Based on the 52 SNP-plex developed by the SNPforID Consortium, we designed two 10-plex to study single nucleotide polymorphisms (SNPs) for human identification and to establish its usefulness in paternity casework. This 20 autosomal SNP set was studied in 56 paternity investigation cases from South Portuguese resident population, also analyzed with 17 Short Tandem Repeats (STRs). Results obtained with both methodologies were consistent with each other, except for one case where the alleged father could not be excluded by SNPs. No mutation was found in the SNP loci, whereas a mismatch in STRs was detected. The use of SNPs as a complement to the analysis of autosomal STRs in paternity casework can result in paternity index and paternity probability values equivalent or higher than those obtained with more STR loci, but with lower costs. This study shows that instead of using additional STR loci, the analysis of 20 autosomal SNPs, as a complement technique to standard methodologies, is an appealing alternative in paternity investigation cases. 相似文献

Analysis of control mitochondrial DNA (mtDNA) hypervariable regions is sometimes the only available method to study hair evidence in forensic casework although being a laborious technique. Nowadays there is a huge interest in new genetic markers such as single nucleotide polymorphisms (SNPs) to type degraded forensic samples. For that purpose, a 10-Plex mitochondrial SNP for haplogroup typing, chosen from several SNP studies and useful to study the most common populations in our laboratory was applied in forensic casework. Hair shafts from three forensic cases with different ethnic backgrounds were studied with mtDNA sequencing and compared with mitochondrial SNPs (mtSNPs) study. Coding mtSNP typing prior to sequencing can allow for a rapid screening in forensic casework, which is emphasized in the first two cases. Moreover, in cases in which mtDNA sequencing fails, mtSNPs can still be detected. This 10 SNP loci multiplex provides a less expensive and simpler method for mitochondrial typing compared to control region mtDNA sequencing, especially when used as a fast screening method. 相似文献

Y-STR mutational rate estimation is very important for the correct evaluation of typing results in forensic casework and specially kinship genetic studies. In this work we studied 95 Southern Portuguese Caucasian father/son pairs in order to estimate mutational rates for the 17 Y-STRs multiplex used in routine casework. In a total of 1615 allele transfers three single step mutations were detected in DYS385a, DYS439, and DYS448, with an estimated mutation rate of 10,526 × 10−3 (95%CI 0.265 × 10−3 to 20.788 × 10−3). The estimated average mutation rate is 1.858 × 10−3 (95%CI 8.08 × 10−4 to 2.908 × 10−3). It would be important to characterize more father/son pairs in order to estimate more reliable allele specific mutation rates for the most widely used Y-STRs markers in forensic genetics. 相似文献

Forensic strategies commonly are proceeding by analysis of short tandem repeats (STRs); however, new additional strategies have been proposed for forensic science. Thus, this article standardized the high‐resolution melting (HRM) of DNA for forensic analyzes. For HRM, mitochondrial DNA (mtDNA) from eight individuals were extracted from mucosa swabs by DNAzol reagent, samples were amplified by PCR and submitted to HRM analysis to identify differences in hypervariable (HV) regions I and II. To confirm HRM, all PCR products were DNA sequencing. The data suggest that is possible discriminate DNA from different samples by HRM curves. Also, uncommon dual‐dissociation was identified in a single PCR product, increasing HRM analyzes by evaluation of melting peaks. Thus, HRM is accurate and useful to screening small differences in HVI and HVII regions from mtDNA and increase the efficiency of laboratory routines based on forensic genetics. 相似文献

Recent advances in nanotechnology applied in forensic sciences have contributed to consider new approaches including chemical evaluation of latent fingermarks. Significant improvement to the detection of small organic molecules has been reached with matrix-free methods associated to laser desorption/ionization mass spectrometry. The present study investigated the application of mesocellular siliceous foam (MCF) as an ionizing agent for laser desorption/ionization (LDI-MS) analysis of fingermarks as a proof of concept research. Fingermarks from three different donors were deposited directly onto a MALDI target plate and α-CHCA matrix solution, MCF ethanolic suspension or MCF/magnetic powder mixture were used for treatment. Microscopy characterization of MCF support showed particles with irregular morphology and variable sizes, and a unordered porous surface with pores diameter ranging from about 10 to 20?nm. Results showed less intense peaks in the spectra produced by the MCF support (control). Analysis of fingermarks showed ions related to endogenous and exogenous molecular components, including possible lipids from human sebum and quaternary ammonium cations commonly present in cosmetics. Promising and reproducible results were obtained for the fingermarks dusted with the MCF/magnetic powder mixture. Considering the forensic applications of nanomaterials for the analysis of small molecules in biological samples by matrix-free LDI techniques, the advantages of silica based materials should be further investigated. 相似文献

Conflict is manifest as promotion of creationism and its euphemisms in school curricula, and science denial, recently in the U.S. Government. Unification, however, is an emerging reality for the science/religion relationship, driven by convergent evolution of each toward saving our shared planet from ourselves. No longer can religion deny science, nor science overlook the power of religious institutions toward achieving our common existential goal. The losses caused by worker theft is one of the most concerning security problems for corporations. Private policing of the workplace is central to cutting down on such losses and keeping up profits. One of the most frequently used methods in such policing is searching workers. Despite the importance of searches and their potential for intrusion into individuals’ right to privacy, the normative bases for and limits to the use of this power have so far been little studied. The aim of this paper is to analyze the legal foundations and limits imposed by the Brazilian State so that private security guards can conduct searches in the workplace. The analysis is based on a qualitative study of Brazilian labor law and a qualitative/quantitative study of 376 judicial decisions on searches collected randomly in two Brazilian states between 2010 and 2013. The data shows that searches can be conducted in the workplace based on the employer’s right to manage production and protect their property. It also indicates the existence of relatively flexible limits to searches. Only a minority of the courts impose restrictions on searches similar to those set for police officers. Most judges allow more extensive searches than those permitted within the scope of public justice systems. The consequences of these findings are discussed. 相似文献

A survey was conducted of a sample of AAPL members to determine their opinions on the inclusion of controversial ethical guidelines for forensic psychiatry. Members appear to appreciate the need to consider traditional Hippocratic values as at least one consideration in their functioning as forensic psychiatrists. They appear to balance their duties to an evaluee with duties to society and the legal system and to appreciate the responsibilities of multiple agency. Support was shown for interpreting ambiguities in AAPL's current guidelines in the directions indicated by most of this survey's proposed guidelines. 相似文献
