A molecular genetic approach for forensic animal species identification   总被引:7,自引:0,他引:7  
This study investigated potential markers within chromosomal, mitochondrial DNA (mtDNA) and ribosomal RNA (rRNA) with the aim of developing a DNA based method to allow differentiation between animal species. Such discrimination tests may have important applications in the forensic science, agriculture, quarantine and customs fields. DNA samples from five different animal individuals within the same species for 10 species of animal (including human) were analysed. DNA extraction and quantitation followed by PCR amplification and GeneScan visualisation formed the basis of the experimental analysis. Five gene markers from three different types of genes were investigated. These included genomic markers for the beta-actin and TP53 tumor suppressor gene. Mitochondrial DNA markers, designed by Bataille et al. [Forensic Sci. Int. 99 (1999) 165], examined the Cytochrome b gene and Hypervariable Displacement Loop (D-Loop) region. Finally, a ribosomal RNA marker for the 28S rRNA gene optimised by Naito et al. [J. Forensic Sci. 37 (1992) 396] was used as a possible marker for speciation. Results showed a difference of only several base pairs between all species for the beta-actin and 28S markers, with the exception of Sus scrofa (pig) beta-actin fragment length, which produced a significantly smaller fragment. Multiplexing of Cytochrome b and D-Loop markers gave limited species information, although positive discrimination of human DNA was evident. The most specific and discriminatory results were shown using the TP53 gene since this marker produced greatest fragment size differences between animal species studied. Sample differentiation for all species was possible following TP53 amplification, suggesting that this gene could be used as a potential animal species identifier.  相似文献

This longitudinal study of 134 female high school seniors investigated the relationship between attachment cognitions and school and work functioning two years later. School functioning was indexed by interviewer-rated strain, stressful events, academic performance, and self-reported attitudes toward college. Work functioning was indexed by interviewer-rated strain, stressful events, and self-reported attitudes toward work. Attachment cognitions were found to be correlated with school strain, number and stress of school-related events as well as self-reported satisfaction, stress, and problems meeting deadlines. Attachment cognitions were found to be correlated with work strain, work satisfaction, tendency to overcommit, and performance anxiety. Most of these relationships persisted even when initial symptomatology was accounted for. These findings lend support to the hypothesis, derived from attachment theory, that secure attachment cognitions provide a secure base that allows the late adolescent to negotiate the challenges of college and work successfully.This research was supported by a PHS National Research Service Award to the first author, and by a grant from the W. Proponents of "reinvention" assert that it will produce governments that are more agile, better able to adapt to rapidly changing conditions, and able to produce public services at much lower cost. However, attempts to make government "work better and cost less" have had limited success. Policy-makers have consequently invested their creative energy in another class of institutional innovations: segregated funding strategies, which protect agencies and programs from the impact of fiscal restraint by excluding them from normal budgeting processes. The proliferation of budget "lockboxes" may actually introduce new rigidities into the public sector and erode democratic influence in policy-making. On the other hand, there may be some circumstances in which these devices can be justified as a method of ensuring that critical long-run interests receive appropriate attention.  相似文献

Proponents of justice for animals often argue that non-human animals have an interest in liberty. Furthermore, they usually claim that this animal interest in liberty is intrinsic rather than instrumental; that is to say, liberty is regarded to be good for animals in itself, irrespective of its contribution to the achievement of other goods, such as pleasure. For this reason they argue that legislating to improve welfare standards in zoos, circuses, laboratories and agriculture is inadequate. Instead, they claim that such practices are analogous to human slavery, necessarily harmful and must be abolished. In this article I refute this assertion and claim that for most animals, their interest in liberty can only ever be instrumental. In doing so I outline and reject two different arguments in favour of an intrinsic animal interest in liberty: first, that liberty is an intrinsic interest of animals because they possess preference autonomy; and second, that it is good for animals to be free, where freedom is defined as the ability to exercise one's natural functionings. I conclude that most animals do not possess an intrinsic interest in liberty because they are not autonomous in the relevant sense; that is, they cannot frame, revise and pursue their own conception of the good. If my conclusion is correct, this would have important effects on our obligations to non-human animals. I end the article by introducing some of these possible implications. Specifically, I propose that for most animals, our obligations do not consist of liberating them, and that it might be permissible to use and interfere with animals more often than other proponents of justice for animals have suggested. If this is the case, not all of the ways we keep and use animals need to be abolished. Nevertheless, many need to be reformed to improve welfare standards.  相似文献

Short placements can provide an important and meaningful experience of politics in action for students who are unable to benefit from the generally lengthy placements currently on offer. This article is based on the findings of the three-year HEFCE-funded project The Scholarship of Engagement for Politics and explores the potential of short, local, research-based placements, which are embedded in modules, supported by online journaling and effectively assessed, to enrich students' understanding of politics and enhance their employability.  相似文献

Abstract

Efforts to pursue ‘deep integration’—agreeing to international rules governing domestic policies to mitigate their adverse trade effects—have been pivotal to the politicization of trade policy. The contributions to this special issue focus on different political dynamics associated with recent high-profile efforts at deep integration. Collectively, they analyse the Transatlantic Trade and Investment Partnership (TTIP), the Comprehensive Economic and Trade Agreement (CETA), the Transpacific Partnership (TPP) and the Japan–European Union Economic Partnership Agreement (JEEPA) negotiations. The special issue, therefore, focuses on extreme examples of deep integration in order to illuminate new political dynamics. This introductory article introduces the concept of ‘deep integration’ and explores how it has been pursued in historical and contemporary trade negotiations. It also relates recent attempts at deep integration to the rise of populist anti-globalization movements. In light of these discussions, this article introduces the contributions to the issue. It concludes by considering whether the politics associated with TTIP and CETA in Europe represent the future of trade policy.  相似文献

Abstract

When the Transatlantic Trade and Investment Partnership (TTIP) negotiations were underway, its politics were distinctively transnational. Numerous alliances between European and American industry associations advocated an ambitious agreement to mitigate the effects of conflicting rules. Some civic interest groups also closely cooperated to shape the agreement, while a broad, loose transatlantic coalition of civic interest groups opposed it. The extent of transnationalism in TTIP was greater than what had come before in the transatlantic relationship and what is evident in contemporaneous analogous trade negotiations. This article argues that while the salience of a trade negotiation affects whether societal actors mobilize, it is not sufficient to prompt transnational cooperation. Rather transnational cooperation stems primarily from what the actors are seeking to achieve and whether they have a motive and opportunity to work together. By clarifying the conditions under which transnational cooperation is more likely, this article informs the emerging literature on the new politics of trade. By making the case that the motives to mobilize and cooperate require distinct analysis, it contributes to the literature on transnationalism.  相似文献
