胸膜肺炎放线杆菌体内表达基因的筛选

利用体内诱导抗原技术对胸膜肺炎放线杆菌(APP)体内表达的基因进行了初步筛选.将收集到的5份感染了APP1型的猪血清混合,分别用体外培养的APP1和大肠杆菌BL21(DE3)的全菌及全菌裂解物进行吸附,用ELISA方法检测吸附效果,经吸附后血清D450nm分别由原来的0.927和0.239降低为0.196和0.078.同时,将APP1基因组DNA以Bsp143 Ⅰ进行酶切,回收500～2000 bp的片段,与pET30a/b/c载体连接,构建了APP1基因组质粒表达文库,库容量(CFU)为2.0×105.用吸附后的血清通过菌落原位免疫杂交筛选该基因组表达文库,从3 000个菌落中获得了12个阳性克隆.     

理性·激情:五四"反传统"的再思考

五四"反传统"的提法并非对五四运动的全面概括,而是其中一种诠释话语,在20世纪80年代末从国外和港台传入大陆后,学术界产生巨大反响和争论.五四知识分子以"西方"为参照,通过对中国道统的否定、学统的解构和文统的革命,来达到思想启蒙、实现人的现代化的目的,其实质并非"全盘反传统主义".五四"反传统"为旧传统向新传统的转化开启了门径、为旧儒家向新儒家的转化提供了契机,至于后来出现的包括"文革"在内的一些过激事件,虽然不能说与"五四"毫无关联,但与"五四"本身并无必然的因果关系.对待"传统"的正确路径应当是遵循"传统"的特性和规律,形成对本民族传统的自觉意识,进而处理好传统与现代化的关系.

国家经济安全预警指标体系的构建

国家经济安全预警指标的选择原则一般应包括完备性原则、主要性原则、可测性原则和独立性原则。在此基础上确定的预警指标体系可以分为财金安全预警指标、社会安全预警指标、外经安全预警指标、资源安全预警指标和产业安全预警指标等。经济安全预警指标体系,应该是一个分层的树状结构。国家经济安全态势的恶化,往往是一个或少数几个风险因素演化和交互作用的结果,而通过整个指标体系层层加权综合得出的指数往往会弱化某些风险因素的影响,导致经济整体安全评价值的不准确。而基于案例的评价方法的推广应用,有赖于案例数据库质量和评价技术的可靠性,这两者将是今后的重要研究方向。     

检察理论研究之反思

检察理论研究在取得一定成果的同时,也存在着不少问题,如较强的部门化倾向、意识形态化话语较多、研究范式以证成性为主等。表面繁荣的背后暴露出学术评价量化机制的某些弊端。应以开放的姿态建构独立自主的检察理论研究体系,把握好历史与逻辑的辩证统一。     

The Aesthetic Account of Everyday Life in Organizations: A Report on Recent Developments in Organizational Research

This article examines recent advances in organizational studies that disputed the dominance of conventional-positivist paradigms by addressing everyday life in organizations through the lens of aesthetic principles. First, I discuss developments in theory and methods, focusing on the “strong” version of Organizational Aesthetics that defends new conceptions about the nature of organizations and experiences of organizational life. Then, I illustrate this new account using two relevant studies. My research finds that the aesthetic account of organizations provides concepts and modes of analysis able to better respond to current conceptual and practical challenges both in organizational research and arts management.     

遗传标记微单倍型在法医学中的研究进展

微单倍型作为一种新型的法医学遗传标记,在国际法医学界已经引起了越来越多的关注。微单倍型是在较短片段内(例如200bp),包含2个或以上个SNP,具有单倍型多态性的序列。相较于STR,微单倍型突变率低,在混合斑鉴定中具有一定优势;与SNP相比较,微单倍型的多态性更高。选择含有祖先信息特征的微单倍型,在种群分析鉴定中具有应用价值。本文就微单倍型的演变,分型方法,命名及群体特征等方面作一综述。     

The Value of Outsourcing Selected Cases in a Medical Examiner Population: A 10‐Year Experience

Due to increasing caseloads and inadequate staffing, the burden on Coroner/Medical Examiner Offices to comply with recommended autopsy limits for forensic pathologists (FPs) has been difficult. Since 2006, pathologists at the University of Alabama at Birmingham have performed select autopsies for the Alabama Department of Forensic Sciences. Each case was reviewed by a state FP and scene investigator to determine appropriateness for referral. All referred cases received full postmortem examination including microscopic examination and collection of toxicological samples, and toxicology was ordered by the referring FP as appropriate. The final cause and manner of death were determined by the referring state FP after review of all findings. A majority of the 421 cases were ruled accidental deaths (233), most due to drug toxicity. Of the 178 natural deaths, 118 were attributed to cardiovascular disease. Outsourcing select forensic cases can be educational and an effective tool to manage workflow without compromising quality.     

Enhancing student performance and employability through the use of authentic assessment techniques in extra and co-curricular activities (ECCAs)

The paper advocates the use of authentic assessment techniques, delivered in extra and co-curricular activities (ECCAs), to augment and improve student performance on academic law degree programmes. A combination of formative and summative methods in ECCA assessment provides the optimum environment to measure and improve the crucial critical reasoning skill – the key transferable skill to academic degree success. This skill, when developed in students, is also highly prized by prospective employers, and therefore, the use of authentic assessment improves graduate employability rates.
