Traditional methods for bone pulverization typically generate heat, risking stability of DNA sample. SPEX? has developed cryogenic grinders which introduce liquid nitrogen to cool the sample and aid in the grinding process. In this study, the Freezer Mill 6970 EFM was used with two DNA extraction methods and routine downstream STR analysis procedures. DNA from as little as 0.1 g of bone powder was used to develop full STR profiles after freezer mill pulverization, and the method was reproducible. Further, no contamination was detected upon cleaning/reuse of the sample vials. There were no significant differences in DNA yield, STR alleles detected, or peak heights using the freezer mill as compared to traditional grinding, and successful DNA profiles were achieved from as low as 0.1 g of bone powder with this method. Overall, this work indicates that this cryogenic mill method may be used as a viable alternative to traditional tissue grinders. 相似文献

This study examined individual and work-level factors that impact job stress and satisfaction for correctional officers. Existing research has explored officer job stress and satisfaction, but very few studies have focused specifically on fear of contracting an infectious disease while at work (HIV/AIDS, hepatitis, and tuberculosis), and the impact fear of and exposure to infectious disease have on correctional officer job stress and satisfaction. Random sample data were collected from 2,999 male and female officers from across the state of Texas to assess job stress, satisfaction, personal safety, and exposure to infectious disease. Ordinary Least Squares analyses indicated that fear of disease was positively correlated with job stress, and inversely correlated with job satisfaction. Exposure to disease however, failed to yield any significant effects on job stress or satisfaction. Officers who felt that their supervisors were supportive of them on the job reported less stress and higher satisfaction levels, while perceived dangerousness of the job was positively correlated with job stress. These findings highlight the importance of supervisory support as well as continuous, in-depth education and training on infectious diseases for officers. 相似文献

This longitudinal study of 134 female high school seniors investigated the relationship between attachment cognitions and school and work functioning two years later. School functioning was indexed by interviewer-rated strain, stressful events, academic performance, and self-reported attitudes toward college. Work functioning was indexed by interviewer-rated strain, stressful events, and self-reported attitudes toward work. Attachment cognitions were found to be correlated with school strain, number and stress of school-related events as well as self-reported satisfaction, stress, and problems meeting deadlines. Attachment cognitions were found to be correlated with work strain, work satisfaction, tendency to overcommit, and performance anxiety. Most of these relationships persisted even when initial symptomatology was accounted for. These findings lend support to the hypothesis, derived from attachment theory, that secure attachment cognitions provide a secure base that allows the late adolescent to negotiate the challenges of college and work successfully.This research was supported by a PHS National Research Service Award to the first author, and by a grant from the W. Abstract: This article presents the novel instrumentation of nanomanipulation coupled to nanospray ionization‐mass spectrometry, which is used to directly probe trace analytes found on individual fibers. The low detection limits and sample volumes associated with nanospray ionization‐mass spectrometry make it the ideal instrument to implement for trace analysis. Nanospray ionization‐mass spectrometry, coupled with the nanomanipulator, allows for the direct probing of trace particulates on fibers. The technique is demonstrated by dissolving an electrostatic particle of cocaine from a fiber, collecting the analyte solution in a nanospray tip, and transferring the tip directly to the mass spectrometer to complete analysis. The utility of this technique is evident through the minimal sample preparation and short analysis time. The use of nanomanipulation coupled to nanospray ionization‐mass spectrometry could improve on current trace particulate analysis by reducing both detection limits and sample size required to complete analysis. 相似文献

Prior research has established a strong and enduring “racial divide” in support for capital punishment, but little research
has explored the processes that explain the racial divide. Following the lead of Unnever and Cullen (Social Forces 85:1281–1301,
2007a), this research explores whether racist sentiment and core values (individualism, egalitarianism, symbolic patriotism,
and authoritarianism) can partially explain the racial divide in public support for capital punishment. The findings suggest
that racist sentiment by Whites and belief in core values by Whites partially explains the racial divide in support for capital

This study tested associations between adolescent perceptions of interparental conflict, adolescent attachment security with parents, and adolescent marital expectations and romantic experiences. Participants were 96 early adolescent females from 2 parent families. Insecurity was examined as a mediator of the association between negative perceptions of parental conflict and romantic outcomes. Results supported the mediation model in which adolescents' negative perceptions of parental conflict was associated with insecure attachment with parents, which was in turn associated with negative marital expectations and romantic experiences. Implications for understanding how parent-adolescent and interparental variables influence adolescent marital expectations and romantic experiences are discussed.

Pubertal Timing and Depressive Symptoms in Early Adolescents: The Roles of Romantic Competence and Romantic Experiences 总被引:1,自引:0,他引:1
In spite of the large literature supporting the link between early pubertal timing and depression in adolescent girls, there
are some exceptions. This suggests that there may be factors that interact with pubertal timing, increasing risk for depression
in some girls, but not others. This study examined two such factors, romantic competence and romantic experiences, and their
role in the cross-sectional and longitudinal associations between pubertal timing and depressive symptoms among 83 early adolescent
females (89% Caucasian). For on-time maturing girls (but not for early- or late-), lower levels of competence were associated
with higher levels of depressive symptoms concurrently, but not longitudinally. In addition, for on-time maturing girls, more
romantic experiences were associated with higher levels of depressive symptoms both concurrently and longitudinally. The discussion
focused on the need for greater conceptual and empirical clarity regarding the pubertal timing-depression association and
its potential moderators.
