Four multi-elementary metal and metalloid quantification methods using inductively coupled plasma mass spectrometry (ICP-MS) were developed and validated in human whole blood, plasma, urine and hair by means of a single preparation procedure for each sample. The ICP-MS measurements were performed using a Thermo Elemental X7CCT series and PlasmaLab software without a dynamic reaction cell. With this procedure 27-32 elements can be simultaneously quantified in biological matrices: Li, Be, B, Al, V, Cr, Mn, Co, Ni, Cu, Zn, Ga, Ge, As, Se, Rb, Sr, Mo, Pd, Ag, Cd, Sn, Sb, Te, Ba, W, Pt, Hg, Tl, Pb, Bi, U. Whole blood, plasma and urine samples (0.4 ml each) were diluted with purified water, acid, triton X100 and butanol. Rhodium was used as internal standard. The urine sample results were corrected for enzymatic creatinine determination. Twenty-five milligrams hair samples were acid mineralized after a decontamination procedure and diluted as previously described for biological fluids. To be validated, each element had to show linearity with a correlation coefficient higher than 0.99. The intra-assay and inter-assay inaccuracy, measured as the variation coefficient, were below 5 and 10% respectively. Global performance was assessed by a quality control program. Our laboratory is a registered participant of the Institut National de Santé Publique du Québec (Sainte-Foy, Canada) inter-laboratory comparison program for whole blood, urine, and beard hair of non-occupationally exposed individuals spiked with selected elements. In our study multi-element metal and metalloid analysis was assessed for 27 elements in whole blood, 27 elements in plasma, 30 elements in urine and 32 elements in hair, from 0 to 25, or 250 to 1000 ng/ml, depending on the element. Quantification limits ranged from 0.002 ng/ml (U) to 8.1 ng/ml (Al) for whole blood, from 0.002 ng/ml (U) to 7.7 ng/ml (Al) for plasma, from 0.001 ng/ml (U) to 2.2 ng/ml (Se) for urine, and from 0.2 pg/mg (Tl) to 0.5 ng/mg (B) for hair. Normal values were determined in whole blood (n=100), plasma (n=100), urine (n=100), and hair (n=45) of healthy volunteers, leading to approximately 10,000 analyses. All results are presented and discussed. Clinical toxicology and forensic toxicology applications are also reported. ICP-MS has made significant advances in the field of clinical biology, particularly in toxicological analysis. This is due to the use of extremely effective equipment that permits better clinical and forensic toxicological analysis of metal and metalloid status of each individual patient.  相似文献

The recent House of Lords decision in Quintavalle v Human Fertilisation and Embryology Authority has raised difficult and complex issues regarding the extent to which embryo selection and reproductive technology can be used as a means of rectifying genetic disorders and treating critically ill children. This comment outlines the facts of Quintavalle and explores how the House of Lords approached the legal, ethical and policy issues that arose out of the Human Fertilisation and Embryology Authority's (UK) decision to allow reproductive and embryo technology to be used to produce a 'saviour sibling' whose tissue could be used to save the life of a critically ill child. Particular attention will be given to the implications of the decision in Quintavalle for Australian family and medical law and policy. As part of this focus, the comment explores the current Australian legislative and policy framework regarding the use of genetic and reproductive technology as a mechanism through which to assist critically ill siblings. It is argued that the present Australian framework would appear to impose significant limits on the medical uses of genetic technology and, in this context, would seem to reflect many of the principles that were articulated by the House of Lords in Quintavalle.  相似文献

The fingernails of Mary Sullivan, the last victim of the Boston Strangler, were examined to determine if any genetic information about the murderer could be obtained. The nails were extremely friable necessitating the development of new techniques for isolating and purifying DNA. DNA yields from nails were optimized by using a NaOH-based preparation technique, which was simple, efficient, and minimized handling. Methods for selectively and thoroughly removing exogenous material on nails were also developed through use of a species-specific PCR assay, wherein mitochondrial DNA from the nail could easily be differentiated from DNA of contaminating cells.  相似文献

The use of Japanese swords for homicidal attempts is rare. A Japanese samurai sword is a sharp and cutting object. When faced with the use of this weapon, one must distinguish between stabs and incised wounds. Incised wounds can rarely lead to death, but because of the size of the weapon, stabs usually cause much more serious injuries. Stabs also imply a penetrating movement, whereas incised wounds can be the consequence of protective circular blows. Therefore, it is important to distinguish clinically between these two kinds of wounds. We report a case where the perpetrator argued he had given a circular blow, unfortunately hitting the victim. Sommaire: L'analyse détaillée des rapports d'autorité entre les acteurs de l'arène politique qui impliquent les différentes instances décisionnelles a contribuéà la compréhension des stratégies et des objectifs poursuivis par chacun des acteurs en présence. En outre, la gestion de la crise du verglas a permis de rendre compte de diverses stratégies, notamment celles qui ont conduit les autorités municipales à exercer davantage leur rôle de porte-parole. Des dew rôles du décideur politique, soit dirigeant et porte-parole, celui de porte-parole est le plus manifestement politique car il participe directement au processus de la médiation et de la régulation. En cela, il contribue à une fonction de repréntation de la collectivité. Cette fonction de porteparole des décideurs municipaux a été soutenue, voire favorisée par la centralisation déisionnelle de la gestion de la crise au niveau national, parfois au détriment du rôle de dirigeant des autorités municipales. Le cas de la crise de verglas participe à une tendance générale du système de régulation des rapports entre les élus des différentes instances décisionnelles et il s'inscrit comme mode de contrôle et d'intervention du pouvoir national sur les pouvoirs qui lui sont subordonnés. L'appropriation des opérations par les acteurs politiques de la gouverne nationale s'est réalisée par l'implantation d'un dispositif organisationnel visant la substitution du modèle fonctionnel par un modèlead hoc centralisé. Le modèlead hoc a permis d'intégrer les liens d'autorité habituels et a conduit par la suite à l'établissement de ce que nous avons désigné le modèle effectif.Le bon fonctionnement du modèle effectif exige l'absence d'acteurs potentiellement en situation d'opposition. Les relations entre les acteurs engagés dans les opérations associées à la gestion du sinistre ont dés lors été soumises aux règles et intérêts des acteurs politiques de la gouveme nationale. Abstract: The detailed analysis of the authority relationships between players in the policy arena and the various decision-makers contributed to understanding the strategies and objectives of each player. Moreover, management of the ice storm crisis highlighted various strategies, including those that led municipal authorities to expand their spokesperson role. The role of the spokesperson is more clearly political because it plays a direct part in the mediation and regulatory process, thus contributing to a community representation function. This spokesperson function of municipal decision-makers was supported and even fostered by the centralization of decisions while managing the crisis at the national level (i.e., Quebec), sometimes to the detriment of the leadership role of municipal authorities. The ice storm case is part of an overall trend within the system for regulating the relationships between elected representatives at the various decision-making levels, and it is also a means of national control and intervention over the powers subordinated to the national authority. National policy-makers operated by implementing an organizational system substituting a centralized, ad hoc model for the functional model. Using the ad hoc model, the usual authority linkages were integrated, subsequently leading to the setup of the effective model. For smooth operation, the effective model requires that there be no players holding diametrically opposite positions. The relationships between the players in crisis management operations were thus governed by the rules and interests of national policy-makers.  相似文献

The military manpower demands created by U.S. operations in Afghanistan and Iraq are causing extraordinary strains on recruitment, and the desirable size of the U.S. armed forces has become a topic of national concern. One key issue is the balance between active-duty forces and the reserve components, citizen-soldiers who face numerous service-related issues that the active-duty forces do not and who have their own unique sociology. We cannot continue to rely on the reserves for missions such as Iraq and Afghanistan, but must look to the use of short-term active-duty enlistments, drawn from recent college graduates, to perform duties currently being performed by reservists and even active-duty personnel.  相似文献

This article uses a purposive method of interpretation to suggest solutions to various questions raised in the application of the National Minimum Wage Act (NMWA). The article first considers the goals of minimum wage laws (and the NMWA in particular) by putting forward the justifications for such laws and addressing critiques. It is argued that the minimum wage is best understood as a mechanism for redistribution of resources and ensuring respect for the human dignity of workers. Building on this articulation of goals, the article then proceeds to consider which group of workers are included within the scope of the NMWA (interpreting terms such as 'worker', 'voluntary workers', apprentices and trainees); what are considered working hours for the purpose of the Act (focusing on cases of work/sleep combinations); and what constitutes part of the wage (focusing on tips, attendance allowances and deductions for accommodations).  相似文献

If there is no single long-standing American grand strategy, one nonetheless sees through the course of U.S. history the tracks of a grand strategy. It started with the idea of a U.S. monopoly in the Western Hemisphere, along with balances of power in the chief theaters of the world; with belief in the primacy of sea and air power and the need for an economic system to support these; and the objective of transforming international politics. Since 9/11, even if the strategic hierarchy, intensity, and political basis have changed, the Bush administration has largely been continuing in this same project, with a sensible strategy but poorly considered tactics.  相似文献
