Most commercially available STR amplification kits have never been fully validated for low template DNA analysis, highlighting the need for testing different PCR kits and conditions for improving single-cell profiling. Here, current strategies rely mainly on adjusting PCR cycle number and analytical threshold settings, with a strong preference for using 30 amplification cycles and thresholds at 30–150 RFU for allele detection. This study aimed to (1) determine appropriate conditions for obtaining informative profiles utilizing a dilution series, and (2) test the outcome on single cells using the DEPArray™ technology. Four routinely applied forensic STR kits were compared by using three different amplification volumes and DNA dilutions down to 3.0 pg, while two well-performing kits were used for single/pooled leucocyte and sperm cell genotyping. Besides reduced costs, the results demonstrate that a 50%–75% PCR volume reduction was beneficial for peak height evaluation. However, this was counteracted by an increased artifact generation in diluted DNA volumes. Regarding profile completeness, the advantage of volume reduction was only prominent in samples processed with Fusion 6C. For single and pooled cells, ESIFast and NGMDetect provided a solid basis for consensus profiling regarding locus failure, although locus dropouts were generally observed as stochastic events. Amplification volume of 12.5 μL was confirmed as appropriate in terms of peak heights and stutter frequencies, with increased stutter peaks being the main artifact in single-cell profiles. Limitations associated with these analyses are discussed, providing a solid foundation for further studies on low template DNA.  相似文献

Social order and security depend on mutual cooperation between the police and the public. Since the majority of crime is not detected by the police itself, informal control is needed to ensure order in society. This article aims to describe the circumstances under which people´s willingness to cooperate with the police is enhanced. Recent studies show that public compliance and cooperation with authorities who carry out criminal proceedings are linked with the extent to which people perceive these authorities as trustworthy and legitimate. Importantly, trust in police procedural fairness leads to the perception that institutions of justice are legitimate, which in turn enhances people´s willingness to cooperate with them in order to fight crime and disorder. This normative perspective is supported in many European countries. However, evidence exists that instrumental judgements, which focus on one´s self-interest and on outcomes of the justice system, could also be important in some countries. Drawing on procedural justice theory, we examine the importance of normative and instrumental factors in eliciting people´s readiness to help the police fight crime in four Central European countries: the Czech Republic, the Slovak Republic, Hungary, and Poland. While the procedural justice pattern, i.e. the normative perspective, holds well in the Czech Republic and Hungary, in other analysed countries trust in police effectiveness or fear of crime, i.e. instrumental judgements, are relevant too.  相似文献

Abstract

Various terms have been used to describe the intersection between computing technology and violations of the law-including computer crime, electronic crime, and cybercrime. While there remains little agreement on terminology, most experts agree that the use of electronic devices to commit crime has increased dramatically and is now commonplace. It is the role of the digital investigator to bring cybercriminals to justice. Cybercrime however differs from traditional crime and presents a variety of unique challenges including the variety of electronic devices available, amount of data produced by these devices, the absence of standard practices and guidelines for analyzing that data, the lack qualified personnel to perform investigations and the lack of resources to provide on-going training. Urban planning in Norway can be characterised as market oriented, with responsibilities for the formulation of planning largely delegated to private developers. Even though the principle of citizen participation has a strong and longstanding tradition in Norway, the market-oriented practices challenge the ability of citizens to influence their spatial surroundings. Based on broad surveys and qualitative case studies, this article maps the attitudes of developers, councillors and planners towards citizen participation and studies the strategies of local community associations. Our analysis shows that developers value citizen participation to a much lesser degree than councillors and planners, which can explain the lack of participatory channels in early phases. Official avenues for participation occur later, primarily through hearings. Local associations find this to be too late, characterising urban planning as a path dependent process; as a consequence, local associations attempt to influence planning processes in a more informal way by contacting councillors directly.  相似文献

Abstract: Using Swiss data from the 2003 International Social Survey Programme (N = 902), this multilevel study combined individual and municipality levels of analysis in the explanation of nationalism, patriotism and exclusionary immigration attitudes. On the individual level, the results show that in line with previous research nationalism (uncritical and blind attachment to the nation) increased exclusionary immigration attitudes, while patriotism (pride in national democratic institutions) was related to greater tolerance towards immigration. On the municipality level, urbanization, socioeconomic status and immigrant proportion (and their interaction effects) were found to affect nationalism, patriotism and immigration attitudes. Nationalist and patriotic forms of national attachment were stronger in German‐speaking municipalities than in the French‐speaking municipalities. Path analyses further revealed that living in a Swiss‐German municipality indirectly led to more negative immigration attitudes through an increase in nationalism. The research is discussed in light of social psychological and political science literature on political attitudes.  相似文献

European economic integration creates unintended consequences for national public administrations. This article offers a conceptual and empirical analysis of how these challenges are met. First, three challenges are identified: a reduced capacity to offer services to citizens who move freely, increasing administrative burdens, and negative externalities for all parts from a single states’ administrative failure. Second, a conceptual framework is developed that links each challenge to a most likely response in form of modes of administrative cooperation. Third, the framework is illustrated by an empirical analysis of the coordination of social security systems, labor inspectorates, and posted workers. The case studies show that horizontal administrative cooperation is developed stepwise over time and in line with the theoretical framework. In sum, we can sustain that horizontal administrative cooperation is a relevant additional integration dynamic that buffers unintended effects of market integration on formally independent but increasingly interdependent member state executive bodies.  相似文献

Previous scholarship on variations in violence within a given terrorist organization has primarily focused on factors that lead to the inception or destruction of that organization. However, violence varies substantially even during the “prime” of an organization's life. This article aims to understand why violence varies in the short term within many organizations, and places a special focus on declines in violence. Specifically, I argue that terrorists face countervailing incentives in terms of how much violence to use, and that when declines in violent activity do occur, they can be divided into two types: a) elective declines, which are usually temporary and used for organizational or reputational recovery; and b) imposed declines, which are dictated by changes in the relative capability of an organization, and are more likely to be permanent. The causal pathways to each type of decline are discussed, and a plausibility probe, consisting of case studies of three terrorist organizations, is then developed to substantiate this theory. The findings have notable implications for counterterrorism policy, as they illustrate not only when and why terrorists choose to curtail violent attacks, but also the conditions that determine whether declines in violence are temporary or permanent.  相似文献

As Britain prepares to leave the European Union after the popular vote of June 2016, the government is embarking on the revision of foreign policy. Boris Johnson, or ‘just Boris’, has been entrusted with forging the new ‘Global Britain’ for the post-Brexit era and reinventing British economy around new relationships. Boris has a track record of misrepresenting and offending foreign peoples, leaders and countries. This article assesses the prospects for Africa in Johnson’s vision for ‘Global Britain’ as presented in his foreign policy speeches. The paper unpacks Johnson’s discursive construction of ‘Africa’ and inserts it into a broader historical and political context of British relations with Africa. It argues that, by constructing Africa as a ‘problem’ and offering liberal values as a condition for development, Johnson is continuing British imperial and post-colonial discourses of ‘developing’ or ‘civilizing’ Africa. In the post-Brexit world of a changing global balance of power, democratic conditionality serves to sustain and reproduce British forms of power and policies.  相似文献
