利用细胞体外培养技术,分别对东方田鼠胚胎、皮肤、肺、腹腔、肾及睾丸组织进行成纤维细胞的分离和传代培养,再与前期建立的两种东方田鼠永生化细胞系在传代次数、冻存复苏以及体外杀伤日本血吸虫童虫等相关生物学特性方面进行比较。结果显示,采用组织块贴壁法或胰蛋白酶消化法,可在体外进行东方田鼠不同组织来源成纤维细胞的培养、传代和冻存,光镜下细胞均贴壁生长,呈长梭形,为典型的成纤维样细胞;不同组织来源的成纤维细胞在最佳分离培养日龄、原代细胞贴壁时间、细胞纯度、传代次数及冻存后细胞活率等方面存在明显差异;与对照组相比,所有东方田鼠成纤维细胞培养上清均没有显著的体外杀伤日本血吸虫童虫的活性。结果表明,东方田鼠不同组织成纤维细胞的体外培养方法及生物学特性不同。  相似文献

采用倒置相差显微镜、环境扫描电子显微镜对新生24 h内SD大鼠大脑皮质原代培养的神经元进行了形态学观察,建立了大脑皮质神经元原代培养方法。应用2.5μg/mL阿糖胞苷(Ara-C)对培养3 d的神经元进行24 h干预,去除神经胶质细胞、成纤维细胞、神经干细胞等杂质细胞,以提高神经元产率;用Nissl’s染色法进行神经元鉴定;利用微量滴定(MTT)法测定了一个培养周期(约10 d)的神经元活性分布。光学显微镜观察结果表明,培养至第4 h绝大多数神经元已贴壁生长,胞体上可见1~2个细小突起;培养至第3 d、第6 d时,Nissl’s染色结果显示,加Ara-C能显著提高神经元纯度(≥92%);MTT结合形态学观察结果表明,神经元培养至第6 d时活性最强,细胞呈卵圆形、蜘蛛状、锥体状等多种形态,细胞胞体饱满,光晕明显,神经网络形成完整。  相似文献

无菌采取5日龄SD大鼠的颅盖骨,用胰蛋白酶预消化后,漂洗,剪碎,再用Ⅱ型胶原酶消化获得成骨细胞。通过倒置相差显微镜观察,并用碱性磷酸酶和矿化结节染色鉴定,利用微量滴定(MTT)法测定一个培养周期的成骨细胞活性分布,探讨一种经济、高效的原代成骨细胞培养方法。倒置相差显微镜形态观察结果显示,未贴壁的细胞呈圆形,贴壁生长的细胞呈不规则梭形、三角形或多角形,单核,有1~3个核仁;碱性磷酸酶和钙化结节染色均呈阳性(阳性率≥98.06%);MTT法结合形态学观察结果显示,成骨细胞培养至第10 d时活性最强。证实用改良的二次酶消化法能够在短时间内获得大量成骨细胞,且所培养的细胞具有典型的成骨细胞形态和功能。  相似文献

从胎龄3个月左右的山羊胚胎中分离出背根神经节,采用组织块培养法,利用血清培养和无血清培养,并在非神经细胞抑制剂作用的条件下,观察了山羊背根神经节神经细胞在体外的生长状况,采用免疫荧光和RT-PCR技术对神经细胞表面标志神经元特异性磷酸化酶(NSE)进行了鉴定。结果显示,接种48h后的细胞从组织中迁出,并长出突起,经阿糖胞苷作用后,神经细胞与非神经细胞分层;经免疫荧光鉴定,上层细胞为神经细胞。随着时间的增加,神经细胞突起延长并交错成网状,至第6~10天神经细胞形态最为成熟饱满,随后逐渐出现细胞老化,神经细胞最长可生存32d。表明本试验成功分离培养得到了山羊背根神经节神经细胞。  相似文献

为寻找一种经济、可重复的鸭心肌细胞最适培养技术,利用胰蛋白酶消化法和差速贴壁法分离樱桃谷鸭心肌细胞,并对培养条件和生长特性进行摸索。用台盼蓝染色法检测培养细胞的活性,用HE染色法检测培养细胞的形态,并用抗α-actin蛋白免疫组织化学染色法鉴定培养的心肌细胞。结果表明,通过差速贴壁法分离的心肌细胞在前3代可观察到节律性搏动,α-actin染色阳性,心肌细胞数量较大,活力高,形态呈梭形、星形和多角形。证实,通过酶消化法和差速贴壁法分离到的鸭心肌细胞,可连续传代至12代,细胞纯度及数量能满足心肌细胞研究的需要,为一种可行的鸭心肌细胞培养方法。  相似文献

以30日龄仔猪睾丸组织块为培养对象,应用飞利普TECNAI10电子显微镜对培养前后的生精细胞类别进行了观察,对培养后的组织块进行生精细胞分化程度鉴定,建立了仔猪睾丸组织生精细胞爬片体外培养模型。培养前后电镜观察及HE染色结果显示,培养前细胞种类主要有2种:支持细胞和精原细胞,有少数几个初级精母细胞;培养第20 d的睾丸组织块HE染色观察到3个精子细胞,电镜观察到2个精子细胞。证实,在没有添加睾酮的情况下,此培养体系能够提供睾丸组织块生精细胞分化所需要的条件,使非精子细胞类生精细胞减数分裂为精子细胞。  相似文献

对猪卵丘细胞和胎儿成纤维细胞的分离、培养和传代进行了系统研究。结果显示,猪的卵丘细胞生长形成单层需要5~6 d;运用组织块法和酶消化法获得胎儿成纤维细胞单层的生长时间分别为10~11 d和8~9 d。猪的卵丘细胞和胎儿成纤维细胞均可以建立并获得稳定的细胞系,这些方法的应用可为猪体细胞核移植研究提供丰富的供体细胞。  相似文献

为建立一种稳定、方便地获得较高纯度的绵羊绒毛膜滋养层细胞的方法,为后续的细胞转染和RNA干扰试验提供细胞学基础,采用组织块法培养滋养层细胞,消化差速法纯化细胞,相差显微镜观察滋养层细胞的形态学特点,应用常规HE染色和免疫组织化学染色、透射电镜及PCR技术鉴定细胞来源。结果显示,倒置相差显微镜下,可见细胞为上皮样细胞形态,呈片状铺展生长。免疫组织化学染色显示,细胞角蛋白染色阳性细胞占90%以上,波形蛋白染色呈阴性;透射电镜可观察到滋养层细胞所特有的结构;台盼蓝排斥试验检测细胞活力,存活率超过95%,细胞活力良好。常规RT-PCR技术可扩增出滋养层细胞所分泌的特有的干扰素蛋白-1基因片段。证实该方法可以有效获得较高纯度的、具有生物学活性的绵羊绒毛膜滋养层细胞,为本实验室后续的体外研究提供试验基础。  相似文献

The EU's strategy for Russia since the end of the cold war has to a large extent failed, and the direct consequences are the currently deadlocked EU-Russia relationship. There are three reasons for this: the EU and Russia's respective strength and status relative to each other have been changing since the beginning of this century; the EU's strategy towards Russia has been more fragmenting in recently years; and the American factor. The EU-Russia relations could continue to be in poor shape in the years to come, and it is very likely sometimes even further worsening. Most of the pages are devoted to a detailed comparison between the old generation and the young generation of American China Watchers regarding to their background, education, and views over China. 1. Introduction
On the morning of April 1st, 2001, a U.S. EP-3 surveillance aircraft was conducting an intelligence flight along the Chinese territorial border in the airspace south-east of China's Hainan Island. Two Chinese fighter jets intercepted the spy plane and followed its flight, as is considered ordinary international practice. For reasons unknown, the EP-3 airplane made a sudden maneuver and crashed into one of the Chinese planes. The Chinese plane dropped into the sea, the pilot missing. The U.S. spy plane was also severely damaged. It intruded into Chinese territorial space and landed on Hainan Island without official permission from the Chinese side. After landing, the spy plane and 24 crew on board were detained by the Chinese authorities. This air collision episode was the first of its kind in the China-U.S. relations and an urgent diplomatic crisis ensued.  相似文献
