The use of phosphate buffered saline for the recovery of cells and spermatozoa from swabs.

In the forensic science laboratory, the recovery of spermatozoa from vaginal swabs, or vaginal cells from penile swabs, can help determine if sexual intercourse may have taken place. There are several methods used to recover spermatozoa and cells from the swabs before visualisation on a microscope slide and most of these methods use water. Phosphate buffered saline (PBS) is a non-toxic solution used in many biological laboratories. Unlike water, PBS prevents cells rupturing or shrivelling up due to osmosis. This study demonstrates that PBS can be used for the extraction of spermatozoa and cells from swabs and that PBS does not affect subsequent DNA profiling.     

Love,sex and the spaces in-between: Kepri wives and their cross-border husbands

In the Riau Islands of Indonesia significant numbers of women have entered into marriages with men from the nearby countries of Singapore and Malaysia. In many cases, neither spouse migrates after marriage: instead, husband and wife continue to reside in their country of origin. Their close geographical proximity means that the couples can see each other regularly while at the same time taking advantage of the economic opportunities presented by living on different sides of the border. These cross-border marriages challenge the normative model of the nuclear cohabiting couple/family. Our research into the motivations and desires of these cross-border couples living in the Riau borderlands reveals that space and mobility mediate their interactions with the Singaporean, Malaysian and Indonesian states, thus producing localized accounts of citizenship in which class mobility (rather than physical mobility) becomes the dominant frame through which they view state regulation of marriage and migration. This research challenges the state-centric tendencies in some of the scholarly literature on international and transnational marriage which places overwhelming emphasis on the ability of states to regulate access to citizenship rights. In presenting a view of inconsistent and sometimes incoherent states, we highlight the significant differences between perceptions of state influence and actual state practices in relation to the regulation of international marriages.     

Systematic differences in electropherogram peak heights reported by different versions of the GeneScan software

DNA profiling using STRs on the 310 and 3100 Genetic Analyzers routinely generates electropherograms that are analyzed with the GeneScan software available from the instrument's manufacturer, Applied Biosystems. Users have been able to choose from three different smoothing options that have been known to result in significant differences in the peak heights that are reported. Improvements in the underlying algorithm of the most recent version of the software also result in significant and somewhat predictable differences in peak height values. Laboratories that have performed validation studies using older versions of GeneScan should either reanalyze the data generated in those validation studies with the newest version of the software or otherwise take into consideration the systematically higher peak height values obtained as they begin following the recommendation of the manufacturer and use the new algorithm.     

Governing concept or political buzzword? Contested perceptions of local control

This article uses a perceptions-based framework to study how local governing actors define local control, and, to identify the determinants of definition alignment among local officials. A mix of survey and administrative data from Wisconsin officials is used to test nine hypotheses. The results indicate there is wide variation in how local government leaders define local control, and that alignment on the concept's meaning is predicted by ideology, perceived erosion of local control, and positional characteristics. The study concludes that local governments should incorporate a working definition of local control in planning and on-boarding processes in order to provide governing relevance to the concept.
