There are no efficient methods to determine the geographic origin of unidentified cadavers. We showed earlier that the geographical distribution of the JC virus genotype detected from human kidneys indicates the host's geographical origin. As there are still cadavers from which we cannot detect the JC virus (JCV), we investigated whether the genotype of another virus species belonging to the same family, human BK virus (BKV), could also be used to detect human geographical origin. BKV was found in 11 of 36 cases (30.5%). Even in the seven JCV-negative cases, the host's geographic origin could be estimated from the BKV genotype. Four subjects were positive for both the BKV and JCV. As the distribution areas of BKV and JCV genotypes are not identical, it is possible to narrow down the geographic area that any cadaver originates from.  相似文献

Abstract: In this study, the specificity, sensitivity, and operability of RSID?‐Urine, a new immunochromatographic test for urine identification, was evaluated and compared with ELISA detection of Tamm‐Horsfall protein (THP). Urine was successfully identified among other body fluids using RSID?‐Urine and ELISA detection of THP. The detection limit of RSID?‐Urine equated to 0.5 μL of urine; although the sensitivity of RSID?‐Urine may be lower than that of ELISA detection of THP, it is thought to be sufficient for application to casework samples. However, results from RSID?‐Urine must be interpreted with caution when the sample may have been contaminated with blood or vaginal fluid, because this might inhibit urine detection. The RSID?‐Urine assay can be performed in just 15 min by dropping the extracted sample onto the test cassette. Therefore, RSID?‐Urine should be an effective tool for the forensic identification of urine, in addition to ELISA detection of THP.  相似文献

We evaluated the performance of real-time RT-PCR and ELISA assays for detection of dermcidin (DCD) in sweat and body-fluid stains. DCD, a small antibiotic peptide secreted into human sweat, was detected by real-time RT-PCR in 7-day-old stains containing as small as 10 μL of sweat, and the assay showed high specificity when testing 7-day-old stains containing 30 μL of other body-fluid. ELISA using anti-human dermcidin mouse monoclonal antibody detected DCD sweat diluted up to approximately 10,000-fold and could specifically detect DCD in 10 μL of body-fluid stains. The performance of the two assays was tested during winter on samples that simulated forensic case samples: an undershirt and a sock worn for 20 h, a handkerchief used to wipe the brow several times within 12 h, a cap and a cotton glove worn for 4 h, and a white robe worn at intervals for 2 years. The result showed that the former assay detected DCD in all sites of the undershirt examined (armpit, back, and breast), and the latter gave a relatively high OD value in the armpit among the three sites. For the socks, although the latter assay gave very high OD values in both the center and toe of the foot sole, the former could not detect DCD in both of them. These results indicate that highly damp conditions, such as inside a shoe, might promote the degradation of mRNA in samples such as socks. In the other case samples, sweat was adequately detected by both assays.This study is the first demonstration of the use of real-time RT-PCR to sensitively identify sweat among body-fluid stains, and it confirmed that dermcidin was an excellent marker for sweat identification. In addition, the usefulness of ELISA was also verified. Positive sweat identification using these assays is expected to assist forensic practice.  相似文献

ABO types obtained from evidentiary samples have been used effectively to obtain the initial information leading to the apprehension of culprits in Japanese criminal investigations. A simple ABO genotyping method using multiplex sequence-specific PCR and capillary electrophoresis was developed as a supplement to serological ABO typing. Limitations in predicting a phenotype based on genotype were evaluated using 1134 randomly selected Japanese peripheral blood samples. A concordance rate of 99.82% (1132/1134 samples) was found between genotypes and phenotypes defined as Groups A, B, AB, and O. Sequencing analysis revealed that one discrepant sample contained an O allele having a previously unreported point mutation at the primer binding site in exon 6, and another discrepant sample contained an O allele lacking the guanine deletion at nt 261 (the O301 allele). Therefore, the existence of such alleles must be given some consideration when predicting phenotype based on genotype.  相似文献

In a Japanese study of glue-sniffers, urinary ortho-cresol and expired air toluene concentrations were determined by gas chromatography, and urinary hippuric acid concentrations by high performance liquid chromatography. There were significant differences among three sample groups (30 male "glue-sniffers", 13 "non-glue-sniffers" and 32 industrial workers) in their expired air toluene, urinary hippuric acid and ortho-cresol concentrations. Moreover, there was a better correlation between expired air toluene and urinary ortho-cresol concentrations than between toluene and urinary ortho-cresol concentrations than between toluene and hippuric acid. These results suggest that urinary ortho-cresol is a good indicator of toluene inhalation in glue-sniffers.  相似文献

ABO genotyping is often performed to identify the blood type of decomposed samples, which is difficult to be determined by a serological test. In this study, we developed a simple method for ABO genotyping using a DNA chip. In this method, polymerase chain reaction-amplified and fluorescent-labeled fragments in the ABO gene and primate-specific D17Z1 were hybridized with DNA probes on a chip designed to detect single nucleotide polymorphisms (SNPs) in the ABO gene and part of the D17Z1 sequence. Using blood samples from 42 volunteers and 10 animal species, we investigated whether the chip could be used to detect SNPs in the ABO gene and the D17Z1 sequence. This method was then applied to various forensic samples, and it was confirmed that this method was suitable for the simultaneous analyses of ABO genotyping and species identification. This method fulfills the recent need for the development of rapid and convenient methods for criminal investigations.  相似文献

Analysis of fatal injuries to motorcyclists by helmet type   总被引:1,自引:0,他引:1  
To clarify the characteristics of injuries of motorcyclists dying in accidents in relation to helmet type, we retrospectively analyzed forensic autopsies of 36 helmeted motorcycle riders. The presence of major injuries and injury severity were evaluated with the injury severity score and the 1990 revision of the Abbreviated Injury Scale. Persons with open-face helmets (19 cases) were significantly more likely to have sustained severe head and neck injuries, especially brain contusions, than were persons with full-face helmets (17 cases). Furthermore, major injuries of the chest or abdomen, rib fractures, lung injuries, and liver injuries were each present in more than one quarter of all cases (26.3% to 70.6%), but their prevalences did not differ significantly between riders with different types of helmet. Because many types of head and neck injuries cannot be prevented and fatal chest and abdominal injuries occur despite the use of full-face helmets, more effective helmets and devices for protecting the chest and abdomen are needed to decrease deaths from motorcycle accidents.  相似文献

We report 10 autopsy cases involving fatal pathological changes in abdominal organs, for which findings of computed tomography (CT) on admission or after death were compared with autopsy findings. Two of the cases were death due to natural causes and eight were death due to traffic accidents. From the findings at autopsy, the causes of death were considered to be rupture of an aortic aneurysm in one case, gastrointestinal bleeding due to gastric cancer in one case, retroperitoneal bleeding in two cases, laceration of the liver in three cases, and traumatic rupture of the small intestine in three cases. CT findings revealed ascites or retroperitoneal bleeding in eight cases. However, in the cases of small-intestinal rupture, CT findings on admission revealed no free air. Therefore, ascites on CT should be regarded as a useful indicator of blunt abdominal trauma. Hepatic portal venous gas, known to be a post-mortem change, was significantly evident on post-mortem CT in cases of traumatic liver or intestinal injury, and was also seen in cases where the period between the accident and CT examination was long.  相似文献

Tobacco policy in the UK and Japan has diverged markedly. In the 1980s, both countries oversaw regimes with minimal economic and regulatory policies. Now the UK has become one of the most, and Japan one of the least, controlled (advanced industrial) states. These developments are puzzling to public health scholars who give primary explanatory weight to scientific evidence and a vague notion of “political will”, because policy makers possessed the same evidence on the harms of tobacco, and made the same international commitment to comprehensive tobacco control. Instead, we identify the role of a mutually reinforcing dynamic in policy environments, facilitating policy change in the UK but not Japan: policy makers accepted the scientific evidence, framed tobacco as a public health epidemic, placed health departments at the heart of policy, formed networks with public health groups and excluded tobacco companies, and accentuated socio-economic conditions supportive of tobacco control. This dynamic helps explain why the UK became more likely to select each tobacco policy control instrument during a series of “windows of opportunity”. Such analysis, generated by policy theory, is crucial to contemporary science/practitioner debates on the politics of “evidence-based policy making”: the evidence does not speak for itself, and practitioners need to know how to use it effectively in policy environments.  相似文献
