Bone samples are valuable for examining the cause of death and circumstance leading up to death when body fluids are not available for forensic toxicological analysis. Examined were heat-induced changes in methamphetamine and amphetamine concentrations in femurs removed from methamphetamine-injected mice to determine if the burned bones could be used for toxicology testing. The femurs were heated at 100°C, 300°C, or 500°C for 10 or 30 min. The tissue structure of the heated femurs was preserved at 100°C for 30 min but was destructed at higher temperatures. Methamphetamine and amphetamine were detected in femurs heated at 100°C for 10 min, 100°C for 30 min, and 300°C for 10 min (with methamphetamine and amphetamine concentrations ranging from 0.36 to 35 μg/g and 0.54 to 47 μg/g, respectively). Methamphetamine and amphetamine were detectable when heated above their decomposition temperature as a result of limited heat transfer do to protection provide by the femoral muscle. Thus, the bone could be a useful analytical sample in cases of burn-related deaths, where it is difficult to collect body fluids.  相似文献

More than two decades ago, Marmarou published a valid model for producing diffuse axonal injury (DAI) in rats. Since then, both mild and severe injuries have been obtained by researchers using the original method and a weight of 450 g. However, the diffuse brain injuries produced in rats were only similar to those seen in humans when the rats sustained severe brain injuries. In these cases, rat mortality in the original article was around 50%, and the cause of death was prolonged apnea post-impact. Rat survival after impact is critical for studying the progression of DAI. In order to explain the cause of death in human victims with cranial trauma who do not show gross brain injury, testing for the presence of DAI is essential. Thus, in order to minimize local and cervical injuries to increase rat survival, attention should be paid to the following aspects: a wider head protector disc should be used, the head of the rat should be elevated at the time of impact, and the foam bed should be soft enough to allow the movement caused by acceleration. With our modified method, rat survival increased by 30% compared to the original model (80% versus 50%). Moreover, 85.7% of rats demonstrated DAI after 24 h of survival. With these modifications, injuries appear in the same locations as in humans; thus, the method is suitable for the study of traumatic DAI in humans.  相似文献

While efforts have been made to reduce the pervasive backlog of sexual assault evidence collection kits, the actual laboratory process remains very time-consuming due to the requirement of a differential lysis step before DNA purification, as well as intricate mixture analysis towards the end of the DNA workflow. Recently, an alternative, direct-to-amplification sperm lysis method (using 1 M NaOH) was identified. However, a direct cell lysis method for non-sperm cells has not been identified yet. Thus, the primary objective of this work was to find an alternative method that is quick, inexpensive, and does not require multiple purification steps for the lysis of non-sperm cells in sexual assault samples. In this study, vaginal swab samples were lysed with the control method, prepGEM™, as well as six alternative reagents: alkaline buffer with 25–200 mM NaOH, high-salt stain extraction buffer, modified radioimmunoprecipitation assay (RIPA) buffer, mammalian protein extraction reagent (M-PER™), digitonin buffer, and urea/thiourea buffer. Quantification using Quantifiler® Trio of vaginal and semen lysates revealed that the alkaline (25 mM NaOH) and M-PER™ methods were efficient for the lysis of vaginal epithelial cells without substantial sperm cell lysis. Following quantification, analysis of STR profiles from vaginal lysates revealed that the M-PER™ method showed promising results across all metrics examined, including the percentage of detected STR alleles, mean peak heights, peak height ratio, and interlocus balance. Thus, this method was recommended as an alternative to the traditional differential lysis method for non-sperm cells given its ability to produce amplification-ready lysates without any DNA purification step.  相似文献

In order to detect latent fingerprints that could be damaged by liquid or powder reagents, non-destructive processes such as gaseous reagents have been developed. In this report, we propose the use of fine mist generated when hot vapor of high-boiling-point liquids is rapidly cooled by surrounding air for fingermark detection. Octyl acetate (OA), 2-phenoxyethanol (2PE), and methyl decanoate (MD) were found to efficiently produce mist when heated to 230°C. By combining these liquids with p-dimethylaminocinnamaldehyde (DMAC) and cyanoacrylate (CN), our team demonstrated effective fluorescence staining of cyano-treated fingermarks using DMAC/OA misting or DMAC/2PE misting, and one-step fluorescence detection of latent fingermarks without cyanoacrylate treatment using DMAC/OA/CN misting or DMAC/MD/CN misting. Fingermark fluorescence was efficiently observed by excitation with a blue LED light (max. wavelength 470 nm) equipped with an interference filter and passing through a 520 nm long-pass filter. We successfully obtained fluorescent images from fingermarks on several substrate materials using the developed misting method.  相似文献

Drawing forensic conclusions from an image or a video is known as “photographic content analysis.” It involves the analysis of an image, as well as objects, actions, and events depicted in images or video. In recent years, photographic depictions of objects suspected as illegal firearms have substantially increased, appearing on CCTV surveillance footage, captured by mobile phones and shared on social media. However, the law in Israel states that a person can be charged with illegally possessing a firearm only if it can be proven that the object is capable of shooting with lethal bullet energy. This becomes more challenging in cases where the firearm was not physically seized, and the evidence exclusively consists of images and video. In this study, photographic content analysis was applied to images and video where objects suspected as commercial or improvised firearms had been depicted. An image and event sequence reconstruction video databases of both firearms and replicas were created in order to better define firearm-specific functional morphological features. We demonstrate that it is possible to classify an object as a firearm by analyzing the functional, and not only the esthetic, morphology in images and video. It is also shown that event sequence reconstruction in video may be used to infer that an object suspected as a firearm has the capacity to shoot by confirming the occurrence of a shooting act or shooting process. Thus, photographic content analysis may be used to forensically establish that an object depicted in an image or a video is a firearm by ruling out other known scenarios, and without physically seizing it.  相似文献

The last few years have witnessed the change in the modalities of smuggling of synthetic cannabinoid receptor agonists (SCRAs) by impregnating them in mail envelopes and fast parcels. Considering the aforementioned scenario, it is important to develop a portable technique to identifying SCRAs through packages. The purpose of this research was to detect SCRAs impregnated into substrates of paper using attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy. Three SCRAs that included 5F-PB-22, AB-FUBINACA and AKB-48 were purchased from Sigma Aldrich. The three SCRAs and four cutting agents were impregnated into paper of variable thickness at four concentrations (10, 15, 20, and 25 mg/mL). Spectra were collected over the wavenumber range of 650–4000 cm−1 using ATR-FTIR spectroscopy and were exported to Matlab 2020b where data analysis was applied. The FTIR spectral data was able to show the three SCRAs could be detected on paper using ATR-FTIR spectroscopy and quantitatively modeled using the partial least squares regression algorithm. Principal component analysis showed separate clustering for the compounds that crystallized (5F-PB-22, AB-FUBINACA and caffeine) onto the papers surface from those impregnated into the bulk of the paper (AKB-48 and procaine) with the latter situated near blank papers in score plots. In summary, ATR-FTIR spectroscopy has proven to be a successful non-destructive technique in detecting and quantifying a selection of SCRAs impregnated into paper.  相似文献

Expanding bullets are preferred by law enforcement because of their wounding potential and ability to avoid over-penetration which could result in unintended targets being struck by bullets that perforate their intended targets. Expansion failure for jacketed hollow point (JHP) bullets is commonly attributed to several causes including damage to the bullet's cavity, velocity loss, bullet destabilization and materials from intermediate targets filling the bullet's cavity which can cause expansion failure when the bullet subsequently impacts a soft, fluid-based target such as human tissue or ballistic gelatin. In this study, JHP bullets were fired into ballistic gelatin after passing through selected intermediate targets representing items common to shooting incidents. Velocity loss and bullet destabilization were not factors that contributed to the JHP bullets that experienced expansion failure; however, materials obstructing the bullets' cavities and damage to the bullets' cavities were considered causes for some of the JHP bullet expansion failures. It was determined through this research that most of the target materials caused JHP bullet expansion failure when shored against the ballistic gelatin, but when placed at distances of 7 ft. from the ballistic gelatin, bullets fired through the same target materials did expand. This original and unique study produced findings that are of significant value to shooting incident reconstruction experts and other forensic professionals as shooting incidents can call into question a victim's proximity to a wall or door when a bullet(s) perforated such a target material.  相似文献

Cocaine and methamphetamine remain highly abused drugs in the United States due to their euphoric effects. This study examines classical stimulant casework, defined as cases positive for methamphetamine and/or cocaine, received by the Toxicology Laboratory and the Drug Analysis Laboratory at the Dallas County Southwestern Institute of Forensic Sciences from local law enforcement agencies and/or the Office of the Medical Examiner (OME) between January 1, 2017, and December 31, 2022. Methamphetamine positivity increased from 10.4% to 20.3% in the Toxicology Laboratory over the 6 years, whereas cocaine positivity remained relatively stable at approximately 17%. Similarly, in the Drug Analysis Laboratory, the methamphetamine positivity rate changed from 24.8% to 33.2%, whereas cocaine identification remained stable at approximately 20%. Blood concentrations of methamphetamine in OME cases ranged from 10.1–42,740.0 ng/mL while they were lower in DWI casework ranging from 10.2–2385.0 ng/mL. The blood concentration trends of cocaine were similar to methamphetamine, with OME casework ranging higher (10.0–24,501.0 ng/mL) than DWI casework (10.2–371.6 ng/mL). Polydrug use was evident for both methamphetamine and cocaine in postmortem cases, and the top three most frequently co-occurring drug/drug class were opioids/opiates, cannabinoids, and ethanol. The results from this study aid in the understanding of historical usage trends of cocaine and methamphetamine in Dallas County and how those trends have changed over time as newer stimulant drugs have emerged.  相似文献

Forensic gait analysis is used to visually and quantitatively analyze information regarding the appearance and style of walking that can be presented as evidence in the court. The demand for analyzing CCTV pedestrian footage in video surveillance has been increasing. The dependence of the accuracy of semiautomatic silhouette-based analysis, often used in forensic science, on the differences in the viewing directions is a very challenging issue that is yet to be resolved for real case applications. Currently, the different viewing directions used in comparison footage significantly decrease the accuracy of same person analysis when using the silhouette-based method, often used in the Japanese forensic science domain. A calibration-based method was previously prosed to resolve this problem, but it requires performing an elaborate measurement procedure at the camera installation site for an accurate analysis. In this study, we propose a novel in-silico silhouette-based analysis method that significantly expands the number of viewing direction pre-set settings to 900 from the 24 used in the previous method. Several software tools have been developed to ensure that all the procedures can be executed on a computer. The experimental results confirm that the accuracy of the proposed method is comparable to that of the calibration-based method. Furthermore, the practical comparison results from actual consultation confirmed the effectiveness of the proposed method under existing viewing direction differences. We therefore anticipate that the proposed method will be beneficial for improving the analysis accuracy in real cases and therefore serve as a substitute of the previous method.  相似文献
