Y-chromosomal short-tandem repeat (Y-STR) amplification has been used in forensic casework at the Bureau of Criminal Apprehension (BCA) Forensic Science Laboratory since 2003. At that time, two separate amplifications were required to type the SWGDAM recommended loci (DYS19, DYS385a/b, DYS389I, DYS389II, DYS390, DYS391, DYS392, DYS393, DYS438, and DYS439). The Yfiler kit coamplifies these loci as well as DYS437, DYS448, DYS456, DYS458, DYS635, and Y GATA H4. The Yfiler kit was validated following the internal validations outlined in the SWGDAM revised validation guidelines. Our studies show that 0.125 ng of male DNA will generate a complete 17 locus profile and that as little as 0.06 ng of male DNA yields an average of nine loci. In the male-male mixtures, a complete profile from the minor component was detected up to 1:5 ratio; most of the alleles of the minor component were detected at a 1:10 ratio and more than half the alleles of the minor component were detected at a 1:20 ratio. Complete YSTR profiles were obtained when 500 pg male DNA was mixed with female DNA at ratios up to 1:1000. At ratios of 1:5000 and 1:10,000 (male DNA to female DNA) inhibition of the YSTR amplification was evident. The YSTR results obtained for the adjudicated case samples gave significantly more information than the autosomal results. Our studies demonstrate that the Yfiler kit is extremely sensitive, does not exhibit cross-reactivity with female DNA, successfully types male DNA in the presence of overwhelming amounts of female DNA and is successful in typing actual forensic samples from adjudicated cases.  相似文献

This article presents research from a three-year study of shifting understandings of threat and security in Britain following the 2003 Iraq War. We develop the case for a more integrated and nuanced approach to studying the relationship between policymakers, media practitioners and media publics given the increasing importance of these relationships to international relations (IR) matters of concern. Our analysis demonstrates the ‘push’ and ‘pull’ factors that explain why certain individuals and groups arrive at certain understandings or perceptions of threats. Responding to recent calls in IR for the use of diverse and interdisciplinary methods, our methodology enables us to demonstrate how disparities emerge between official and public understandings of threats. These understandings result from people's engagement with political and media discourses, and the experience of this engagement can be characterized by connectivity, (un)certainty and contradiction.  相似文献

Sommaire: Plusieurs gouvernements ont mis en place des processus de réformes qui se traduisent par un processus de changement visant la modernisation de l'État. Ces processus cornportent une dimension axiologique influencée par la Nouvelle gestion publique. Considérant la réforme comme un phénomène psychosocial et culturel, cet article expose la perception d'acteurs clés vis‐à‐vis des processus de changement qu'implique la modernisation de la fonction publique quebecoise (Loi sur l'administration publique). Cette recherche, basée sur vingt entrevues réalisées auprès de direc‐teurs des ressources humaines de ministères et organismes, démontre que les perceptions de la réforme sont diverses et que l'influence de la politique dans les réformes se situe davantage dans les objectifs que dans le design. En plus de faire ressortir les principales initiatives et quelques obstacles au changement, la recherche illustre que la perception qu'ont les drh sur la nature réelle des changements est variable, et permet de les regrouper sous trois catégories. Enfin, cette recherche présente différents types de cultures témoignant de l'importance d'ajuster les modes d'implémentation pour le succès des réformes. Elle illustre l'importance de soigner la mise en cruvre des réformes plutôt que d'essayer de définir un modèle idéal. Un regard sur l'avenir vient enfin compléter cette étude. Abstract: Many governments have implemented reform processes that lead to modernizing the state. These experiences involve a value dimension influenced by new public management. Stating administrative reform as a cultural and psychosocial phenomenon, this article exposes the perception that key actors have of the change processes entailed by the Quebec public administration's modernization (Loi sur l'administration publique). This research, based on twenty interviews of human resources directors from ministries and public organizations, shows that the perceptions of the reform are diverse and that the political influence of the reforms appears more in the objectives than in the design. In addition to showing the principal facilitating initiatives and a few obstacles to change, the research illustrates that the human resources directors' perception of the changes' actual nature is variable and permits grouping them into three categories. Finally, this research presents different types of cultures, showing the importance of adjusting implementation modes for the reforms' success. It illustrates the importance of taking care of the implementation rather than trying to define an ideal model. A look at the future completes the study.  相似文献

Social science research contains a wealth of knowledge for people seeking to understand collaboration processes. The authors argue that public managers should look inside the "black box" of collaboration processes. Inside, they will find a complex construct of five variable dimensions: governance, administration, organizational autonomy, mutuality, and norms. Sommaire: Depuis le milieu des années 1990, plusieurs gouvernements ont affirmé leur volonté de tenir compte des impacts de leurs actions sur la santé des populations. En juin 2002, le gouvernement du Québec met en vigueur l'article 54 de la Loi sur la santé publique (L.R.Q. chapitre S‐22, 2001). Cet article atteste de l'obligation pour les ministères de tenir dorénavant compte des impacts sociaux, environnementaux et économiques de leurs actions sur la santé. La mise en Oeuvre de cet article suppose l'intégration d'un processus d'évaluation prospective des politiques publiques dans l'appareil gouvernemental, remet en question le fonctionnement vertical des administrations publiques et souligne l'importance d'une gestion stratégique des connaissances dans un monde sans frontières. Dans ce contexte, quatre dilemmes sont soulevés. Les deux premiers dilemmes sont d'ordre interministériel. Ils concernent les relations qu'entretiennent les ministéres entre eux et avec les organismes centraux. Les deux autres dilemmes s'intéressent à la dimension intra organisationnelle, c'est‐à‐dire la maniére dont chaque ministère réagit face aux demandes d'évaluations prospectives de ses politiques ou programmes et répond aux exigences dc l'article 54. Des méthodes qui devraient permettre aux administrations publiques de remédier à ces différents dilemmes sont également présentées. Abstract: Since the mid‐1990s, several governments have declared their commitment to ensuring that the impact of their actions on the health of the population is taken into account. In June 2002 the Quebec provincial government implemented article 54 of the Public Health Act (R.S.Q., chapter 2.2, 2001). This article stipulates the obligation on the part of the government departments to take into account the social, environmental, and economic impacts of their actions on public health in the future. The implementation of this article means integrating a process for the prospective assessment of the government's public policies, and re‐examining the vertical model of operation of public administrations; it also underlines the importance of strategic knowledge management in a world without borders. Four dilemmas are raised within this context. The first two dilemmas are interdepartmental in nature, and involve the relationships the government departments have with each other and with the central agencies. The other two dilemmas fall within the intra‐organizational sphere ‐ that is, the way in which each government department reacts to the demands for the prospective assessment of its public policies or programs, and how each department responds to the requirements of article 54. Processes that should allow the public administrations to solve these various dilemmas are also presented.  相似文献

This paper uses the official juvenile offenses among delinquent girls in the 1958 Philadelphia Birth Cohort to investigate the two overarching questions. This research investigates the nature of delinquency conduct, offense-by-offense, and its relationship to adult crime status. Although it is convenient to think of an offender’s delinquency career as a whole, such a career actually consists of one or more specific offenses, and offense conduct can be worth studying in its own right. Thus, it is necessary to determine whether the timing, type, severity, court disposition, and so on, of these juvenile offenses can be used to predict adult career pathways. An extensive review of the literature revealed that investigations of early offense conduct and its connection to adult crime are exceedingly scarce. This study indicates as follows. First, the way a delinquent begins her criminal career is predictive of the adult trajectory that will be followed. Second, we also found that aspects of the first few offenses doe influence whether delinquent girls do continue committing crimes as adult. The strongest predictor of adult crime status was juvenile court dispositions.  相似文献

Sommaire : Les politiques publiques du travail ont été conçues pour offrir des mesures de protection aux salariés travaillant à temps plein, pour un seul employeur, sous son contrôle et sur les lieux de l'entreprise. Or, le marché du travail est aujourd'hui composé d'une pluralité de relations d'emploi qui limitent l'efficacité de ces protections. Ce constat soulève la question suivante : que devraient contenir les politiques publiques pour être mieux adaptées au marché du travail? La première partie de cet article examine les voies de renouvellement possible des politiques publiques pour tenir compte des transformations du travail et de l'entreprise. Certaines suggèrent de faire reposer l'octroi de droits et d'avantages sur l'existence d'un travail ou d'une activité professionnelle alors que d'autres les rattachent à l'individu ou au citoyen. Ces propositions ne pourront cependant voir le jour que si elles font l'objet d'une appropriation par les acteurs sociaux et étatiques. Une réflexion sur le contenu des politiques doit ainsi être combinée à l'étude des processus sociaux susceptibles de faire émerger de telles réformes. Cette question est abordée dans la deuxième partie de l'article à partir de trois exemples récents de réformes législatives. Abstract: Public labour policies were developed to ensure the protection of workers who work for one employer on a full‐time basis, at the employer's place of work and under his control. However, today's labour market is composed of a multiplicity of employment relations that limit the effectiveness of the protection. This raises the following question: what features should public policies include to better meet the needs of the labour market? The first part of this article examines how public policies could be renewed in order to take work and organizational transformations into account. Some policies suggest that rights and benefits should be granted on the basis of a job or professional activity, whereas others link rights and benefits to the individual or the citizen. However, these proposals will only emerge if they are appropriated by social and governmental stakeholders. A reflection on the content of the policies must therefore be undertaken in combination with a study of the social processes that may influence the emergence of such reforms. This question is examined in the second part of the article through a review of three recent examples of legislative reform initiatives.  相似文献
