旋毛虫组织蛋白酶B基因TsCB1的克隆与生物信息学分析

为了研究旋毛虫组织蛋白酶B的功能,对GenBank中旋毛虫基因组数据库和EST数据库进行检索,获得旋毛虫组织蛋白酶B的cDNA序列并设计引物。以旋毛虫肌幼虫总RNA为模板,进行RT-PCR,将PCR产物克隆到pMD18-T载体后测序并进行生物信息学分析。结果表明,成功克隆到旋毛虫组织蛋白酶B基因(TsCB1)的cDNA序列,TsCB1cDNA含有1个由1 449个核苷酸组成的完整的开放阅读框,编码482个氨基酸残基组成的多肽,蛋白质分子质量理论值为55.1ku,理论等电点为7.66。TsCB1第1～35位氨基酸残基为信号肽序列,有2个潜在的N-糖基化位点,具有1个生长调节素B结构域和半胱氨酸蛋白酶结构域,半胱氨酸残基活性位点被丝氨酸残基所替换,同源性分析表明与其他线虫组织蛋白酶B的一致性在60%左右。     

Feudalism in the Service of the Revolution

This article discusses the way the past is being reexamined in modern-day Vietnam, particularly through the medium of heritage. Hue, the old royal capital of Vietnam, provides the case study, as this city reflects the great themes and events of Vietnamese history over the last two hundred years, from the establishment of a unified nation under the Nguyen, through the imposition of colonial control, the devastation of war, reunification, and the establishment of communism, to the consolidation of an independent postcolonial nation. The importance of Hue's heritage is recognized in its status as a “world heritage” site. The author argues that Hue's heritage is, nevertheless, problematic for Vietnam's ruling communists, because to them it largely represents a regime—the Nguyen Dynasty—that was “reactionary” and that had sold out the country to the French. The apparent contradiction between the standard communist view of the Nguyen past and the value accorded to Nguyen heritage in Hue is resolved, the author contends, by recourse to the depoliticized practices of heritage preservation and tourist promotion.

Abstract

American social scientists have recently expressed deep concern over the stare of “human rights” in the third world The source of this concern can be pinpointed precisely as the “new moralism” of the Carter Administration. Like Mr Carter, these social scientists address the problem by isolating human rights as a distinct phenomenon which can be dealt with in the abstract, detached from the root characteristics of the political economy within which it occurs. The typical analysis focuses on the individual dictator, who, ostensibly, has available to him a range of choices on the amount of social control that is to be maintained. For example, if the United States were to exert pressure and to discontinue aid, then, according to this view, the dictator would restore democratic rights.     

房屋征收补偿协议的性质与救济——对《国有土地上房屋征收与补偿条例》第25条的分析

因《国有土地上房屋征收与补偿条例》的相关规定不明确,导致实践中对补偿协议的性质和救济的认识不同,进而产生了救济上的混乱和困境.究其本质,补偿协议作为一种新型行政合同,属于行政法律关系的范畴,但又具有一定的民事合同形式特征.本文认为,从该条例的立法本意、诉讼便利以及控制行政权、保障公民权的行政法法理出发,应通过行政诉讼渠道解决相关争议,而目前之问题在于需要行政诉讼法的修改.     

The mental health association and prevention

Summary

The history of the Mental Health Association's involvement in prevention is described. From its inception in 1909 the Association has played a key role in advocating for prevention. It contributed to the Child Guidance Center movement, the community mental health center movement, and helped focus the 1978 President's Commission on Mental Health on prevention. In recent years, it has served as the catalyst for the formation of a coalition of national organizations with an interest in prevention.     

Strategic Disorder,the Office of Policy Coordination and the Inauguration of US Political Warfare against the Soviet Bloc, 1948–50

This article examines the inauguration of US political warfare operations against the Soviet bloc, 1948–50. It builds on diplomatic and military historiography of US Cold War foreign policy, and engages with more recent scholarship of the ‘covert’ and unconventional confrontation between east and west. Two key interrelated themes are explored. The failure of Truman's policymakers, particularly the State Department's Policy Planning Staff, to devise a coherent strategy to wage political warfare against communist regimes was fundamental to its ultimate failure. Also, the disordered US government bureaucracy further impaired and strategically dislocated the application and overall success of political warfare operations by the Office of Policy Coordination.

Validation and implementation of the PowerPlex 16 BIO System STR multiplex for forensic casework

The PowerPlex 16 BIO multiplex short tandem repeat (STR) system contains the 13 CODIS loci (FGA, TPOX, D8S1179, vWA, D18S51, D21S11, TH01, D3S1358, CSF1PO, D16S539, D7S820, D13S317, and DS5S818), plus two pentanucleotide repeat loci (Penta D and Penta E) and the sex-identifying locus. Amelogenin. The PowerPlex 16 BIO System is optimized for use with the Hitachi FMBIO gel imaging systems. A consortium of seven independent laboratories collaborated to perform the studies defined by the FBI standards for performing a developmental validation, including the evaluation of sample concordance, percent stutter determination, nonprobative casework, precision, sensitivity, mixture determination, effect of substrates, the impact of environmental insults, and species specificity. All samples tested for concordance were consistent except for one sample from the Virginia Division of Forensic Science database that displayed discordance at D13S317, a locus whose primer sequence was altered. Stutter values were comparable to those of other STR multiplex systems, the precision was comparable to other multiplexes analyzed by gel electrophoresis, the DNA profiles were unchanged by the substrate upon which the blood samples were placed, and the nonprobative casework samples re-typed for the PowerPlex 16 BIO System were consistent with previous typing results. When greater than 0.125 ng of DNA was placed into the PowerPlex 16 BIO System amplification reaction, a full profile was generated by all laboratories. The mixture study results were comparable to those reported for other multiplex systems, the environmental study demonstrated a loss of larger molecular weight loci when samples were incubated at elevated temperatures for a prolonged period of time, and the only notable cross species hybridization was observed with primate DNA samples. This extensive validation work performed demonstrates that the PowerPlex 16 BIO System provides STR data of a quality comparable with other PowerPlex STR multiplex kits as well as other widely used STR multiplexes and is thus suitable for evidentiary casework analysis as well as database sample profiling.
