参照GenBank中盖他病毒株E2蛋白氨基酸序列并进行密码子优化后,人工合成该基因,并将其定向克隆到表达载体pGEX-4T-1中,经酶切及测序鉴定均正确后,转化宿主菌BL21(DE3)进行IPTG诱导表达。以该表达蛋白作为诊断抗原,建立了检测猪盖他病毒抗体的E2-ELISA方法,并对临床血清样品进行检测。结果显示,重组表达的E2蛋白具有良好的抗原性和特异性。用建立的间接ELISA检测471份临床血清样品,检出抗体阳性119份,阳性率为25.27%。结果表明,本研究制备的重组E2蛋白具有良好的免疫原性,可应用于ELISA等相关检测方法的建立。 相似文献

China's Internet companies and citizens are now world leaders in developing and using the Internet and related information technologies for financial transactions. Accordingly, it is important that China becomes a world leader in identifying challenges posed by Internet finance, and providing law and governance solutions to address these challenges. While the Internet and its associated technologies are now globally available, a core question is whether, and to what extent, regulatory challenges and opportunities are common across different jurisdictions, or whether they reflect local circumstances. In short, an interesting question is what can the world learn from China as it takes the lead in addressing Internet finance challenges, and what can China learn from the world as it seeks to do so?This article first identifies the landscape of China's burgeoning Internet finance market, including key technologies and services and government and nongovernment players. The article then turns to key regulatory challenges, with a focus on factors especially significant in China. The article then examines the “top down” “campaign style” approach to regulation, which is China government's initial response to emerging challenges. Following an analysis of the campaign, some suggestions are then made for future possible governance strategies. We explain how emerging “information” based and experiment-based approaches to governance are drawing on both global and Chinese experiences to harness the capabilities of the Internet and the collective energies of Internet finance enterprises and users to advance the regulation of the China Internet finance system in a way that is conducive to the public interest. 相似文献

Pratt and Turanovic (European Journal of Criminology, 13(1):129–146, 2016) argue that previous studies operationalizing risky lifestyles as mere “going out” (problematic indicators of risky lifestyles) were misspecified and that “improved” indicators of risky lifestyle (risky behaviors) would perform better than “problematic” indicators in models that explain victimization. This study examines these propositions by testing the self-control/lifestyle framework of victimization using the data from a random sample of Filipino high school students at a state university in Dumaguete City, Philippines. Results show strong support to Pratt and Turanovic’s claims. Self-control has stronger effects on improved indicators than on problematic ones. And, improved indicators have stronger effects than problematic indicators on property, violent, peer/sibling and sexual victimization. Moreover, the findings provide partial support for the self-control/lifestyle framework of victimization. 相似文献

This paper suggests that a gap exists between policy theorists who are formulating postpositivist models of analysis and the beliefs of practitioners whose labor is guided by the traditional hand of positivism. The tension between postpositivist theory and traditional practice has created a dialectic that contributes to both, but the postpositivist theory is not likely, for reasons discussed in the paper, to spark a revolution in practice. The paper maintains that members of the discipline's intellectual infrastructure can assist the evolution to a policy analysis paradigm that includes elements of postpositivism by introducing incremental changes in practice that reinforce it. For this purpose, one important incremental change is the introduction of Q‐methodology as a common tool for policy analysis. The use of Q‐methodology—a method for the study of subjectivity—would help subvert the assumptions of dominant objectivism that underlie the R‐methods typically learned and used by traditional analysts and could influence analysts to adopt a postpositivist perspective of their work. © 1999 by the Association for Public Policy Analysis and Management. 相似文献

Based on the concept model of vitae system, the fund invested into a disaster affected region is divided into three parts: Circumstance factor, economy factor and community factor. By using AHP (analytical hierarchy process), the relative weights of those three parts in vitae system are given. When the region is suffering from multiple low-frequency and high-impact disasters, the fund should be optimally managed to ensure the vitae index is larger than the critical value to achieve the region harmony. 相似文献
