Direct PCR Improves the Recovery of DNA from Various Substrates

This study reports on the comparison of a standard extraction process with the direct PCR approach of processing low-level DNA swabs typical in forensic investigations. Varying concentrations of control DNA were deposited onto three commonly encountered substrates, brass, plastic, and glass, left to dry, and swabbed using premoistened DNA-free nylon FLOQswabs^™. Swabs (n = 90) were either processed using the DNA IQ^™ kit or, for direct PCR, swab fibers (~2 mm²) were added directly to the PCR with no prior extraction. A significant increase in the height of the alleles (p < 0.005) was observed when using the direct PCR approach over the extraction methodology when controlling for surface type and mass of DNA deposited. The findings indicate the potential use of direct PCR for increasing the PCR product obtained from low-template DNA samples in addition to minimizing contamination and saving resources.

ABSTRACT

China has a highly centralized bureaucracy that is no longer strictly monitored by political loyalty but by governance performance (e.g., economic growth), rewarded with promotion and monetary incentives. In the early 2000s, environmental criteria were added to this system. As part of this effort, a high-powered performance management system was introduced in 2006. It held high-level provincial officials, who are part of the nomenklatura, personally responsible for meeting specific emissions targets. Using data from China Statistics Yearbooks and several official news archives, the empirical results indicate that the implementation of the new performance management system reduced emissions only for air pollutants, which are the most publicly visible among the targeted pollutants. Water pollution, which is less visible but also a mandated target, was unaffected. Emissions of soot, an untargeted pollutant, were also unaffected. The findings imply that, even in centrally managed systems like China, compliance with a high-stakes reward for measured performance is not universal.

Urgent issues and prospects at the intersection of culture,memory, and witness interviews: Exploring the challenges for research and practice

The pursuit of justice increasingly relies on productive interactions between witnesses and investigators from diverse cultural backgrounds during investigative interviews. To date, the role of cultural context has largely been ignored by researchers in the field of investigative interviewing, despite repeated requests from practitioners and policymakers for evidence-based guidance for the conduct of interviews with people from different cultures. Through examining cultural differences in human memory and communication and considering specific contextual challenges for investigative interviewing through the lens of culture, this review and associated commentaries highlight the scope for considering culture and human diversity in research on, and the practice of, investigative interviewing with victims, witnesses, and other sources. Across 11 commentaries, contributors highlight the importance of considering the role of culture in different investigative interviewing practices (e.g., rapport building, questioning techniques) and contexts (e.g., gender-based violence, asylum seeking, child abuse), address common areas of cultural mismatch between interviewer–interviewee expectations, and identify critical future routes for research. We call for an increased focus in the investigative interviewing literature on the nature and needs of our global community and encourage constructive and collaborative discussion between researchers and practitioners from around the world to better identify specific challenges and work together towards evidence-based solutions.     

Personality Profiles of Women in Multiple Abusive Relationships

Personality profiles of women with multiple abusive relationship histories (N = 42) were compared to either abused women with 1 abusive relationship (N = 33) or a control group (N = 52) on the Coolidge Axis II Inventory (Coolidge & Merwin, 1992, J. Pers. Assess. 59: 223–238), a self-report measure based on the Diagnostic and Statistical Manual of Mental Disorders (DSM-IV; American Psychiatric Association, 1994). Women with multiple abusive relationships had higher rates and greater levels of dependent, paranoid, and self-defeating personality disorders than women in the other 2 groups. Women in multiple abusive relationships had significantly more depression, and women in this group with posttraumatic stress disorder (PTSD) had significantly more personality disorders than women with single abusive relationships with PTSD. Women in single abusive relationships did not exhibit more psychopathology than women in the control group with matched marital status. Theoretical and methodological issues, and treatment recommendations are discussed.
