The relationship between components found on vaginal swabs was examined to determine whether the presence and quantity of a particular component could be used to predict the presence of others or to help interpret grouping results. Vaginal swabs were tested for the presence of blood, cellular material, spermatozoa, acid phosphatase, p30, ABH, Lewis, EsD, GLO I, PGM and PGM subtype. Methods included rocket electrophoresis and p-nitrophenyl phosphate assay for acid phosphatase; rocket and cross-over electrophoresis for p30. Results from 323 semen-free and postcoital vaginal swabs from known donors are presented. Comparison of methods showed that seminal acid phosphatase and p30 were detected more often by the rocket techniques. Attributing the grouping results to semen, based on the reactivity of any single component, could lead to erroneous conclusions. Activation of vaginal components in the presence of semen and endogenous vaginal levels are discussed.  相似文献

Comparative investigation of seminal stains and other secretions from human body and blood as well as objects of plant origin by gel electrophoresis using alkaline values of pH buffer solutions and subsequent enzymography for acid phosphatase made it possible to develop optimal conditions for detection of seminal presence in stains. Good preservation of seminal acid phosphatase in stains during several years is shown.  相似文献

A novel approach to the presumptive screening of questioned semen stains has been developed which enables the rapid identification of stains which are devoid of semen. Questioned semen stains can be swabbed with a moist cotton swab, and the prostatic acid phosphatase (SAP) activity transferred to the swab identified through assay with 5-bromo-4-chloro-3-indolyl phosphate (BCIP). Controlled laboratory studies revealed that the BCIP swab procedure was as sensitive as the semiquantitative SAP test currently employed in the FBI Laboratory for the presumptive screening of semen stains. A validation study of the BCIP swab procedure in parallel with the current procedure using 4305 case evidence stains indicated that the BCIP swab procedure was as effective as the current procedure in identifying those questioned stains which lack semen. The advantage of the BCIP swab procedure is that it can be performed on questioned stains in situ and thereby avoids the requirement of removing and extracting the stain before assay of SAP activity.  相似文献

Although elevated acid phosphatase (AP) activity in vaginal fluid is a consistent indicator for semen, differentiation between vaginal AP and seminal AP provides a more meaningful result. Detection of seminal AP in mixtures of vaginal AP, feces, and blood is accomplished by starch gel electrophoresis, employing the substrate thymolphthalein monophosphate as a selective visualization agent. Genetic phosphoglucomutase isoenzymes are simultaneously separated by this method and allow differentiation in some semen/vaginal fluid mixtures.  相似文献

The modified zinc test and a commercially available acid phosphatase test were compared as to their screening parameters according to the microscopical finding of spermatozoa in cases of alleged sexual assault. The study involved 65 pieces of evidence material. It was found that the modified zinc test has a higher sensitivity and higher predictive values than the acid phosphatase test. However, when both tests are combined in parallel, the sensitivity and the negative predictive value could be raised to 99%. This finding suggests that a negative result obtained from the parallel combination of the modified zinc test and the acid phosphatase test predicts very well that no spermatozoa will be found at microscopical examination. Since the latter technique is the only one to give absolute proof of the seminal origin of stains or traces, the parallel combination of the zinc test and the acid phosphatase test might be useful in sorting out these cases or materials that deserve further investigation by more elaborate techniques.  相似文献

Acid phosphatase (AP) reagent (Fast Black) is used as a presumptive test for the presence of seminal fluid on exhibits submitted in allegations of sexual assault. Research was carried out to determine whether the direct application of AP reagent to exhibits is a viable alternative to the traditional indirect (blot) testing method used routinely in the laboratory. The relative sensitivity of the indirect and direct testing methods was investigated as was the effect of AP reagent on histological staining of spermatozoa, the incidence of false positives from vaginal material and saliva, and the effect of AP reagent on subsequent DNA testing. Also included are the results of specificity studies from validations of the direct AP testing method. The results of this research show that, provided the incidence of false positives is borne in mind, direct AP testing can be especially useful when screening exhibits which are difficult to indirectly (blot) AP test or when it is problematic to relocate an AP positive stain. Direct application of AP reagent can also be beneficial for locating dilute semen stains. Three case examples are given which illustrate the use of direct AP testing in laboratory casework.  相似文献

Prostate specific antigen (PSA, also known as p30), a glycoprotein produced by the prostatic gland and secreted into seminal plasma, is a marker used for demonstrating the presence of seminal fluid. Methods for the detection of PSA include Ouchterlony double diffusion, crossover electrophoresis, rocket immuno-electrophoresis, radial immunodiffusion, and ELISA. The extremely sensitive ELISA technique can detect PSA in concentrations as low as approximately 4 ng/mL. However, all these techniques are cumbersome and time consuming to perform in forensic laboratories, especially when only a few samples per week are processed. Various membrane tests are currently used in clinical settings to screen a patient's serum for the presence of PSA at levels greater than 4 ng/mL. In this study we evaluated three immunochromatographic PSA membrane tests by analyzing semen stains stored at room temperature for up to 30 years, post-coital vaginal swabs taken at different time after intercourse, semen-free vaginal swabs, and various female and male body fluids, including urine. The data demonstrate that PSA membrane test assays offer the same sensitivity as ELISA-based tests and provide a rapid approach for the forensic identification of seminal fluid. Furthermore, when the supernatant from a DNA extraction is used for the assay, there is essentially no DNA consumption for determining the presence of PSA in a forensic sample.  相似文献

The application of p30 detection to casework analysis of seminal traces on vaginal swabs is reported and compared with the levels of acid phosphatase determined.A simple crossed-over immunoelectrophoresis system was used for batch identification of swab extracts using a commercially obtained anti-p30 serum.Positive p30 results were obtained in less than 20% of the casework swab extracts, but they provide conclusive proof of the presence of semen which is a substantial advantage over the quantitative determination of acid phosphatase.  相似文献

The detection of haptoglobin (Hp) from serum and bloodstains is utilized extensively in forensic science laboratories in order to include or exclude possible donors. There is an increasing need to make the same discriminations utilizing genetic markers from urine samples. This paper describes the use of enzyme immunoassay and Western blotting (electrophoretic) techniques to determine Hp phenotypes from concentrated urine samples. Serum and urine specimens were collected from volunteer donors. The serum sample from each donor was typed for Hp. The urine specimens were concentrated 3000-fold from the starting volume of 15 mL to a final volume of 5 microL and applied to the gradient polyacrylamide gels. This procedure allows the separation of Hp samples into the three common phenotypes as well as the other rare variants found in humans. The Western blotting electrophoretic technique was used to achieve the transfer of Hp bands from the gels to the nitrocellulose membranes. Enzyme immunoassay with goat anti-Hp antiserum and rabbit anti-goat immunoglobulin alkaline phosphatase conjugate were used to identify the Hp bands from the concentrated samples. Specimens stored for six months at -22 degrees C were also concentrated and typed successfully. Recent implementation of drug-screening policies has resulted in an increase in the submission of substituted urine specimens. The above procedure can be used to detect an additional genetic marker from urine samples and thus facilitate the identity of the donor.  相似文献

The application of an enzyme-linked immunosorbent assay (ELISA) for 19-OH Prostaglandin F1 alpha/F2 alpha (19-OH PG F) to casework analysis of seminal contamination of swabs and stains is reported. The results are compared to those where the identification of semen was based on the presence of acid phosphatase and spermatozoa. Five hundred and one samples were analysed and there was good agreement between the results of ELISA and conventional methods. The determination of 19-OH PG F confirmed both the presence of semen where spermatozoa were absent and indicated semen was present when acid phosphatase and spermatozoa were both absent. The results indicate that 19-OH PG F represents a useful marker for the casework identification of semen and is particularly valuable where spermatozoa are absent.  相似文献

The value of the acid phosphatase spot test in the primary visualization and identification of seminal traces is hampered by the sensitiveness of the enzyme to biodegradation. An alternative spot test is proposed, based on the high concentration of the more stable zinc metal in seminal plasma. The proposed zinc spot test is simple and suitable for on site investigation. Although the sensitivity in fresh stains is lower than that of the acid phosphatase spot test, this is largely compensated by the lower sensitiveness to biodegradation. The specificity for semen is higher than that of the acid phosphatase spot test. In vaginal swabs it was nevertheless seen, that samples should be taken within 24 h after alleged sexual assault to give reliable results.  相似文献

The detection of potentially harmful substances presents a multifaceted challenge. On one hand, it can directly save lives, on the other, it can significantly aid and enhance police work, thereby increasing the effectiveness of investigations. The research conducted in this study primarily aims to identify paracetamol in fingerprints, considering situations involving direct contact of a person with paracetamol either chronically or in a single dose. The identification procedure presented, utilizing Raman spectroscopy, aims to rapidly detect the xenobiotic following ingestion by an individual, which involves touching the tablet with their fingers—this can be termed as touch evidence in forensic science investigations. Additionally, the authors focus on assessing the impact of additives present in drugs containing paracetamol as the main active ingredient. The screening results obtained will enable us to analyze the composition of drugs in terms of potentially toxic substances, and their influence on the physicochemical activity of the active substance. We successfully identified the paracetamol molecule using a noninvasive forensic trace detection method. Samples in the form of common drugs containing 500 mg of paracetamol were studied. Throughout the study, comprehensive validation of the method was ensured through the utilization of a statistical model, which excluded sensitivity to the presence of other substances, whether additives or from the external environment. The proposed approach to trace the content of substances in fingerprint using Raman scattering analysis provides a useful starting point to enhance current analytical methods not only in forensic science but also in toxicology.  相似文献

The sensitivity and specificity of a zinc spot test for the detection of semen were compared with those of an acid phosphatase detection method. As screening techniques both tests were found to be very sensitive, but the zinc test was more specific and was more reliable in older and especially in deteriorated specimens. It is concluded that the zinc spot test deserves at least the same place as the acid phosphatase test in the primary investigation of suspected semen stains and might well be the test of choice in older and poorly preserved stains.  相似文献

The Body Fluid Forum of the Association of Forensic Science Providers recognised concerns raised by forensic practitioners regarding the lack of data to inform on the incidence of significant AP (Acid Phosphatase) reactions from vaginal and oral samples, i.e. those which might be misinterpreted as indicating the presence of semen. This is particularly relevant in the light of appeal court rulings regarding the need for data to support evaluations. This paper presents collaborative data on the nature and incidence of AP reactions from vaginal and oral swabs provided by donors. The results demonstrate that caution is required with mid to strong purple AP reactions from direct testing of oral swabs and with mid purple reactions from vaginal swabs. The use of a Bayesian approach to assist with the provision of opinions regarding the presence of seminal fluid is highlighted.  相似文献

When tablet residues are found in the gastrointestinal tract during autopsy, this does not only indicate the possible presence of intoxication, but may also provide indications with the regard to the kind of intoxication (e.g. suicide) if the amount of tablets is considered. If tablets have already dissolved and thus large portions can no longer be detected with the naked eye, a definitive appraisal with regard to the presence of tablet residues is often difficult or even impossible on the autopsy table. A polarization microscopic screening method is described which enables identification of characteristic tablet auxiliary substances (maize starch, sodium carboxymethyl starch, microcristalline cellulose or sodium carboxymethyl cellulose) to be identified immediately and simply in the gastrointestinal contents. It also enables a rapid orientative screening for tablet residues in glasses found or the fluid these contained as well as in aspirated material and vomit. If the active agent of the tablet can be detected by chemical toxicology, the polarization microscopic diagnosis of abundant tablet auxiliary substances is compatible with intake of large amounts of drugs, which makes self-administration highly probable.  相似文献

The identification of seminal traces is exceptionally difficult, if the semen of the assailant is azoospermic. The evident value of prostatic acid phosphatase (PAP) activity must be evaluated in such cases with caution. In a murder investigation of a 13 year old girl a positive PAP reaction was found in vaginal swabs and in her underpants. Spermatozoa could not be found. Using the gas-chromatographic method, described by Douse (1985) the presence of prostaglandin E could be demonstrated in the swabs as well as in the crotch of the underpants. The offender was found to be a man with azoospermia, who admitted intercourse but with the consent of the victim. The E prostaglandins are mainly synthesized in the vesiculae seminal and seen to be specific for semen. Swabs taken from mouth and rectum showed negative reactions for prostaglandins in this case. Prostaglandins could never be detected in vaginal swabs taken at least 7 days after intercourse. Conversely Douse could detect prostaglandins in swabs up to 58 hours after intercourse. Apparently the prostaglandin detection by Douse provides a suitable alternative besides to the quantitative and immunological PAP detection or the immunological detection of the protein p 30.  相似文献

This study compares the detection of choline in seminal stains by both an enzymatic method and by the standard Florence crystal test. The tests were conducted on 293 actual casework samples. In those samples identified as containing semen, choline was detected twice as often by the enzymatic method compared to the Florence method (84.6 versus 40.3%). The choline results were correlated with spermatozoa and acid phosphatase tests. The enzymatic detection of choline in seminal stains was found to be a fast, easy, sensitive, and reliable test.  相似文献
