木丹颗粒对痛性糖尿病周围神经病变大鼠钙通道基因表达的影响

目的研究木丹颗粒对痛性糖尿病周围神经病变(PDPN)大鼠钙离子通道蛋白mRNA表达的影响,探讨木丹颗粒对PDPN的治疗作用。方法雄性Wistar大鼠90只,随机取30只设为正常组,其余大鼠造模,参照Sally等方法,腹腔注射链脲佐菌素STZ(53mg/kg)制作PDPN模型,4周后,用Vonfrey纤维丝测双后足机械痛阈,痛阈明显下降,与基础值相比下降50%者为PDPN造模成功。再将在成模的54只大鼠随机分为模型组,木丹颗粒组。木丹颗粒组给予10ml/(kg·d)灌胃,分别于药物治疗后第2、4、8周末,各组随机取8只大鼠,用RT-PCR技术法检测大鼠背根神经节N型钙离子通道mRNA表达。结果与正常组比,模型组N型钙离子通道mRNA表达在各时间点呈上升趋势(P<0.05);与模型组比,木丹颗粒组N型钙离子通道mRNA表达在各时间点呈下降趋势,8周时,本组N型钙离子通道mRNA表达显著减少(P<0.05)。结论木丹颗粒可以下调PDPN大鼠N型钙离子通道蛋白mRNA表达,对PDPN有一定的治疗作用。
