Ultrafast STR Separations on Short‐Channel Microfluidic Systems for Forensic Screening and Genotyping

There are situations in which it is important to quickly and positively identify an individual. Examples include suspects detained in the neighborhood of a bombing or terrorist incident, individuals detained attempting to enter or leave the country, and victims of mass disasters. Systems utilized for these purposes must be fast, portable, and easy to maintain. DNA typing methods provide the best biometric information yielding identity, kinship, and geographical origin, but they are not portable and rapid. This study details the development of a portable short‐channel microfluidic device based on a modified Agilent 2100 bioanalyzer for applications in forensic genomics. The system utilizes a denaturing polymer matrix with dual‐channel laser‐induced fluorescence and is capable of producing a genotype in 80 sec. The device was tested for precision and resolution using an allelic ladder created from 6 short tandem repeat (STR) loci and a sex marker (amelogenin). The results demonstrated a precision of 0.09–0.21 bp over the entire size range and resolution values from 2.5 to 4.1 bp. Overall, the results demonstrate the chip provides a portable, rapid, and precise method for screening amplified short tandem repeats and human identification screening.     

Case report of a criminal dismemberment in Northeast Brazil

Several decomposed body parts were received for examination by the Forensic Anthropology section of the Medico-legal Institute of João Pessoa, Paraíba, Brazil. The portions of the lower and upper limbs, ribs, vertebrae, and a skull were thoroughly examined. The biological profile indicated a male individual with an estimated age range between 23 and 57 years and a mean age of 35.2 years (SD = 9.4; phase IV, Suchey-Brooks). The skeleton showed injuries caused by sharp force and sharp-blunt force trauma that affected all body segments. Macroscopically, the lesions are mainly in the diaphyseal segments of the long bones, sacrum, pelvis, mandible, maxilla, scapulae, sternum, vertebrae, the distal epiphysis of the left fibula, and the distal epiphysis of the left tibia displayed characteristics compatible with injuries produced perimortem. It was not possible to determine the cause of death. DNA analysis resulted in a positive identification. Because of common difficulties faced in forensic practice, it is often not possible for forensic anthropologists to go to the crime scene, X-ray or body scanners are frequently unavailable, and the victim’s medical and/or dental records are sometimes absent. These difficulties make identification ultimately depend on genetic analysis, which is more time-consuming than other identification methods. Despite this, bone fragment examination in dismemberment cases is a complex task. Forensic Anthropology can shed light on cases involving the identification of dismembered remains, which are challenging because of the number of traumatic injuries, as well as different injury patterns, on bones.     

Climate-smart agriculture: what is it good for?

Leading international organisations presently argue that a transition to ‘climate-smart agriculture’ (CSA) is an obligatory task to ensure food supply for an anticipated nine billion people by 2050. Despite the rubric’s newfound importance, the conceptual underpinnings of CSA are often left unclear. Focusing on the World Bank’s framework, this paper critically interrogates the principles and concepts that underpin CSA. It argues that while CSA provides greater policy space for more holistic approaches to agriculture, it nonetheless operates within an apolitical framework that is narrowly focused on technical fixes at the level of production. This depoliticised approach to the global food system tends to validate existing policy agendas and minimise questions concerning power, inequality and access. By highlighting four strong tensions that permeate the CSA framework, the paper extols the need to greatly widen the scope of debate. To this end, it proposes an alternative ‘climate-wise’ framework to foreground the inherently political dimensions of food and agriculture in an era of climatic change.
