STR analysis of artificially degraded DNA-results of a collaborative European exercise

Degradation of human DNA extracted from forensic stains is, in most cases, the result of a natural process due to the exposure of the stain samples to the environment. Experiences with degraded DNA from casework samples show that every sample may exhibit different properties in this respect, and that it is difficult to systematically assess the performance of routinely used typing systems for the analysis of degraded DNA samples. Using a batch of artificially degraded DNA with an average fragment size of approx. 200 bp a collaborative exercise was carried out among 38 forensic laboratories from 17 European countries. The results were assessed according to correct allele detection, peak height and balance as well as the occurrence of artefacts. A number of common problems were identified based on these results such as strong peak imbalance in heterozygous genotypes for the larger short tandem repeat (STR) fragments after increased PCR cycle numbers, artefact signals and allelic drop-out. Based on the observations, strategies are discussed to overcome these problems. The strategies include careful balancing of the amount of template DNA and the PCR cycle numbers, the reaction volume and the amount of Taq polymerase. Furthermore, a careful evaluation of the results of the fragment analysis and of automated allele calling is necessary to identify the correct alleles and avoid artefacts.     

Female repeat offenders negotiating identity

This study examines the process of identity negotiation for Israeli female ex-convicts who were separated for extensive periods of time from their children and eventually lost custody over them. The content analysis of in-depth interviews reveals that these women were able to reconstruct their biographies and retrospectively account for their crimes and drug addiction in terms of the sexual, physical, and economic abuse they had endured and by appeal to higher loyalties, their children who they had to provide for. However, when having to account for their fallings as mothers, all biographical reconstruction, external blame, and accusation collapsed. Looking at themselves through their children's eyes, female offenders were simply unable to renegotiate the imputed identity of incompetent mother. They could neither confront their children's anger nor explain to them why they had abandoned them. Permanently alienated from the center of motherhood, these women were doomed to an existential chaos.     

Correlates of self-concept among black early adolescents

This study examines the self-concept of Black eighth-grade students from the Midwest in relation to Black acceptance, social intimacy, locus of control, and sex-role type. Twenty-eight students high in self-concept were compared with 31 students whose self-concept was low. As predicted, the high self-concept group scored higher than the low self-concept group in intimacy, internality, and acceptance of black identity. As also predicted, the high self-concept group had a significantly greater number of adolescents with masculine and androgynous sex roles than the low self-concept group; Black females with high self-concepts included a larger proportion of individuals with androgynous sex roles than low self-concept females. However, the prediction that high self-concept males would have a larger proportion of masculine sex roles than low self-concept males was not supported. The difficult situation of the low self-concept adolescents is discussed, along with the implications for intervention.On clinic internship at Smolian Psychiatric Clinic in Birmingham, Alabama. Linking Policy Networks and Environmental Policies: Nitrate Policy Making in Denmark and Sweden 1970-1995

The policy network literature has provided important insight into the way in which public policy is made in Western societies. Most network studies have focused on processes within networks and have paid little attention to the conceptualization of policy outcomes and the theoretical link between network type and policy type. This article defines and categorizes environmental policy and suggests a proposition on the link between network types and environmental policy types. It is argued that the existence of tight and closed policy communities in sectors subject to environmental regulation is associated with the introduction of low cost environmental policies. In contrast, open and loose issue networks are associated with high cost environmental policies.
