根据GenBank中已登录的猪生殖与呼吸综合征病毒(PRRSV)全基因组序列,设计合成了1对特异性引物,对PRRSV非结构蛋白nsp4基因进行了RT-PCR扩增,将回收的目的基因片段克隆到大肠埃希氏菌表达载体pET28a中,构建了重组质粒pET-nsp4,测序结果证实了重组质粒pET-nsp4的可靠性。将pET-nsp4转化至大肠埃希氏菌表达菌株BL21(DE3),用IPTG诱导表达,SDS-PAGE结果表明,重组菌可表达分子质量约23 ku的蛋白。经镍离子亲和层析柱(Ni-NTA)纯化,获得了高纯度的重组蛋白,Western-blot分析结果表明,nsp4重组蛋白在大肠埃希氏菌系统中获得了正确表达。  相似文献

根据已克隆的堆型艾美球虫 (Eimeriaacervulina)广东株子孢子表面抗原基因cSZ1的cDNA序列设计了特异性引物 ,用PCR方法扩增cSZ1的开放阅读框架 (ORF)后克隆至表达载体pET 32a( ) ,构建了重组表达质粒 pET 32a( ) cSZ1,并将其转化至大肠埃希氏菌BL2 1(DE3)。经IPTG诱导 ,获得了cSZ1重组抗原在大肠埃希氏菌中的高效表达 ,表达产物量可达菌体总蛋白的 9.3% ,融合蛋白的分子质量约为 4 0ku。重组菌诱导表达的产物经SDS PAGE后 ,用堆形艾美球虫感染鸡的超免疫血清进行免疫印迹分析 ,结果为阴性 ,提示cSZ1所编码的抗原可能主要是T细胞抗原表位  相似文献

将猪繁殖与呼吸综合征病毒(PRRSV)ORF6基因从pGEM-T-ORF6重组质粒中亚克隆到pET-5a原核表达载体上,成功构建重组表达质粒pET5-ORF6.将其转化大肠埃希氏菌BL21(DE3)感受态细胞,重组菌经IPTG诱导表达,其细菌裂解物经SDS-PAGE可检测到分子质量约为19 ku的目的蛋白带,经薄层扫描分析,目的蛋白占菌体总蛋白的26.8%,采用重组菌裂解物作为包被抗原进行ELISA检测,结果表明表达的蛋白显示特异性.  相似文献

采用PCR方法分别对类志贺氏菌毒素A亚单位基因第 2 98~ 80 5序列、818~ 12 0 1序列进行了扩增 ,使其第 80 6~ 817序列的碱基 (编码 16 7~ 170位氨基酸 )缺失 ,并将两片段连接后克隆至质粒载体pGEX 6P 1中 ,获得了含有重组质粒 pGEX Ade的重组大肠埃希氏菌 ;经SDS PAGE以及使用特异性抗SLT ⅡeA单克隆抗体的Western blotting ,证明该重组大肠埃希氏菌在IPTG诱导条件下可表达SLT ⅡeA。  相似文献

以含长白猪IFN-γ基因的质粒载体为模板,用原核表达引物扩增了长白猪IFN-γ基因,将其定向克隆至原核表达载体pET28b的EcoRⅠ和XhoⅠ位点,构建了长白猪IFN-γ基因的原核表达载体pET28b-PoIFN-γ。经酶切、PCR鉴定,表明所构建的重组质粒为特异性长白猪IFN-γ基因原核表达质粒。将该重组质粒转化大肠埃希氏菌BL21(DE3)细胞,阳性菌落筛选、SDS-PAGE分析以及Western-blotting分析表明,成功构建了表达重组猪IFN-γ的大肠埃希氏菌基因工程菌株。亚细胞定位分析表明,目的蛋白经原核表达后主要以不溶性包涵体形式存在,其他少量可溶性目的蛋白存在于细胞浆中。  相似文献

根据绵羊肺腺瘤病毒(SJRV) NM株基因组全序列,设计了 1 对包含囊膜(env)基因在内的引物,并在5′端分别设计了HindⅢ和 BamHⅠ酶切位点,利用 PCR技术扩增 env基因,通过连接反应将其克隆于pET 30b表达载体上。序列分析表明,env基因全长 1 836 bp,编码 612 个氨基酸残基;构建表达重组质粒env pET 30b,转化大肠埃希氏菌 BL21,经 IPTG于 37 ℃诱导培养,SDS PAGE电泳分析表明,表达的env基因融合蛋白分子质量约为69 ku。  相似文献

以胸膜肺炎放线杆菌(APP)血清1型标准菌株基因组为模板,用PCR扩增ApxⅠBD基因的特异性片段;对PCR产物纯化回收后与载体pMD 18-T进行连接、转化,经酶切和序列分析鉴定后亚克隆到原核表达载体pET28a中,构建重组表达质粒pETⅠBD;转入大肠埃希氏菌BL21中,以IPTG诱导表达,进行SDS-PAGE电泳。结果,从APP中克隆的ApxⅠBD DNA序列与已发表序列的同源性为100%,长1 447 bp,编码482个氨基酸,所表达的融合蛋白相对分子质量约54ku,与预期的大小相符。结果表明,含有ApxⅠBD基因特异性片段的重组表达载体pETⅠBD已成功构建。  相似文献

采用PCR方法,以牛疱疹病毒Ⅰ型基因组DNA为模板扩增gB基因,克隆至pGEM-T载体。将以克隆质粒pGEM-T-gB为模板扩增的gBⅠ、gBⅡ和gBⅢ基因片段分别连接至原核表达载体pET32a。获得的重组质粒分别转化感受态细胞BL21(DE3),在IPTG诱导下表达。经SDS-PAGE鉴定,gBⅠ、gBⅡ和gBⅢ基因片段在大肠埃希氏菌BL21(DE3)中以融合蛋白形式获得了表达。以磁化组氨酸蛋白纯化系统对融合蛋白纯化后,Western-blotting和ELISA分析结果表明,这些融合蛋白均与BHV-1标准阳性血清反应,可作为抗原用于牛传染性鼻气管炎ELISA检测方法的建立。  相似文献

构建了TGEV TH-98株M基因序列的原核表达载体,利用RT-PCR方法从TGEVTH-98株中扩增出M蛋白基因,并亚克隆至原核表达载体pGEX-6P-1上,转化大肠埃希氏菌BL21(DE3)pLysS,用IPTG诱导重组菌株表达了融合蛋白。结果表明,从TGE TH-98株克隆到了M基因cDNA,该基因全长789 bp,与T014株、Purdue株、TFI株和96-1933株的M基因同源性分别为92.37%、98.35%、99.87%和96.96%。SDS-PAGE电泳结果显示,重组表达质粒获得了约57 ku的目的带,其中M蛋白的分子质量约为31 ku,与预期的结果一致;Western-blotting分析表明,融合表达产物可与TGEV全病毒制备的阳性血清发生反应。证实,TGEV的M基因高度保守,构建的M基因原核表达载体pGEX-6P-TM能在大肠埃希氏菌中获得高效表达。  相似文献

利用RT-PCR扩增了2株H5N1亚型禽流感病毒NS1基因,并将其克隆到pMD 18-T载体上,进行序列分析。结果显示,这2株禽流感病毒NS1基因核苷酸序列的同源性为70.2%,分别属于NS等位基因群A和等位基因群B。再将克隆的NS1基因插入到pET-28a质粒中构建原核表达载体,将其转化到DH5α大肠埃希氏菌感受态细胞中,经双酶切鉴定及序列分析,表明获得了重组质粒pET-52NS1和pET-174NS1。经SDS-PAGE分析,重组质粒转化BL21(DE3)(pLysS)感受态细胞后,经IPTG诱导,获得了分子质量约为30 ku的NS1融合蛋白。用AIV多克隆血清做Western-blotting分析,发现来自2个等位基因群的NS1蛋白都具有较好的抗原活性。  相似文献
