Since 1928, hemagglutinins have been known to exist in saliva; however, they have not been utilized as evidence in criminal investigations because in the past, techniques for measuring them have not been sufficiently sensitive. In this paper we describe improved techniques for detecting salivary hemagglutinins and report initial results obtained with these methods. The stability of salivary hemagglutinins at several different temperatures was examined in liquid samples and in dried stains on filter paper, cigarette butts, and envelope flaps. Our observations indicate that salivary hemagglutinins may be sufficiently stable, over periods of one to several days at ambient room temperatures, to be of value to forensic science investigators. The results of the hemagglutinin assay are not affected by the age or sex of the sample donor. Because salivary hemagglutinins can be used to determine ABO blood type, analyses of this kind can serve as an important confirmatory test which the forensic serologist can use in conjunction with salivary agglutinogen determinations.  相似文献

Cocaine is rapidly degraded in blood samples, and its degradation was found to be highly dynamic in nature. The analysis of blood spots dried on filter paper may provide a method to minimize the break-down of cocaine and to largely preserve the analytical profile of the parent drug and its hydrolysis products at the time of sampling. The short term stability of cocaine in 100 microL blood spots prepared from unpreserved and preserved (sodium fluoride, 0.25%) blood samples was compared to the stability of the particular whole blood specimens stored in tubes at ambient temperature and at -20 degrees C. Due to dehydration, both the chemical and the enzymatic hydrolysis of cocaine and its products could be stopped in dried blood spots. More than 75% of the initial cocaine concentration could be detected in the blood spots, and the analytical profile was ensured for 17 days. Provided its practical suitability, the spot technology should offer a simple approach to detect actual impairment of motorists taken in police custody in the view of section 24a of the German traffic act as well as in cocaine associated criminal cases.  相似文献

DNA was extracted from human and non-primate dried blood stains. Human male and female specimens were readily distinguished by analysis with a Y-chromosome specific DNA probe. Human and non-primate blood stains were also readily differentiated using a repeat sequence (Alu) DNA probe. A sensitive microplate hemagglutination-inhibition technique has been used to ascertain the distributions of secreted blood group substances (BGS) in a population of 176 semen specimens and to characterize the stability of these substances in dried semen stains. The BGS concentrations in semen were found to vary throughout a wide range of titer. Despite this latitude of variation, the titers for the component BGS within the blood groups could be described by a log-normal distribution function. Studies of a number of sequential semen specimens obtained from the same donors revealed that the intraindividual variation in BGS titers was much more limited than the interindividual BGS titers. Attempts to correlate variations in titers between A and H in Group A semen or B and H in Group B semen indicated that the levels of these component substances vary independently. Studies of the stability of BGS in Groups A and O semen suggested that these substances were stable when the semen stains were stored at -20 degrees C, 4 degrees C, or at ambient laboratory temperature in a dry state. In contrast, stains stored at 37 degrees C under humid conditions suffered a dramatic loss in BGS titer, with the half-life of the BGS being on the order of 30 days.  相似文献

The efficiency of detection of blood in stains on material evidence by spectrofluorometry and fluorescent hemotest developed at Bureau for Forensic Medical Expert Evaluations of the Leningrad region and by traditional methods used in Russia and abroad is compared. The proposed methods are 1000 times more effective than the methods routinely used in Russia; moreover, they allow objective computer recording and processing. These methods are 50-70 times cheaper than the methods used in foreign countries and are virtually as sensitive. Fluorescent hemotest persuasively proves the presence of blood in stains on material evidence under laboratory conditions, at the site of accident, and even under field conditions (express analysis).  相似文献

Alpha-feto-protein can be detected in the dried bloodstains of fetal blood, blood from pregnant women, and in the amniotic fluid by radioimmunoassay. Alpha-feto-protein alone is not sufficient from exact differentiation. In addition, quantitative analysis of the total protein or electrophoresis is required.  相似文献

When a blood typing is made for mixed stains of sweat and blood, erroneous results may be obtained. The reason is that the blood group substance in the sweat is detected at the same time as that in the blood. Mechanical forces applied to the neck region are known to release certain amounts of thyroglobulin into circulation. In this experiment, an attempt was made to detect thyroglobulin in bloodstains as an aid in the diagnosis of mechanical asphyxia. Experimental bloodstains containing thyroglobulin at concentrations of 1, 2, 5 and 10 mu g/mL were prepared on a sheet of filter paper. Small pieces of bloodstains, measuring approximately 2.4 cm2 in area, were extracted with 0.1 mL of distilled water and the extracts were tested against an antihuman thyroglobulin serum by precipitation-electrophoresis. Bloodstains containing more than 1 mu g/mL of thyroglobulin formed distinct precipitin lines for up to one month of storage, while bloodstains containing more than 5 mu g/mL of thyroglobulin formed distinct precipitin lines for up to three months of storage. The present results suggest that the bloodstains can be utilized in the diagnosis of mechanical asphyxia.  相似文献
