Multicolor Real-Time PCR Genotyping of ABO System Using Displacing Probes

Abstract:  Rapid and informative ABO genotyping has become increasingly popular in forensic use. We developed a multiplex real-time polymerase chain reaction (PCR) approach to genotype ABO major groups and subgroups. Seven differently fluorophor-labeled displacing probes for O¹(261delG), A(261G), A(796C/803C), B(796A/803C), O² (802G>A), A² (1059delC), and A² (1009A>G) were combined in one or two PCRs to determine either ABO major groups or subgroups. The method correctly detected 13 reference DNA samples. A blind test of 237 samples resulted in complete agreement with their phenotypes, and 110 of these 237 samples as well as with PCR-SSP method. The whole analysis could be finished in less than 100 min at substantially low material cost and the template DNA ranging from 0.16 to 500 ng per reaction could be quantitatively detected. Despite the limited informativeness of ABO genotyping, the developed methods could find application in rapid and inexpensive screening of forensic settings.

用GC／ECD方法分析海洛因中毒尿液吗啡代谢物

目的　考查尿检材中海洛因的代谢物吗啡和单乙酰吗啡的液液萃取条件、三氟乙酰化和气相色谱电子捕获检测 (GC/ECD)条件。方法　以烯丙吗啡为内标 ,氯仿∶异丙醇 (9∶1)为液相萃取剂萃取尿中的吗啡和单乙酰吗啡 ,采用MBTFA衍生化 (三氟乙酰化 ) ,GC/ECD检测。结果　尿中加样相对回收率吗啡 89% ,单乙酰吗啡 75 % ,最小检测量吗啡 5 0ng ,单乙酰吗啡 10 0ng。通过实验兔的中毒实验 ,对尿检材进行了分析。 结论　所建立的萃取与检测方法分析海洛因中毒尿检材中的吗啡准确、灵敏 ,可用于海洛因的吸毒检验

The Evolution of American ＂New Politics＂

Since the Presidential election in 2012, the ＂fiscal cliff＂ issue has plagued the United States. Although this issue relates to the economy, it reflects the unprecedented political polarization brought by the ＂New Politics＂, an escalating conflict between ＂Red America＂ and ＂Blue America＂. The ＂New Politics＂ refers to the intensifying and increasingly ill-tempered rivalry between the Republicans and Democrats, which has seen a decline in presidential authority, a deepening of divisions in society, and a shrinking of the political middle ground. In the past, the tension between the two main political parties was regarded as the motive power of U.S. politics, a source of dynamism and innovation. Now, partisan conflict is preventing both sides from seeking common interests, and seems to have become a struggle only for party interests.

中越边贸经济交流历史悠久,两国边境居民服务与商品交流频繁,为了满足边境货品买卖及交易活动的结算需求,“货币市场”也随之自发形成。本文分析了越南农业农村发展银行之发展现状、中越边贸发展与结算业务状况,对越南农业农村发展银行边贸结算服务进行了综合评价,并提出了完善发展其边贸结算服务的措施。

改革开放以来,我国青年的社会参与意识不断增强,经历了以反思与寻觅为特征的探索式参与、以认同与担当为特征的主体参与,和以冷静、务实、自主、逐步为特征的全方位参与的发展阶段。考察青年参与的历史可以发现,青年社会参与热情高涨是改革开放的伟大成果,中国特色社会主义事业发展需要青年社会参与,执政党应该重视青年社会参与,对待青年社会参与既要放手锻炼又要保持安定团结的政治局面。     

关于马克思主义大众化问题的思考

马克思主义本身是一个科学的、整体的、开放的体系,马克思主义大众化也不是一项孤立的活动和进程,而是一项体系性活动和进程。所以,关于马克思主义大众化,须从多个角度和层次去考察,它至少包括马克思主义大众化的缘起、现实路径、深层意义及困境和反思等几个方面的有机统一。马克思主义大众化之多维审视有利于推动马克思主义大众化整体性进程。
