Haplotype diversity in human mitochondrial DNA hypervariable regions I–III in the city of Caracas (Venezuela)

In order to expand the database of variable DNA for forensic identification purposes in Venezuela, we analyzed the sequence polymorphisms of mitochondrial DNA (mtDNA) hypervariable regions (HVR) I–III from 100 unrelated individuals from the city of Caracas, using PCR amplification and fluorescent-based capillary electrophoresis sequencing method. Dominant haplogroups corresponded to Native Americans followed by African ones. The inclusion of HVR III although useful for sub-haplogroup assignation, added little to the discrimination capacity of our database.     

At Boiling Point: Like Water for Chocolate and the Boundaries of Mexican Identity

Abstract — This article explores the mechanisms of Mexican identity as they are constructed in Alfonso Arau's film Como agua para chocolate (1991) (Like Water for Chocolate). In re-designing the characters of Laura Esquivel's novel, Arau produces a range of filmic stereotypes drawn from both the Hollywood and the Mexican traditions of film-making. Through the careful manipulation of filmic devices such as editing, framing and close-ups, many of the features of Mexican otherness perpetuated by Hollywood throughout the twentieth century are inscribed. I apply the metaphor of boiling, derived from the film's title, to examine certain key concepts of cinematic 'mexicanness' including the tropes of 'revolution', 'border', 'race' and 'sex'. Crucial to this argument is a consideration of the contemporary political climate in which Like Water for Chocolate was both produced and released. In Mexico, it was released halfway through the sexenio (six-year period of rule) of President Carlos Salinas de Gortari's rule and is clearly one of the most succesful cultural products (and exports) of his government's now infamous rule. In the United States, the climate of anti-immigrant attitudes in 1992 and 1993 and the corresponding political tension provokes new readings of certain stereotypical images of Mexicans and mexicanness. It is the tension that is produced by the collision between these two contexts — cultural, political and ethnic — that forms the principal focus of discussion in this article.     

A highly specific and sensitive sandwich enzyme immunoassay for human hemoglobin A

A highly specific and sensitive sandwich enzyme immunoassay for human hemoglobin A (Hb A) is described. A rabbit anti-human Hb A IgG-coated polystyrene ball was incubated with human Hb A and subsequently with affinity-purified anti-human Hb A Fab'-horseradish peroxidase conjugates, which had been prepared before and after absorption with Japanese monkey Hb-Sepharose 4B and dog Hb-Sepharose 4B. Bound peroxidase activity was measured by fluorimetry using 3-(4-hydroxyphenyl)propionic acid as a substrate. The detection limits of human Hb A using the conjugate before and after the absorption were 0.65 pg/tube (3 X 10(10)-fold dilution of whole blood) and 2.0 pg/tube (1 X 10(10)-fold dilution of whole blood), respectively. Human Hb A could be discriminated from Hb of animals such as Japanese monkey, dog, cat, pig, horse, sheep, chicken and cow by measuring bound peroxidase activity in the presence and absence of the conjugates prepared before and after the absorption. Human Hb A in bloodstains on cotton gauze could be discriminated from Hb of animals described above even after seven washings. Human Hb A in 220,000-fold diluted bloodstains on cotton gauze could also be discriminated from Hb of animals described above.     

Identification of propellants of small arms ammunition

Existing methods seem inadequate to identify propellants of small arms ammunition. The present article describes that viscosity may be effectively used to differentiate and identify those. The temperature dependence, intra- and inter-batch variation and storage effect of propellants were studied.

DNA recovery after sequential processing of latent fingerprints on copy paper

Forensic examiners must determine whether both latent fingerprint development and DNA profiling can be performed on the same area of an evidence item and, if only one is possible, which examination offers the best chance for identification. Latent fingerprints can be enhanced by targeting different components of fingerprint residues with sequential chemical treatments. This study investigated the effects of single-reagent and sequential latent fingerprint development processes on downstream DNA analysis to determine the point at which latent fingerprint development should be stopped to allow for DNA recovery. Latent fingerprints deposited on copy paper by one donor were developed using three sequential processes: 1,8-diazafluoren-9-one (DFO) → ninhydrin → physical developer (PD); 1,2-indanedione-zinc (IND-Zn) → ninhydrin → PD; and IND-Zn → ninhydrin → Oil Red O (ORO) → PD. Samples were examined after the addition of each chemical treatment. DNA was collected with cotton swabs, extracted, quantified, and amplified. DNA yields, peak heights, number of alleles obtained, and percentage of DNA profiles eligible for CODIS upload were examined. DNA profiles were obtained with varying degrees of success, depending on the number and type of treatments used for latent fingerprint development. The treatments that were found to be the least harmful to downstream DNA analysis were IND-Zn and IND-Zn/laser, and the most detrimental treatments were DFO, DFO/laser, and PD. In general, as the number of treatments increase, the opportunities for DNA loss or damage also increase, and it is preferable to use fewer treatments when developing latent fingerprints prior to downstream DNA processing.

Zoning in this country has traditionally been a waiting game with the development proceess. Not knowing what specific property will be used for and not controlling the ball game suficiently to say what will be the use, land has been zoned in a “wait and see” classification. Some land has been overzoned for certain uses; other land has been left fairly open as to its potential use. Thus the system itself necessitates constant change in a property's classification as circumstances change. Many of these changes are made to accommodate a specific development proposal, so they are to the landowner's benefit. However, other rezonings result in a reduction in the landowner's potential for profit. It is this type of change—downzoning—that is the subject of this discussion.     

Marriage,residence, and occupational choices of senior and junior siblings in the english past

The aim of this study is to throw light on the importance of social networks by examining the choice of godparents and witnesses to marriage in early nineteenth-century Iceland. The article analyzes parish records concerning two communities representative of different socio-economic environments in southwestern Iceland. The results show that godparents were chosen predominantly among people of standing in the neighborhood, such as communal directors, church wardens, merchants, and midwives. Grandparents figured only rarely as godparents. In the case of marriage, however, close kin played a markedly more important role as witnesses, particularly in the upland community that had a more stable population than the coastal one. Observed local variations in this respect can be explained in light of different socio-economic settings.
