Analysis of length polymorphisms at STR loci in the human genome has become a standard approach for comparative genotyping in many areas including disease research and diagnostics, parentage assessment, investigations of human diversity, and forensic science. The simultaneous analysis of multiple STR loci through multiplex PCR and multicolor fluorescence detection offers sample conservation, high throughput, and automated genetic analysis. Careful design and optimization of tetranucleotide STR multiplexes has led to reliable, standardized systems that powerfully differentiate and distinguish individual human DNA profiles. The development of these multiplex systems involved a rigorous experimental strategy that included careful selection of PCR primer sequences (for yield, specificity, and multiplex compatability), along with optimization of PCR component concentrations, thermal cycling parameters, and fluorescence detection conditions. This developmental approach rendered well-characterized DNA typing systems that are high performing (sensitive, specific, and balanced), optimized to universal parameters (same reaction conditions), resilient to fluctuations in reaction conditions, and simple to implement and use routinely.  相似文献

Given the drive to balance the federal budget, cutbacks andchanges in federal aid are likely to place added burdens onstate governments. This article examines whether state governmentsare fiscally up to task. A review of the recent and currentfiscal position of the states suggests that current state fiscalstrength may be temporary, and is unevenly distributed amongthe states. In addition, many factors will differentially influencestate responses to the changing federal environment. Recent empirical literature has shown that the determination of intergovernmental grants is highly influenced by the political bargaining power of the recipient states. In these models federal politicians are assumed to buy the support of state voters, state politicians and state interest groups by providing grants. In this paper we provide evidence that the fiscal referendum reduces the reliance of states on matching grants received from the central government and thus the possibility of interest groups and state bureaucrats to obtain more grants. If referendums are available, voters serve as a hard budget constraint.  相似文献

In this article we shall argue the importance of approaching the relationship between party organizations and party members by analyzing the interaction between characteristics of the national party system, the party organization as a more or less open or closed system, and the resources of individuals particularly conducive to party membership. We shall mainly discuss the organizational level, taking as a starting point different models from organizational theory: the rational behaviour model, the survival model, the bureaucratic model, and the symbolic action model. We apply these models in our search for how political parties seek to mobilize members. Examples illustrating the relevance of the different models are a warning to those who wish to develop a general theory of party organization.  相似文献

Changing behavior is often necessary to tackle societal problems, such as obesity, alcohol abuse, and debt problems. This article has two goals. First, it aims to highlight how governments can try to change the behavior of citizens. Government can use policy instruments to do so, including incentives, bans and mandates, information campaigns, and nudges. However, the government should not be a manipulator that applies policy instruments without societal support. Therefore, the second goal of the article is to provide a conceptual framework that helps analyze whether public policies to stimulate behavior change are effective and supported by key stakeholders. This conceptual framework has five criteria indicating the extent to which there is effective and supported behavior change: the policy is (1) effective and (2) efficient, and there is support for the policy among (3) politicians, (4) implementing organizations, and (5) citizens. The article ends with suggestions to study public policy aimed at behavior change.  相似文献

Research recognizes that strategic business interests can provide an important driver of private regulation, even in the absence of significant societal pressure and non‐governmental organization‐constructed demand. This article examines a range of competition and collective action‐related interests that can motivate firms to promote and adopt certification schemes. We pay particular attention to the hitherto underexplored collective action interest to safeguard common‐pool resources, upon which an industry may depend to sustain yields. Based on a case study of salmon aquaculture certification, the article argues that while the corporate motives repertoire includes strengthening competitiveness and industry reputation, safeguarding shared waters for culturing salmon is key to explaining industry commitment to and adoption of private regulation in this sector.  相似文献

We investigate the impact of policy consistency on frontline workers’ perceptions of policy meaningfulness and legitimacy. The results from an experiment involving 779 teachers indicate that policy consistency does have a positive effect on legitimacy and to a lesser extent on meaningfulness. However, the extent depends on policy content and the degree of autonomy. Overall, our findings emphasize the potential positive impact of policy consistency. Although this, to some extent, conflicts with the nature of political decision‐ and policy‐making (i.e., democratically elected governments have been mandated to change policy), our study suggests that policy consistency could be a valuable strategy for governments to strengthen successful policy implementation. This adds a new perspective to the continuing debate within policy implementation and street‐level bureaucracy research on how to account for the complex, messy and sometimes contradictory implementation of public policies.  相似文献

The immediate advantages of adolescent friendships and disadvantages of peer rejection are well documented, but there is little evidence that these effects extend into adulthood. This study tested the hypothesis that peer relationships during adolescence predict life satisfaction during middle adulthood, using data from a 30-year prospective longitudinal study. Participants included 996 (49.5 % female) 8th grade students from a community sample of Swedish youth. Self-reports of friendship and peer reports of rejection were obtained when participants were age 15. Self-reports of global life satisfaction and perceived relationship quality were collected at age 43 for women and age 48 for men. Path analyses tested a direct-effects model that examined links from adolescent friendship participation and peer rejection to middle adulthood outcomes, and a buffered-effects model that examined links from adolescent peer rejection to middle adulthood outcomes, separately for those with and without friends during adolescence. Strong support emerged for the buffered-effects model but not the direct-effects model. Adolescent friendship participation moderated associations between adolescent peer rejection and adult global life satisfaction and between adolescent peer rejection and adult perceived relationship quality such that peer rejection predicted poorer adult outcomes for youth without friends but not for youth with friends. The findings suggest that the risks of peer rejection—and benefits of friendship—extend from adolescence well into middle age.  相似文献
