广东广西地区5个群体9个STR基因座的频率调查   总被引:11,自引:0,他引:11  
目的 调查广东汉族、广西汉族、广西侗族、广西壮族、广西苗族5个群体9个STR基因座多态性,探讨其在法医学检验中的应用价值。方法 应用AmpFISTR Profiler PlusTM荧光标记复合扩增系统,对广东广西5个群体4个民族的1191个无关个体的血样DNA进行9个STR基因座的复合扩增;用ABI 3100遗传分析仪对扩增产物进行分型,统计9个STR基因座的群体遗传学参数。结果 9个STR基因座在广东广西地区5个群体中的累积偶合率为1.51×10-11~8.08×10-11,累积非父排除率为0.99981—0.99990。,结论 该9个STR基因座可满足汉族、壮族、侗族、苗族群体法医学的个体识别及亲权鉴定的需要。  相似文献

In the present study, forensic parameters were estimated for three populations residing in the United Arab Emirates (UAE) including UAE Arabs, Pakistanis and Indians based on the population data of 23 autosomal short tandem repeats (STRs). The UAE Arabs is a vital population to study due to high rates of consanguineous marriages. Therefore, it is essential to estimate the allele distribution and frequencies within this population. In addition, it is crucial to study the largest communities living in the UAE such as Indians and Pakistanis. A total of 1272 blood samples were collected on FTA® cards, comprising of 571 UAE Arabs, 352 Indians and 349 Pakistanis. All of these samples were amplified directly using Verifiler® Express PCR Amplification Kit that focuses on 23 autosomal STR loci, namely D3S1358, vWA, D16S539, CSF1PO, TPOX, D8S1179, D21S11, D18S51, D2S441, D19S433, TH01, FGA, D22S1045, D5S818, D13S317, D7S820, D10S1248, D1S1656, D12S391, D2S1338, D6S1043, Penta D and Penta E loci. The PCR products were electrophoresed on ABI 3500 Genetic Analyzer and analyzed using GeneMapper ID-X v1.4 software. Arlequin v3.5 and PowerStats software were utilized to determine the forensic parameters and population structure using AMOVA. Gene diversity, ranged from 0.67406 (TPOX) to 0.9226 (Penta E) in the UAE Arabs, 0.69955 (TPOX) to 0.9214 (Penta E) in Indian and 0.69853 (TPOX) to 0.921 (Penta E) in Pakistani population. The most discriminating autosomal STR loci observed was Penta E (PD = 0.985), (PD = 0.986), (PD = 0.986) in the UAE Arabs, Indian and Pakistani population, respectively. The obtained results showed the 23 STR loci had a relatively high genetic variation, confirming the suitability for forensic identification and kinship analysis, in the relevant populations. The significance of this study is to build an allelic frequency database for one of the most powerful commercially available STR amplification kits by using the current forensic workflow.  相似文献

目的 调查青岛地区汉族人群无关个体的 13个STR基因座 (D3S135 8、VWA、FGA、D8S1179、D2 1S11、D18S5 1、D5S818、D13S317、D7S82 0、D16S5 39、TH0 1、TPOX、CSFIPO)的基因频率分布 ,研究其遗传多态性及其在法医学个体识别及亲子鉴定中的应用价值。 方法 用美国ABI - 310型遗传分析仪对ProfilerPlus和Cofiler两个系统的 13个STR基因座的复合扩增产物进行毛细管电泳及四色荧光自动分析检测 ,基因分型软件为GeneScanv3.1和Genotyperv2 .5 .2。  结果 获得 13个STR基因座在青岛地区汉族人群的基因频率分布数据 ,13个STR基因座的PIC >0 .5 ,DP >0 .71,CCE =0 .999999,TDP值接近 1,TPm =1.2× 10 -14 ,家系调查符合孟德尔遗传规律。 结论 ProfilerPlus和Cofiler两个系统的 13个STR基因座在法医学个体识别及亲子鉴定中具有较高的应用价值。  相似文献

目的调查湖南地区汉族人群21个STR基因座(D3S1358、D13S317、D7S820、D16S539、Penta E、D2S441、TPOX、TH01、D2S1338、CSF1PO、Penta D、D10S1248、D19S433、v WA、D21S11、D18S51、D6S1043、D8S1179、D5S818、D12S391和FGA)的遗传多态性。方法共采集560例湖南汉族健康无关个体血液样本,使用Chelex-100法提取DNA,应用AGCU EX22试剂盒及9700 PCR扩增仪进行复合扩增,扩增产物使用310遗传分析仪进行分离分析。结果共发现248个等位基因,等位基因频率分布在0.001~0.518。除Penta E(P=0.023)外,其余基因座的基因型分布均符合Hardy-Weinberg平衡。21个基因座的累积个人识别率、累积非父排除率、累积匹配率分别为0.999 999 999 999 999 999 999 999 8、0.999 999 998和1.36×10-25。结论 21个STR基因座在湖南汉族人群中呈高度多态性。本研究可为法医学个人识别及亲子鉴定提供有价值的数据及理论基础。  相似文献

目的调查21个非CODIS系统STR在浙江汉族人群的遗传多态性,为其法医学应用提供基础数据。方法应用AGCU21+1荧光标记复合扩增系统,对浙江汉族481名无关个体进行21个STR基因座的复合扩增,用ABl3130XL型基因分析仪对扩增产物进行检测,并统计其STR基因座的群体遗传学参数。结果获得21个STR基因座的等位基因频率分布,分别检出8、11、9、10、8、9、6、5、13、9、6、15、8、7、8、9、8、9、9、16、7个等位基因,并分别获得21个STR基因座的H、He、DP、PM及EP等法医遗传学参数。结论21个STR基因座具有较强个体识别能力,可应用于法庭科学中的个体识别与亲权鉴定。  相似文献

目的 构建6个五核苷酸STR基因座荧光复合扩增体系。方法筛选6个多态性程度较高的五核苷酸STR基因座D10S2325、Penta B、Penta W、PentaX、Penta D和PentaE,按照复合扩增引物设计要求,重新设计引物并标记荧光染料,经反复调整和优化,构建6基因座荧光复合扩增体系,并用该复合扩增体系对239名武汉汉族无关个体进行分型。结果6个五核苷酸STR基因座荧光复合扩增体系分型稳定,可重复性好,与各自相应单基因座分型结果完全一致;累积个人识别率达0.999999988,累积非父排除率达0.998063807。结论本文构建的6个五核苷酸STR基因座荧光复合扩增体系具有很高的法医学实用价值,可作为商品化试剂盒的有效补充。  相似文献

DNA typing of degraded DNA samples can be a challenging task when using the current commercially available multiplex short tandem repeat (STR) analysis kits. However, the ability to type degraded DNA specimens improves by redesigning current STR marker amplicons such that smaller sized polymerase chain reaction (PCR) products are generated. In an effort to increase the amount of information derived from these types of DNA samples, the AmpFlSTR MiniFiler PCR Amplification Kit has been developed. The kit contains reagents for the amplification of eight miniSTRs which are the largest sized loci in the AmpFlSTR Identifiler PCR Amplification Kit (D7S820, D13S317, D16S539, D21S11, D2S1338, D18S51, CSF1PO, and FGA). Five of these STR loci (D16S539, D21S11, D2S1338, D18S51, and FGA) also are some of the largest loci in the AmpFlSTR SGM Plus kit. This informative nine-locus multiplex, which includes the gender-identification locus Amelogenin, has been validated according to the FBI/National Standards and SWGDAM guidelines. Our results demonstrate significant performance improvements in models of DNA degradation, PCR inhibition, and nonprobative samples when compared to the AmpFlSTR Identifiler and SGM Plus kits. These data support that the MiniFiler kit will increase the likelihood of obtaining additional STR information from forensic samples in situations in which standard STR chemistries fail to produce complete profiles.  相似文献

Allele and genotype frequencies for the ten STR loci D3S1358, VWA, D16S539, D2S1338, D8S1179, D21S11, D18S51, D19S433, TH01, FGA were determined in a Swiss Caucasian population sample (n=206) using the AmpFISTR SGM Plus Amplification kit. Electrophoresis was carried out on an ABI PRISM CE 310 Genetic Analyzer instrument. Previously, allele frequencies were published for the 13 STR loci D3S1358, VWA, FGA, D8S1179, D21S11, D18S51, D5S818, D13S317, D7S820, THO1, TPOX, CSF1PO and D16S539 for the same samples (n=206) amplified with the AmpFISTR Profiler Plus and Cofiler PCR Amplification kits. Since the results for the eight loci D3S1358, VWA, FGA, D8S1179, D21S11, D18S51, THO1, D16S539 shared between the AmpFISTR SGM Plus, Profiler Plus and Cofiler PCR Amplification kits already are published, only the allele frequencies for the two STR loci D2S1338 and D19S433 are reported in this paper. The two loci meet Hardy-Weinberg expectations. In addition, there is little evidence for association of alleles among the 15 loci (amplified with the Profiler, Cofiler, and SGM Plus amplification kits). The allelic frequency data can be used in forensic analyses to estimate the frequency of a multiple STR locus DNA profile in the Swiss population.  相似文献

目的为研发复合扩增荧光检测试剂盒,对现有的STR基因座进行分析研究并优选新的高鉴别力基因座。方法收集汉族、锡泊族、畲族、壮族、藏族等5个民族群体血样共1200份,提取DNA,应用复合扩增方法检测1200名5个民族群体无关个体的24个基因座的等位基因分布。结果TPOX和TH01基因座的等位基因在5个民族群体中分布不平衡;D2S1338、D6S1043和Penta E等3个STR基因座在5个民族群体中均具有高度遗传多态性,等位基因频率分布均匀,在各群体间无显著差异,而且等位基因传递遵循孟德尔遗传规律。结论确定出DNATyperTM15试剂盒中的14个适合中国人群体遗传学特征和法医学应用的STR基因座。  相似文献

Abstract: Analysis of length polymorphism at short tandem repeat (STR) loci utilizing multiplex polymerase chain reaction (PCR) remains the primary method for genotyping forensic samples. The AmpF?STR® Identifiler® Plus PCR Amplification Kit is an improved version of the AmpF?STR® Identifiler® PCR Amplification Kit and amplifies the core CODIS loci: D3S1358, D5S818, D7S820, D8S1179, D13S317, D16S539, D18S51, D21S11, CSF1PO, FGA, TH01, TPOX, and vWA. Additional loci amplified in the multiplex reaction are the sex‐determinant, amelogenin, and two internationally accepted loci, D2S1338 and D19S433. While the primer sequences and dye configurations were unchanged, the AmpF?STR® Identifiler® Plus PCR Amplification Kit features an enhanced buffer formulation and an optimized PCR cycling protocol that increases sensitivity, provides better tolerance to PCR inhibitors, and improves performance on mixture samples. The AmpF?STR® Identifiler® Plus PCR Amplification Kit has been validated according to the FBI/National Standards and Scientific Working Group on DNA Analysis Methods (SWGDAM) guidelines. The validation results support the use of the AmpF?STR® Identifiler® Plus PCR Amplification Kit for human identity and parentage testing.  相似文献

Autosomal short tandem repeats (STRs) have been widely used in forensic investigations. Prior to the application of any DNA based identification method, it is essential to estimate the allele frequencies and forensic statistical parameters of targeted STR loci in each population in order to provide a more precise reference database for forensic investigation. The GlobalFiler™ Kit is a multiplex assay that combines the 13 original CODIS loci with 7 non-overlapping loci from the expanded European Standard Set (ESS), as well as the highly discriminating SE33 locus, two Y-based loci and the sex determining maker, Amelogenin. The full complement of loci in the GlobalFiler™ Kit are: D13S317, D7S820, D5S818, CSF1PO, D1S1656, D12S391, D2S441, D10S1248, D18S51, FGA, D21S11, D8S1179, vWA, D16S539, TH01, D3S1358, AMEL, D2S1338, D19S433, DYS391, TPOX, D22S1045, SE33 and a Y-specific insertion/deletion locus (Yindel). The 6-dye GlobalFiler™ PCR Amplification kit (ThermoFisher Scientific) comprises 21 autosomal STRs have already been proven to be able to provide reliable DNA profiling results and enhance the power of discrimination between individuals. A population study of Caucasians residing in Maine was conducted using the AmpF1STR Profiler PCR Amplification Kit and the AmpF1STR Profiler Plus PCR Amplification Kit (Applied Biosystems Division (ABD) of Perkin Elmer, Foster City, CA). The kits contain the reagents necessary to amplify 12 different STR loci and the gender marker Amelogenin using two multiplex PCR, each containing nine STR loci. Thus, there is an overlap of six STR loci. The 12 STR loci are TH01, TPOX, CSF1PO, D3S1358, vWA, FGA, D8S1179, D21S11, D18S51, D5S818, D13S317, and D7S820. These loci represent 12 of the 13 core loci selected by the CODIS STR standardization project. Dye-labeled amplification products were separated and detected using the capillary electrophoresis instrument ABI Prism 310 Genetic Analyzer. Allele frequencies were determined for the 12 STR loci. Statistical analysis of the data included Hardy-Weinburg equilibrium (HWE) analysis, pairwise independence testing, power of discrimination (PD), and probability of exclusion (PE).  相似文献

目的评估SiFa~(TM) 23 Plex试剂盒(提取测试版)的性能及其所包含的STR基因座在汉族人群中的群体遗传学信息。方法应用试剂盒对1 000名汉族健康无关个体进行分型检测,检测试剂盒有效性并统计分析STR基因座的频率数据及群体遗传学参数信息。结果该试剂盒最小扩增体系可以为6.25μL,标准体系25μL中DNA含量低至125 pg可获取理想分型图谱。试剂盒中包含的21个常染色体STR基因座在1000人份中共检测到267个等位基因,均符合Hardy-Weinberg平衡。新增加的两个STR基因座D1S1656和D10S1248分别检测到15和11个等位基因,多态信息含量丰富。结论 SiFa~(TM) 23 Plex试剂盒(提取测试版)对于血液样本检测性能优越,准确性、重复性、灵敏度均可满足法医学检案需求,可以应用于法医学实际检案及DNA建库工作。  相似文献

The aim of this study was to estimate the allelic frequencies of the 15 STR loci included in the AmpFlSTR Identifiler PCR Amplification Kit in a sample of 342 unrelated Caucasian individuals autochthonous from Spain to be used for forensic purposes and population studies. The combined power of discrimination and the combined power of exclusion for all of the 15 loci were 5.68x10(-18) and 0.9999964, respectively. According to the obtained data, the D18S51 locus may be considered the most informative among the tested loci.  相似文献

目的研究21个常染色体STR基因座(CSF1PO,D3S1358,D5S818,D7S820,D8S1179,D13S317,D16S539,D18S51,D21S11,FGA,TH01,VWA,D2S1338,D19S433,D1S1656,D12S391,D2S441,D10S1248,TPOX,D22S1045,SE33)在新疆汉族人群中的遗传多态性。方法用GlobalFiler^TM R PCR Amplification荧光标记试剂盒对1066例新疆汉族无关个体的DNA进行PCR扩增,3500遗传分析仪电泳分析,用GeneMapper■ID-X v1.4软件分析等位基因片段大小,用Modified-Powerstates和Arlequin v3.5分析软件进行等位基因频率和法医学常用参数统计分析。结果在新疆汉族人群中,21个常染色体STR基因座不存在连锁不平衡现象,基因型分布符合Hardy–Weinberg平衡,共检出282个等位基因和1147种基因型,杂合度期望值(He)范围从0.6291(TPOX)到0.9428(SE33),多态信息含量(PIC)范围从0.5648(TPOX)到0.9393(SE33),累计个体识别率(CDP)>0.99999999999999999999。结论新疆汉族人群21个常染色体STR基因座具有较高多态性,可以用于法医学亲权鉴定和个体识别,也可以用于人类学和遗传学研究。  相似文献

目的调查19个常染色体STR基因座在贵州汉族人群中的等位基因分布,评估其在法医学中的应用价值。方法应用Goldeneye~(TM) DNA身份鉴定系统20A试剂盒,研究贵州520名汉族无关健康个体19个常染色体STR基因座多态性。用310型遗传分析仪进行毛细管电泳,Gene Mapper~ID v3.1进行基因分型。结果 19个常染色体STR基因座的杂合度为0.603 8~0.916 4,个体识别率为0.790 0~0.985 6,非父排除率为0.295 5~0.826 9,多态信息含量为0.553 5~0.908 9,累积个体识别率为1-1.230 0×10~(-22),累积非父排除率为0.999 999 99。贵州汉族和其他五个地域的汉族两两之间等位基因频率比较,仅贵州汉族与山东汉族、辽宁汉族、山西汉族之间存在基因频率差异具有统计学意义。结论 D19S433等19个常染色体STR基因座在贵州汉族人群中具有良好的遗传多态性,对群体遗传学和法医物证学研究有应用价值。  相似文献

中国汉族人群15个STR基因座的等位基因频率调查   总被引:14,自引:7,他引:14  
目的 调查10071名中国汉族无关个体15个STR基因座的等位基因的类型及其频率,并与以往相关文献报道的汉族群体资料进行统计比较。方法 应用PowerPlex~(TM)16荧光标记复合扩增系统,对10071份中国汉族无关个体的血样DNA进行15个STR基因座的复合扩增;用ABI 377或3100遗传分析仪对扩增产物进行分型,统计15个STR基因座的基因频率。结果 15个STR基因座共发现226个等位基因,频率在0.0001~0.5512;除D8S1179基因座外,其它基因座均发现稀有等位基因,数目1~7个不等,共34个。在中国汉族人群,稀有D21S11基因座的等位基因32.1和36.2,D18S51基因座的等位基因15.2和17.2,Penta E基因座的等位基因15.2、17.4、18.4、19.4、26和27,D7S820基因座的等位基因9.2、10.1、11.1和15,Penta D基因座的等位基因18、19和20,TPOX基因座的等位基因14,FGA基因座的等位基因13,以及较常见但欧洲稀有的D21S11基因座的等位基因30.3和D7S820基因座的等位基因9.1和9.2等均为首次报道。结论 大样本基因频率调查有利于观察STR基因座的稀有等位基因;本研究结果与以往相关文献报道的结果有不同程度的差异。  相似文献

目的 研究PentaD和PentaE基因座分型引物设计 ,调查PentaD和PentaE基因座在武汉汉族人群中的遗传多态性。方法 用重新设计的PentaD和PentaE基因座分型引物 ,采用热启动PCR和PAGE技术对 2 81名武汉地区汉族无关个体进行分型调查 ,并将其分型结果与Promega公司的PowerPlexTM16系统荧光标记复合扩增试剂盒分型结果进行比较。结果 新设计引物扩增产物的片段大小范围分别为 15 3~ 198bp和 10 7~ 2 12bp ,其分型结果与PowerPlexTM16系统的分型结果完全一致 ,且用银染法检测的灵敏度显著提高 (由 0 5ng提高到 0 2ng)。PentaD和PentaE基因座在武汉汉族群体分别检出 10个和 2 1个等位基因 ,其基因型频率分布均符合Hardy Weinberg平衡。家系调查证实了其等位基因的传递符合孟德尔遗传规律。两基因座的个体识别能力 (DP)分别为 0 92 62、 0 9860 ,非父排除率 (PE)分别为 0 665 1、 0 83 2 5。结论 新设计引物用于PentaD和PentaE基因座的分型检测准确可靠 ,两基因座多态性程度高 ,在法医学个人识别和亲子鉴定中具有使用价值。  相似文献
