Development and validation of the AmpFℓSTR® Identifiler® Direct PCR Amplification Kit: a multiplex assay for the direct amplification of single-source samples

Abstract: The AmpF?STR^® Identifiler^® Direct PCR Amplification Kit is a new short tandem repeat multiplex assay optimized to allow the direct amplification of single‐source blood and buccal samples on FTA^® card without the need for sample purification and quantification. This multiplex assay has been validated according to the FBI/National Standards and SWGDAM guidelines. Validation results revealed that slight variations in primer concentration, master mix component concentration, and thermal cycling parameters did not affect the performance of the chemistry. The assay’s sensitivity was demonstrated by amplifying known amounts of white blood cells spotted onto FTA^® cards, and the assay’s specificity was verified by establishing minimal cross‐reactivity with nonhuman DNA. No effect on the age of the sample stored on the FTA^® substrate was observed and full concordance was established in the population study. These findings of the validation study support the use of the Identifiler^® Direct Kit for forensic standards and database samples genotyping.     

Rapprochement between Taiwan and the Chinese Mainland: implications for American foreign policy

This article examines US policy toward the improving relations between the Republic of China on Taiwan and the People's Republic of China. It also analyzes several policy options that the Barack Obama administration may wish to consider. In conclusion, the author suggests that, despite some arguments to the contrary, continuing the current policy supporting the growing rapprochement between Beijing and Taipei is in the best interest of the US.     

Determining Bruise Etiology in Muscle Tissue Using Finite Element Analysis

Bruising, the result of capillary failure, is a common physical exam finding due to blunt trauma and, depending on location and severity, a potential indicator of abuse. Despite its clinical relevance, few studies have investigated the etiology of capillary failure. The goal of this study was to determine whether capillaries primarily fail under shear stress or hydraulic‐induced tensile stress. An arteriole bifurcating into four capillaries was modeled using ANSYS 14.0 ^®. The capillaries were embedded in muscle tissue and a pressure of 20.4 kPa was applied. Any tensile stress exceeding 8.4 × 10⁴ Pa was considered failure. Results showed that failure occurred directly under the impact zone and where capillaries bifurcated, rather than along the line of greatest shear stress, indicating that internal tensile stress is likely the primary mode of capillary failure in bruising. These results are supported by the concept that bruising can occur via blunt trauma in which no shearing lacerations occur.     

Massive ovarian cyst and sudden death

ABSTRACT: Massive enlargement of an ovarian cyst is an uncommon cause of morbidity and a rare cause of mortality due in large to part to noninvasive imaging techniques that usually permit early detection. When an ovarian cyst reaches giant proportions, it produces abdominal enlargement often with a fluid wave resulting in a condition that mimics ascites, called pseudoascites. Despite their impressive appearances, such cysts often are operable for cure. We describe a case of a middle-aged woman who presented 3 years before her death with symptoms from an undiagnosed giant cyst and given a diagnosis of ascites of undetermined etiology. She subsequently died at home unexpectedly, and at autopsy, she was found to have a massively enlarged but otherwise benign mucinous cystadenoma.     

The application of venture analysis to public policy decisions: A case study of photovoltaics

Venture analysis is a group of analytical techniques normally used by the private sector to assist in major capital decisions. The approach can be applied to public policy decisions in those cases where changing private sector investment decisions are the central focus. A case study concerning a temporary subsidy to an emerging energy technology is reviewed. Major strengths of the approach are its use of numerous data- and opinion-gathering techniques and its explicit treatment of market and supply uncertainties. The major weakness is its lack of ability to handle those social costs and benefits for which no market exists. Implications for other policy studies are also presented.
