Development and validation of the AmpFlSTR MiniFiler PCR Amplification Kit: a MiniSTR multiplex for the analysis of degraded and/or PCR inhibited DNA

DNA typing of degraded DNA samples can be a challenging task when using the current commercially available multiplex short tandem repeat (STR) analysis kits. However, the ability to type degraded DNA specimens improves by redesigning current STR marker amplicons such that smaller sized polymerase chain reaction (PCR) products are generated. In an effort to increase the amount of information derived from these types of DNA samples, the AmpFlSTR MiniFiler PCR Amplification Kit has been developed. The kit contains reagents for the amplification of eight miniSTRs which are the largest sized loci in the AmpFlSTR Identifiler PCR Amplification Kit (D7S820, D13S317, D16S539, D21S11, D2S1338, D18S51, CSF1PO, and FGA). Five of these STR loci (D16S539, D21S11, D2S1338, D18S51, and FGA) also are some of the largest loci in the AmpFlSTR SGM Plus kit. This informative nine-locus multiplex, which includes the gender-identification locus Amelogenin, has been validated according to the FBI/National Standards and SWGDAM guidelines. Our results demonstrate significant performance improvements in models of DNA degradation, PCR inhibition, and nonprobative samples when compared to the AmpFlSTR Identifiler and SGM Plus kits. These data support that the MiniFiler kit will increase the likelihood of obtaining additional STR information from forensic samples in situations in which standard STR chemistries fail to produce complete profiles.     

The Conjunctive Analysis of Case Configurations: An Exploratory Method for Discrete Multivariate Analyses of Crime Data

Derived from comparative approaches in both qualitative and quantitative research, the current study describes a simple exploratory technique for the multivariate analysis of categorical data. This technique is referred to as the conjunctive analysis of case configurations. After describing the logic and underlying assumptions of this conjunctive method, it is applied and illustrated in the study of the federal sentencing of drug offenders. The relative value of this conjunctive approach for purposes of exploratory data analysis and its overall utility as a method for confirmatory research are also discussed.

Supporting farmer participation in formal seed systems: lessons from Tharaka,Kenya

This article examines contemporary challenges of formal seed sector participation for resource-poor farmers in Tharaka and engages in a wider discussion of national seed policy and formal seed sector development in Kenya. While many farmers reported utilising modern seed varieties developed by the formal seed sector, the majority of these were introduced through seed aid and maintained through seed saving, supporting seed system integration strategies. Building on these findings, the article discusses ways in which national seed policy in Kenya might be refined to better meet national and regional development goals focused on decreasing the incidence of hunger and poverty.     

Separated by Bars or Dollar Signs? A Comparative Examination of the Financial Literacy of Those Incarcerated and the General Population

This study examines the level of financial literacy of inmates in Arkansas correctional institutions. Furthermore, it compares the financial knowledge, planning, and practices between not only white and non-white inmates but also between males within and outside of penal institutions. Specifically, this research combines primary data on the financial realities of those within correctional institutions and existing statistics on the public to examine the relationship between demographics, banking history, use of non-traditional lenders, and financial literacy. While prior literature on the public is extensive, research on the financial literacy of individuals currently incarcerated is sparse. Findings indicate vast differences between the public and those within penal institutions, particularly in financial knowledge and planning. For our incarcerated sample we find similar disparities between our white and non-white respondents. Last, we find that youth, minority status, and lowered education are predictors of lower financial knowledge, use of predatory lender use, and poor financial planning among inmates. This is crucial because low levels of financial literacy, use of predatory lenders, and poor financial planning often provide barriers to asset accumulation, which increases the probability of incarceration and recidivism.     

Idle musical community: Dischord Records and anarchic DIY practice

Originally formed in 1980 to document the Teen Idle’s Minor Disturbance EP, Dischord Records has continued to document and support the local Washington, DC, hardcore punk music scene for over three decades through an anarchic Do-It-Yourself (DIY) ethic. Dischord’s DIY ethic affords a manner of music documentation, in contrast to production, and community formation that reconfigures the space of dominant commercial production and exchange. Dischord’s business practices, such as forming agreements with bands via handshake instead of legal contract, allow the label to resist consumerism and exploitation of both artists and audiences while fostering a creative space, where profit does not dictate the sound of the music or produce a commodified musical product. Ultimately, Dischord’s political potential is in its ethics to the extent that anarchic DIY practice constitutes a sort of inoperative community of circulation rather than production and profit. The implications of these practices relate to the ways in which music constitutes social and communal relations and its radical possibility of reconfiguring a space outside of commercial exchanges, whether in the political sphere or in the digital realm.
