Quantitation using GC-TOF-MS: example of bromazepam

Time-of-flight mass spectrometry (TOF-MS) offers new perspectives for forensic toxicology. Qualitative and quantitative analyses of a mixture of three selected benzodiazepines (diazepam, nordazepam and bromazepam) were used to compare gas chromatography (GC-TOF-MS, quadrupole GC-MS, GC-ECD) and liquid chromatography (HPLC-DAD) data. Method validation parameters like LOD, LOQ, S/N-ratios reflect the capabilities of GC-TOF-MS. Five-point calibrations for bromazepam in human peripheral blood (50, 100, 160, 200, 300 ng/ml) using medazepam as internal standard (1000 ng/ml) were performed. The calibrations using GC-TOF-MS (using the fragments of m/z 236 and 288), GC-ECD (dual system) and HPLC-DAD (at 235 nm) all showed correlation coefficients close or superior to 0.99. Quadrupole GC-MS data was not used in the comparison of extracted samples due to the low sensitivity in the full scan mode. Two analyses of real cases concerning bromazepam are presented. In the first case, the presence or absence of bromazepam could not be established with both HPLC-DAD and GC-ECD due to background signals. The extracted ion chromatograms and spectrum traces after the analysis with the GC-TOF-MS could clearly excluded the presence of bromazepam. The second case illustrates the quantitation of bromazepam, where both HPLC-DAD and GC-ECD were unable to give satisfactory results, again due to interfering background signals. The analyses performed on the GC-TOF-MS-system demonstrated high sensitivity and also high selectivity due to the high quality of mass spectra obtained. The advantages of GC-TOF-MS make it a promising analytical technique for forensic toxicology.     

Dimensions of self-esteem and psychological symptoms among chinese adolescents in Hong Kong

Self-esteem and psychological symptoms were assessed in a sample of 1082 Chinese adolescents who responded to the Chinese versions of Coopersmith's Self-Esteem Inventory and the General Health Questionnaire. Three dimensions of self-esteem, interpreted as Family Acceptance/Rejection, Self-Depreciation, and Peer Affirmation/Disaffirmation, were found to relate substantially to general psychological symptoms and particularly to specific symptoms of anxiety, social dysfunction, and depression as reported by these adolescents. The implications of the relative importance of appraisal from internal feedback as sources of self-esteem as opposed to the external feedback from family and peers were discussed in relation to self-esteem enhancement in adolescents.Received his Ph.D. from the University of Western Ontario in 1978. Paint and tape: collection and storage of microtraces of paint in adhesive tape

The collection and preservation of microtrace evidence with the aid of an adhesive tape is a method of choice in forensic science. This technique is rapid and easy and allows the concentration of microtraces on a carrier, which facilitates further investigations in the laboratory. Adhesive tapes are currently used to secure microtraces of fibers and glass, but hardly for traces of automotive paint and other lacquers for fear of interference with the analysis of binders. A collection of automotive paint consisting of original and repair lacquers collected by tape has been evaluated. After various times of storage within the tape, these samples were compared with untreated references by microscope FT-IR and microspectrophotometry (MSP). Another set of paints was collected in 1984, stored within the tape until 1995, and examined the same way. About 170 layers of lacquer with various types of binder were examined. With the exception of one clear lacquer no difference between treated samples and references was detected. This small difference observed could be correlated to the exposure to xylene (extractant) and was not caused by the storage within the adhesive tape.     

Autoerotic accident by inhalation of propane-butane gas mixture

We present a case of an accidental autoerotic death involving the inhalation of a propane-butane gas mixture, also known as LPG (liquefied petroleum gas). A 19-year-old male was found dead in supine position in his bed in a residential accommodation one day after he was last seen alive. On a personal computer at the end of the bed, a pornographic movie was still running. On his left shoulder, an empty rubber balloon and on the bedside 2 empty "Kisag-Gas" cartridges were found. Toxicologic investigations revealed an intoxication with propane and butane, together with a recent consumption of cannabis. This case report compares the toxicologic findings with other recently published cases, and the theories of the toxic effects are discussed.     

Hochwasserschutz im Spannungsverhältnis zum übrigen Fachplanungsrecht, Raumordnungsrecht und zur Bauleitplanung

Durch das Oderhochwasser, aber vor allem durch die Flutkatastrophe an der Elbe im August 2002, ist der Hochwasserschutz verstärkt in das öffentliche Bewusstsein getreten. Dies hat auf der Ebene von Bund und Ländern zu verschiedenen Aktionen geführt, die sich nunmehr in Gesetzesänderungen niederschlagen sollen 1. Der Hochwasserschutz kann dabei durchaus in ein Spannungsverhältnis zu anderen öffentlichen und privaten Anliegen und Belangen treten—vor allem, wenn sich aus deren Sicht Einschränkungen für die bauliche oder sonstige Nutzung der betroffenen Flächen ergeben können. Ein wichtiger Teilausschnitt aus diesem Gesamtproblem ist das Spannungsverhältnis des Hochwasserschutzes zum übrigen Fachplanungsrecht, zum Raumordnungsrecht und zur Bauleitplanung. Hierüber soll auf der Grundlage des vorliegenden Gesetzentwurfs der Bundesregierung zum verbesserten Hochwasserschutz berichtet werden. *) Der Beitrag beruht auf einem Vortrag, den der Verfasser auf dem 9. Leipziger Umweltrechts-Symposium Rechtliche Aspekte des vorbeugenden Hochwasserschutzes am 22. 4. 2004 gehalten hat und der auch in dem von dem Direktoren des Instituts für Umwelt- und Planungsrecht, Prof. Dr. Martin Oldiges und Prof. Dr. Wolfgang Köck betreuten Tagungsband erscheinen wird.     

Toward a contemporary tradition for menarche

A cultural tradition to acknowledge a girl's first menstrual period is proposed in order to overcome the negative connotation of the event. Three basic questions regarding such a tradition are addressed: Who would participate? What would it mean? What type of activity should it be? A research team consisting of a psychologist, a nurse, and a social worker suggests answers based on their interviews with mothers and daughters, as well as their collection of menarche anecdotes from women psychologists.

The relevance of the detection of troponins to the forensic diagnosis of cardiac contusion

The forensic diagnosis of cardiac contusion has hitherto been based mainly on anamnesis, concomitant thoracic injuries and the detection of macroscopic changes to the heart. Parallel histological and serological investigations of the heart-specific troponins have been conducted with varying results. This paper aims to show whether heart-specific troponins are suitable as a means of securing the diagnosis in proven cases of cardiac contusion and of determining which of the three heart-specific troponins cTnT, cTnI and cTnC are most significant in serology and histology for postmortem diagnosis.In the study, 25 cases of known cardiac contusion and 11 controls without vital myocardial trauma taken from autopsy material were prospectively investigated. Investigation of the venous serum revealed significant differences in the concentrations of the case and control groups for troponin T (mean value 5.5056 versus 0.4982; p = 0.014), for troponin C (mean value 263.9280 versus 68.5640; p = 0.001) and for troponin I (mean value 1404.0560 versus 36.1650; p = 0.003). In histology there are also significantly different depletions between the groups investigated (cTnT: p = 0.002; cTnC: p = 0.003; cTnI: p < 0.001) taking into account the autolysis time.
