Analysis of sensitivity and specificity: precise recognition of neutrophils during regeneration of contused skeletal muscle in rats

In this report, we applied the TissueFAXS 200 digital pathological analysis system to rapidly and accurately identify neutrophils during regeneration of contused skeletal muscle, and to provide information for follow-up studies on neutrophils to estimate wound age. Rat injury model was established, and skeletal muscle samples were obtained from the control group and contusion groups at 1, 1.5, 2, 3, 4, and 6 h, as well as at 1, 3, 5, and 15 d post-injury (n = 5 per group). The expression of nuclei and neutrophils was detected by hematoxylin and eosin (HE) staining and immunohistochemical (IHC) staining. A total of 20 injury site areas of 0.25 mm² (0.5 mm × 0.5 mm) were then randomly selected at all time points. A TissueFAXS 200 digital pathological analysis system was used to identify the positive and negative numbers. Knowledge of five professional medical workers were considered the gold standard to measure the false positive rate (FPR), false negative rate (FNR), sensitivity, specificity, and area under the curve (AUC) of receiver operating characteristic (ROC) curves. As a result, with a staining area of neutrophils from 8 µm² to 15 µm², the FPR was 4.28%–12.14%, the FNR was 12.42%–64.08%, the sensitivity was 35.92%–87.58%, the specificity was 87.86%–95.72%, the Youden index was 0.316–0.754, the accuracy was 82.80%–88.30%, and the AUC was 0.771–0.826. The AUC was largest when the cut-off value of the staining area was 12 µm². Our results show that this software-based method is more accurate than the human eye in evaluating neutrophil infiltration. Based on the sensitivity and specificity, neutrophils can be accurately identified during regeneration of contused skeletal muscle. The TissueFAXS 200 digital pathological analysis system can also be used to optimize conditions for different cell types under various injury conditions to determine the optimal cut-off value of the staining area and provide optimal conditions for further study. Furthermore, it will provide evidence for forensic pathology cases.

Fatal motorcycle accidents and alcohol

A series of fatal motorcycle accidents from a 7-year period (1977-1983) has been analyzed. Of the fatalities 30 were operators of the motorcycle, 11 pillion passengers and 8 counterparts. Of 41 operators 37% were sober at the time of accident, 66% had measurable blood alcohol concentration (BAC); 59% above 0.08%. In all cases where a pillion passenger was killed, the operator of the motorcycle had a BAC greater than 0.08%. Of the killed counterparts 2 were non-intoxicated, 2 had a BAC greater than 0.08%, and 4 were not tested. The results advocate that the law should restrict alcohol consumption by pillion passengers as well as by the motorcycle operator. Suggestions made to extend the data base needed for developing appropriate alcohol countermeasures by collecting sociodemographic data on drivers killed or seriously injured should be supported.     

A simplified procedure for simultaneous detection of salivary proteins and its application in paternity testing

An alternative simplified procedure for the genetic analysis of salivary protein complex (SPC) which eliminates the need for a specific concentrated parotid sample is proposed. After the technique is applied isoelectric focusing and protein detection is carried out by silver staining. The chance of exclusion of non-fathers for these systems in our population is 0.161711 for Pr, 0.07948 for Db, 0.07836 for Pa and 0.00995 for PIF. The total exclusion rate for these systems is 0.30042.

浅析危险化学品安全储存管理

随着我国经济的发展,化学品的种类急剧增加,使用量不断加大,加上操作人员的安全意识薄弱,危险化学品事故时有发生,对人民群众的生命财产安全构成极大威胁。而与危险化学品相关的法律法规、制度并不完善。针对我国现阶段危险化学品的储存状况,对危险化学品储存过程中面临的问题进行分析,并探讨解决这些问题的对策和措施。     

The politics of landscape as ways of life in the ‘divided’ city: reflections from Mostar,Bosnia–Herzegovina

ABSTRACT

‘Divided’ cities – places of extreme exclusion and polarization – are sites where the paradox of both fragmentary and cosmopolitan capacity of the city comes to the fore. This article asks: What practices-in-landscape consolidate, construct and deconstruct the impression of a divided city? Drawing insights from the post-conflict Mostar, Bosnia and Herzegovina, the main argument I put forward is that urban infrastructures are a crucial medium through which social divisions and cohesions are performed and rearticulated on the ground. The article attends to the everyday of youth in Mostar turning to the politics of landscape as ways of life in the city.     

司法专业化与社会化的适度平衡:台湾观审制改革及其启示

观审制改革是目前台湾讨论最热烈的"司法改革"议题。所谓观审制,是让经由一定程序选出的普通民众担任"观审员",针对一些重罪案件,全程参与第一审法院的审判程序。评议时,观审员虽不能参与表决,但可以表示意见,为法官下判作参考。观审制改革是台湾民众对司法的不信任与日韩的国民参与审判立法共同作用的结果。观审制主要包括观审员及其选任程序、观审法庭组成及适用案件类型、审判程序、评议程序四部分内容。观审制对目前河南省正在进行的人民观审团机制改革具有重要借鉴意义。
