fingerprint age based on its quality after heat exposure.  相似文献

The application of powders to fingerprints has long been established as an effective and reliable method for developing latent fingerprints. The powders adhere to the ridge pattern of the fingerprint only, thus allowing the image to be visualised. Fingerprints developed in situ at a crime scene routinely undergo lifting with specialist tapes to facilitate subsequent laboratory analysis. As with all recovered evidence these samples would be stored in evidence bags to allow secure transit from the scene to the laboratory and also to preserve the chain of evidence. In this paper, the application of Raman spectroscopy for the analysis of exogenous material in latent fingerprints is reported for contaminated fingerprints that had been treated with powders and also subsequently lifted with adhesive tapes. A selection of over the counter (OTC) analgesics were used as samples for the analysis and contaminated fingerprints were deposited on clean glass slides. The application of aluminium or iron based powders to contaminated fingerprints did not interfere with the Raman spectra obtained for the contaminants. In most cases background fluorescence attributed to the sebaceous content of the latent fingerprint was reduced by the application of the powder thus reducing spectral interference. Contaminated fingerprints developed with powders and then lifted with lifting tapes were also examined. The combination of these two techniques did not interfere with the successful analysis of exogenous contaminants by Raman spectroscopy. The lifting process was repeated using hinge lifters. As the hinge lifters exhibited strong Raman bands the spectroscopic analysis was more complex and an increase in the number of exposures to the detector allowed for improved clarification. Raman spectra of developed and lifted fingerprints recorded through evidence bags were obtained and it was found that the detection process was not compromised in any way. Although the application of powders did not interfere with the detection process the time taken to locate the contaminant was increased due to the physical presence of more material within the fingerprint. The presence of interfering Raman bands from lifting tapes is another potential complication. This, however, could be removed by spectral subtraction or by the choice of lifting tapes that have only weak Raman bands.  相似文献

In forensic science, fingerprints are a common source of evidentiary information. However, latent examination is not always successful and trace human DNA cannot always be obtained. Thus, examining the fingerprint microbiome may offer a suitable alternative to more traditional methods of forensic identification. The Zymo Research ZR Bacterial/Fungal DNA MicroPrep™ Kit, Qiagen QIAmp® DNA Mini Kit, Promega Wizard® Genomic DNA Purification Kit, and the MPBio FastDNA® Spin Kit were compared for their ability to yield a sufficient amount of bacterial DNA for next-generation sequencing in order to obtain a microbiome profile. Prints were deposited onto slides, allowed to sit for up to 1 month, and total DNA isolated and quantified using each kit. The kit from Zymo Research yielded the most concentrated DNA sample (0.0084 ng/µL) in the least amount of time as compared to other kits examined. Although this amount of DNA was far below the recommended DNA concentration threshold recommended for next-generation sequencing, a microbiome profile was successfully obtained. As interest in using the microbiome of an individual as a forensic tool continues to increase, there is the possibility that the microbiome of a fingerprint could complement traditional human DNA profiling in the future. This study provides evidence that trace amounts of bacterial DNA from fingerprints is quantifiable and sufficient for microbiome analysis.  相似文献

Latent fingerprint deposits on thermal paper have been visualized noninvasively at visible wavelengths when illuminated with a UV‐A light source (peak 365 nm). A higher intensity UV source (250 W/m2 at 0.38 m) gave superior fingerprint visibility when compared with a 60 W/m2 (at 0.4 m) source. Removing the visible (blue) component of the light source emission did not adversely affect the visibility of the fingerprint. Sample fingerprints from 100 donors, when examined 24 h after deposition, produced identifiable fingerprints from nearly 34% of fingerprint deposits. A mechanism for the observed visibility is proposed based on low emission of visible wavelengths from areas of thermal paper coincident with the fingerprint deposit, when illuminated with UV. This is likely due to a weak color change in the thermal paper dye arising from protonated amino acid components of the sweat. This effect was not observed on nonthermal paper.  相似文献

Fingerprints deposited at crime scene can be a source of DNA. Previous reports on the effects of fingerprint enhancement methods have focused mainly on fingermarks deposited in blood or saliva. Here, we evaluate the effects of fingerprint enhancement methods on fingerprints deposited on porous surfaces. We performed real‐time quantification and STR typing, the results of which indicated that two methods (iodine fuming and 1,2‐indanedione in ethyl acetate enhancement) had no effect on the quantity of DNA isolated and resultant STR alleles when compared to control samples. DNA quantities and allele numbers were lower for samples enhanced with silver nitrate and 1,2‐indanedione in acetic acid when compared to control samples. Based on DNA quantity, quality, and observable stochastic effects, our data indicated that iodine fuming and 1,2‐indanedione in ethyl acetate were the preferred options for the enhancement of fingerprints on porous surfaces.  相似文献

One of the crucial factors determining the cyanoacrylate deposit quality over latent fingerprints appeared to be the extent of the humidity. This work focuses on the enhancement/refreshment of age‐degraded latent fingerprints by irradiating the samples with UV, X‐ray, or thermal neutrons prior to the cyanoacrylate (CA) fuming. Age degradation of latent fingerprints deposited on glass surfaces was examined through the decrease in the number of characteristic minutiae counts over time. A term “critical day” was introduced for the time at which the average number of identifiable minutiae definitions drops to one‐half. Fingerprints older than their “critical day” were exposed to either UV, X‐ray, or thermal neutrons. Identical reference samples were kept unexposed. All samples, both reference and irradiated, were developed during a single CA fuming procedure. Comparative latent fingerprint analysis showed that exposure to ionizing radiation enhances the CA fuming, yielding a 20‐30% increase in average minutiae count.  相似文献

Based on the phenomenon that static electricity attracts dust, a polyethylene terephthalate (PET) semirigid sheet coated with printing ink was used to visualize impressions of fingerprints on the skin of living and dead bodies. Compared with visualization methods for perspiration fingerprints, this method recovers better images for a longer time after the fingerprint has been deposited on skin. Fingerprints transferred to the PET sheet are photographed with sidelighting using an ordinary light source. For fingerprints that yield inadequate contrast, an argon-ion laser can be used to improve the contrast.  相似文献

An examination was done to investigate whether certain lifting techniques can lift recovered latent fingerprints on human skin surfaces of living subjects. For recovery Swedish Soot powder mixture (Swedish Black) was used.Donors intentionally placed fingerprints on the skin surface of living subjects. Finger marks were then in all cases recovered with Swedish Black powder. The procedure was repeated after 1 h and 4 h. Treated finger marks were secured and preserved as latent fingerprint evidence by different lifting processes. Having examined skin surfaces and finger marks we observed that the lifters such as white instant lifter, white fingerprint gelatin, black fingerprint gelatin, silicone, transparent adhesive tape, are suitable. Moreover, white fingerprint gelatin and white instant lifter proved to be very good at lifting treated finger marks. Black fingerprint gelatin was very good also, but finger marks were examined by slant light.  相似文献

Corrosion of α‐phase brass by sebaceous sweat fingerprint deposits produced identifiable impressions in a majority of samples (n = 40) 4 days after deposition. Combining sebaceous with eccrine sweat yielded a greater percentage of identifiable fingerprint deposits, although this increase was not statistically significant. Production of identifiable fingerprints from eccrine sweat deposits was dependent on the sampling time of year with deposits taken during summer months giving similar percentages of identifiable fingerprints to sebaceous deposits. A statistically significant positive correlation was found between elapsed days after deposition and identifiable eccrine (ρ = 0.787, p < 0.05), sebaceous (ρ = 0.724, p < 0.05), and eccrine/sebaceous mixture (ρ = 0.908, p < 0.01) fingerprints deposited during summer months. The summer increase in the percentage of identifiable eccrine sweat deposits was statistically significant compared to winter eccrine deposits (p < 0.0001). Observations were consistent with results obtained from artificial sebaceous and eccrine sweat.  相似文献

In unison, fingerprinting and DNA analysis have played a pivotal role in forensic investigations. Fingerprint powders that are available on the market can come in a range of colors and with specific properties. This study evaluated the efficiency of DNA extraction from samples coated with 3 brands of fingerprint powders: Lightning, Sirchie, and SupraNano, covering a range of colors and properties. A total of 23 fingerprint powders were tested using the Chelex, Promega DNA IQ™, and Applied Biosystems™ PrepFiler™ DNA extraction protocols. The DNA IQ™ and PrepFiler™ methods extracted higher yields of DNA in comparison to Chelex, which also accounted for better quality of PowerPlex x00AE; 21 DNA profiles recovered. There were no signs of degradation or inhibition in the quantification data, indicating that samples returning low DNA yield was due to interference during DNA extraction and not PCR inhibition. DNA profiles were recovered from the majority of fingerprint powders with only a single powder, Sirchie Magnetic Silver, failing to produce a profile using any of the methods tested. A link was observed between the DNA extraction chemistry, fingerprint powder property, that is, nonmagnetic, magnetic and aqueous, and the brand of fingerprint powder. Overall, the DNA IQ™ method was favorable for nonmagnetic fingerprint powders, while magnetic fingerprint powders produced more DNA profiles when extracted with the PrepFiler™ chemistry. This study highlights the importance of screening DNA extraction chemistries for the type of fingerprint powder used, as there is not a single DNA extraction method that suits all fingerprint powder brands and properties.  相似文献

A method for lifting bloody footwear impressions using alginate casts and enhancing the lifted impressions with amido black is presented. On rough or dark substrates, background interferences may conceal significant details of footwear impressions. Illumination with alternative light sources and chemically enhancing the bloody footwear impressions may reveal additional details, but sometimes, lifting footwear impressions prior to enhancing is the only way to expose hidden details (by using blood reagents not adequate on the original). Several cast formulations were tested for lifting the footwear impressions. The best results were achieved using Aroma fine®. Enhancement of the footwear impressions was attempted with several reagents prior to lifting, during the casting process, and on the lifted footwear impressions. Applying amido black to footwear impressions lifted with alginate produced the sharpest and most detailed footwear impressions. Alginate castings followed by chemical enhancement with amido black may produce high‐quality footwear impressions for comparison.  相似文献

Former studies [Nature (1997) 387, Electrophoresis 20 (1999) 2870, P. Van Renterghem, D. Leonard, C. De Greef, Progress in Forensic Genetics, Vol. 8, 2000, p. 501, J. Forensic Sci. 45 (3) (2000) 687] have shown that even a single skin contact, documented by a latent fingerprint, can transfer enough DNA for a genetic analysis. It was proven in these studies that it is possible to swab fingerprints from surfaces [Nature (1997) 387, Electrophoresis 20 (1999) 2870, P. Van Renterghem, D. Leonard, C. De Greef, Progress in Forensic Genetics, Vol. 8, 2000, p. 501] and use them as a DNA source. Usually, however, discovered fingerprints are removed with scotch tape and placed on evidence cards for further investigation. In this study, we tried to assess the potential use of latent fingerprints as a DNA source for STR typing. The materials (magnetic powder, soot powder and scotch tape) used for visualization and archiving fingerprints in Germany were tested for their PCR inhibitory characteristics. Then, fingerprints were placed on clean glass surfaces, visualized and tested for their usefulness as a DNA source.Obtained DNA was quantified and tested in an STR system. Partly it proved possible to type fingerprints taken directly from the surface as well as fingerprints removed from the surface with scotch tape.  相似文献

The purpose of this research is to determine if latent fingerprints deposited on the exterior glass surfaces of vehicles, then covered in debris, can be recovered. Past research used liquid latex to lift soot to recover trace evidence. Recently, liquid latex has been used to recover latent fingerprints along the bottom of vehicles. In this study, a total of 216 latent fingerprints were deposited on the exterior windows of three vehicles. Three control and three experimental latent fingerprints were placed on each side window. The vehicles collected debris for either 2, 3, or 4 weeks. After debris collection, liquid latex was applied to the experimental sections. The underlying fingerprints were developed with white granular powder. Control fingerprints were developed directly with white granular powder. A chi-square test revealed a significant difference in fingerprint recovery between the control and liquid latex method (X2 = 9.026, d.f. = 1, p = 0.003). An odds ratio determined that the control method increases the probability of latent fingerprint recovery by 2.68. Fisher's exact test indicated that there is no statistically significant difference between the detail of the recovered control and experimental fingerprints (p = 0.065). This study demonstrates that recovery of fingerprints is possible using the liquid latex method; however, the control method recovers more fingerprints on the glass exterior of vehicles. If latent fingerprints are thought to be present on the exterior glass surfaces of vehicles, the control method should be used to improve vehicle processing by investigators.
