Postmortem stability of DNA   总被引:11,自引:0,他引:11  
High-molecular-weight DNA was recovered postmortem in sufficient quantities from various human organ tissues as well as from blood, although not all organs were equally well suitable. Good DNA stability was found in brain cortex, lymph nodes and psoas muscle over a period of three weeks postmortem. Spleen and kidney showed good DNA stability up to five days postmortem but after longer periods, rapid degradation was observed. Yields of DNA from blood were not consistent because of the non homogeneity of samples. Blood clots were rich with DNA. Generally, the amount of degraded DNA correlated directly with the duration of the postmortem period. However in some cases, DNA degradation was already prominent after a short period. Case histories showed that high environmental temperature at the site of death and/or infectious diseases prior to death were the main factors for rapid autolysis. Gradual disappearance to complete loss of the long fragments (15-23 kb) was observed in DNA fingerprinting using the minisatellite probe 33.15. No extra-bands were noted, thus excluding erroneous conclusions. However, evidentiary value of older samples was lower.  相似文献

This article introduces the concept of ‘deep marketisation’ as a relatively new, contemporary phase of neoliberal development policy in Bangladesh. By looking into the development strategy of the country’s energy sector, the article shows how an emphasis on marketisation through public-private partnerships (PPPs) and other strategies advances a market fundamentalist agenda to strengthen the private sector and establish a world market. By drawing on interviews with development practitioners from various development organisations in Bangladesh, the article further reveals how development conceptualisations are shaped by the strategy of deep marketisation, leading to the impoverishment of development by constraining its field of actions to measures based on the primacy of economic growth and private sector-led economic development, at the same time leading to a re-legitimisation of flawed neoliberal development policies that result in further inequality, poverty and environmental degradation.  相似文献

As we know from comparative public policy, bureaucracies contribute to a considerable degree to the contents and the ways of policy-making. One important driver of administrative policy-making are their specific “styles” or “cultures”. “Administrative styles” are understood here as the standard operating procedures and routines that characterize the behavior and activities of administrative bodies in initiating, drafting and implementing policy. In this article, we convey the concept of Administrative Styles to the level of International Organizations (IOs) and apply it to the Organization for Economic Development and Co-Operation (OECD). The article proceeds in three steps: First, the concept of administrative styles is introduced and refined. Drawing on expert interviews with OECD staff, we secondly show that consideration of OECD administrative styles significantly advances the literature’s understanding of the organization. Finally, we give an outlook on new research avenues and the relevance of our findings for the study of International Public Administrations (IPAs) more generally.  相似文献

Public and academic debates often lament a “crisis of trust” endangering democratic stability. In contrast, this article seeks to demonstrate the value of distrust in political relations, a certain form of which should be considered, such as trust, a prerequisite of democracies. To underline this thesis, this contribution discusses categories that clarify the relationality of the concept of distrust (its directedness and modalities) and that allow for a discussion of the relation of trust and distrust, which are introduced as an asymmetrical pair of opposites – with far-reaching consequences for the common discourse on trust. After presenting several historical and current arguments in favor of the value of political distrust, this article will end with a deeper look on those forms of distrust that should indeed be considered dysfunctional and dangerous for democratic orders.  相似文献

Since the 1980s, regulated markets and New Public Management have been introduced in the public sector across the world. How they have affected existing governance mechanisms such as self‐regulation and state regulation has remained largely unexplored, however. This article examines the origins and consequences of institutional layering in governing healthcare quality. Dutch health care, where a market‐based system has been introduced, is used as a case study. The results show that this market‐based system did not replace but modified existing institutional arrangements. As a result, hospitals have to deal with the fragmentation of quality demands. Using the concept of institutional layering, this study shows how different arrangements interact. As a consequence, the introduction of a certain policy reform will work out differently in different countries and policy sectors. Our ‘archaeological’ study in this layering can be seen as an example of how such incremental change can be studied in detail.  相似文献

The standard view of the negotiations on mutual and balanced force reductions 1973–86 is that no progress was made because the Warsaw Pact sought to protect its existing advantage in conventional forces in Europe, while the western side was against any reductions. Moreover, the political objectives of the talks (for the eastern side the conclusion of CSCE, for the western side the maintenance of domestic support for troop commitments to Europe) were achieved without any progress in the negotiations. Research in German archives shows that for a time there was strong support in the west German government for far more ambitious goals seeing the talks as a mechanism to affect fundamentally the structure of European security on the basis of more radical force reductions. This paper analyses the results of this research, its influence on the official west German position on MBFR and the interaction with the decision‐making process in the NATO alliance.  相似文献

The article comparatively investigates reforms of human resource management in the European Commission and the OECD Secretariat. It analyzes to what extent both organizations have adjusted their respective structures towards the ideal type of the so-called New Public Management (NPM). The empirical findings show that reforms towards NPM are more pronounced in the Commission than in the OECD Secretariat. These findings are surprising for two reasons. First, it seems rather paradoxical that the OECD as central promoter of NPM at the international level lags behind the global trend when it comes to reforming its own structures. Second, this result is in contradiction with theoretical expectations, as they can be derived from theories of institutional isomorphism. To nevertheless account for the surprising results, it is necessary to modify and complement existing theories especially with regard to the scope conditions of their causal mechanisms.  相似文献
