The problem of DNA contamination in forensic case work—How to get rid of unwanted DNA?

The PCR technique has become a powerful and very sensitive tool in a broad field of research, that is, molecular biology, medical diagnostics, population genetics, ancient DNA analysis and forensic casework.However, the high sensitivity down to single molecules can easily cause false-positive PCR results due to different types of contamination. In this study, artificial DNA contaminations (saliva and pure DNA) were treated with UV irradiation and other decontamination procedures. A satisfactory DNA removal could not be achieved, emphasizing the necessity of contamination avoidance.     

Sequence polymorphisms at the DXS6789, DXS8377 and DXS101 loci in three Asian populations

Sequence analyses of X-chromosomal short tandem repeats, DXS6789, DXS8377 and DXS101 were performed for representatives of 3 Asian populations: 130 Japanese, 61 Bangladeshi and 89 Indonesian males. At DXS6789, the sequence polymorphism was found in 7 alleles in the Japanese, 3 in the Bangladeshis and 3 in the Indonesians. At DXS8377, the sequence polymorphism was found in 13 alleles in the Japanese, 9 in the Bangladeshis and in all alleles identified in the Indonesians. At DXS101, the sequence polymorphism was found in 7 alleles in the Japanese, 9 in the Bangladeshis and 8 in the Indonesians. Because sequence polymorphisms were found in most of the alleles at the DXS6789, DXS8377 and DXS101 loci, it was concluded that sequencing was essential for identifying the alleles at these loci in all 3 Asian populations.     

Optimization and validation studies of the Mentype Argus X-8 kit for paternity cases

The use of ChrX-STRs is enormous in forensic case as these have proven to be powerful tools, mainly in deficiency paternity cases when the disputed child is female, and also some special cases involving blood relatives, incest cases, fetal typing in abortion material. The Mentype^® Argus X-8 kit is a commercial multiplex system which contains Amelogenin for gender determination as well as gonosomal STR markers (DXS8378, HPRTB, DXS7423, DXS7132, DXS10134, DXS10074, DXS10101 and DXS10135). Validation studies were being performed on blood obtained from the volunteers in Turkish population. In this study, some parameters were taken under consideration for validation like DNA extraction using different protocols, quantitated by using commercially available Invitrogen Qubit Fluorometer, reaction volume validation of Master Mix and the analysis of female/male, female/female and male/male mixtures were performed. The conditions were optimized and validated using GenAmp 9700 and reducing reaction volume from 25 μl to 12.5 μl and 6.5 μl. After reducing the total volume of the reaction, the results were same and there was no effect on peak height and quality when analyzed on ABI 310 genetic analyzer. 2 paternity cases were also performed which gave the same power of discrimination as has been mentioned in Mentype^® Argus X-8 kit.     

Mortgage fraud: A risk factor analysis of affected communities

Mortgage fraud is a fast-growing form of white-collar crime that has received much press coverage in the United States of America. Mortgage fraud has an adverse effect on individual homeowners, communities, and many indirect victims of the crime. While past research has focused on the personal motivating factors behind the commission of white-collar crime, this particular article reviews several facets of the crime itself and explores the potential neighbourhood risk factors that help attract the crime. From a national perspective, mortgage fraud seems to occur more frequently in neighbourhoods that have low socioeconomic indicators. These associations become even more pronounced when the degree of fraud occurrences within the community is factored in as a variable. Upon disaggregating the data according to region, the fraud indicator variables also display differing trend levels, perhaps indicating that as mortgage fraud practices begin to mature within an area, its community dynamics tend to change as well. The article concludes with recommendations for policymakers, community organizations, and law enforcement officials as to how to address mortgage fraud once it appears within a community, and also addresses future avenues of research for what is largely an untapped area of financial crime research.     

Psychopathy and instrumental aggression: Evolutionary, neurobiological, and legal perspectives

In the study of aggression, psychopathy represents a disorder that is of particular interest because it often involves aggression which is premeditated, emotionless, and instrumental in nature; this is especially true for more serious types of offenses. Such instrumental aggression is aimed at achieving a goal (e.g., to obtain resources such as money, or to gain status). Unlike the primarily reactive aggression observed in other disorders, psychopaths appear to engage in aggressive acts for the purpose of benefiting themselves. This is especially interesting in light of arguments that psychopathy may represent an alternative life-history strategy that is evolutionarily adaptive; behaviors such as aggression, risk-taking, manipulation, and promiscuous sexual behavior observed in psychopathy may be means by which psychopaths gain advantage over others. Recent neurobiological research supports the idea that abnormalities in brain regions key to emotion and morality may allow psychopaths to pursue such a strategy—psychopaths may not experience the social emotions such as empathy, guilt, and remorse that typically discourage instrumentally aggressive acts, and may even experience pleasure when committing these acts. Findings from brain imaging studies of psychopaths may have important implications for the law.     

Forensic analysis of explosives using isotope ratio mass spectrometry (IRMS) — Discrimination of ammonium nitrate sources

An evaluation was undertaken to determine if isotope ratio mass spectrometry (IRMS) could assist in the investigation of complex forensic cases by providing a level of discrimination not achievable utilising traditional forensic techniques. The focus of the research was on ammonium nitrate (AN), a common oxidiser used in improvised explosive mixtures.The potential value of IRMS to attribute Australian AN samples to the manufacturing source was demonstrated through the development of a preliminary AN classification scheme based on nitrogen isotopes. Although the discrimination utilising nitrogen isotopes alone was limited and only relevant to samples from the three Australian manufacturers during the evaluated time period, the classification scheme has potential as an investigative aid.Combining oxygen and hydrogen stable isotope values permitted the differentiation of AN prills from three different Australian manufacturers. Samples from five different overseas sources could be differentiated utilising a combination of the nitrogen, oxygen and hydrogen isotope values. Limited differentiation between Australian and overseas prills was achieved for the samples analysed.The comparison of nitrogen isotope values from intact AN prill samples with those from post-blast AN prill residues highlighted that the nitrogen isotopic composition of the prills was not maintained post-blast; hence, limiting the technique to analysis of un-reacted explosive material.     

“‘I have done it and I have got to die’: Coroners' inquests of abortion deaths in Rhode Island, 1876–1938.”

This article offers an analysis of abortion deaths among white working-class women in Providence County based on thirty-three coroners records from 1876 to 1938. Most women were single and in their twenties: they chose abortion either because their lover was married, or they were too ashamed of their premarital sexual activity to confront their parents. Married women, on the other hand, did so primarily due to economic factors, extramarital affairs, or a strong desire for no more children. In seeking abortions, single women depended on lovers while married women relied on sisters or sisters-in-law. In investigating the deaths resulting from these abortions, coroners called husbands, but not lovers, before the inquests and sought out antemortem statements. Yet only 39% of cases had dying declarations: either doctors refused to participate in interrogating women on their death beds, or women refused to identify abortionists. Of the abortion providers identified, 45% were physicians. Working-class status did not prevent these women, and later their families, from securing expensive medical care from doctors. Both inquests and newspaper coverage in Providence County differ from studies by other scholars who find these venues used as mechanisms to embarrass women and warn single women of urban threats, and to target midwives over physicians. The evidence points to analogous handling of doctors and laypeople, and of married and single women. No intimate details of women's lives were exposed and newspapers did not use sensationalized headlines to draw attention to the illegal activity of abortion. Coroner inquests asked questions necessary to investigate death from a criminal activity, not to take a moral stance on female sexuality.     

Determining the sequence of intersecting gel pen and laser printed strokes — A comparative study

The relative chronological sequence of intersecting gel pen ink lines and laser printed marks has been determined using optical methods. The results were obtained by observing spectral reflection, relative sheen, skipping and gaps of gel pen ink strokes at the point of intersection. The intersections are observed under stereomicroscope and digital microscope. The effectiveness of both the optical instruments for sequence of the strokes has been compared. The sequence of intersecting lines (of same colour, different colours and where one of the strokes is darker) has been established.
