National Institute of Standards and Technology SRM 2372 human DNA quantitation standard has been produced to support the need for a human-specific DNA quantitation standard in forensic casework and calibration of new quantitative polymerase chain reaction (qPCR) assays. The conventional DNA concentration has been assigned with one of the U.S. National Reference UV/Visible Spectrophotometers, assuming an absorbance of 1.0 at 260 nm equals 50 ng/μL of double stranded DNA. In addition, an interlaboratory study has been conducted, to verify that the SRM 2372 materials perform well in currently used DNA quantitation assays by the forensic DNA community. Each unit of SRM 2372 consists of three well-characterized DNA extracts. Component A is a single-source human male material derived from blood. Component B is a multiple-source human female material derived from blood. Component C was purchased as a purified unsheared genomic human DNA (Sigma-Aldrich Co., St. Louis, MO) obtained as a lyophilized human genomic extract and has both male and female donors. SRM 2372 is intended to enable the comparison of DNA concentration measurements across time and place. Manufacturers can use SRM 2372 to validate the values assigned to their own reference materials. Individual forensic laboratories can use SRM 2372 to validate DNA quantitation methods and to verify the assigned concentration of in-house or commercial DNA calibration standards.  相似文献

Abstract: The emergence of the “New Public Management” (NPM) and its faith in markets led governments to search for alternative methods in the delivery of public services. One of the most popular methods was privatization. The rationale behind the choice of privatization is based on what Charles Wolf describes as “non‐market failure.” This article argues that the market may not be as efficient as its proponents have asserted, especially when there is a monopoly over service delivery. This has been the case in many municipalities, in some developed countries, where privatization of water service delivery has reverted to public delivery. Using the City of Hamilton's experience with its water and wastewater services as an example, the authors' finding is that the nature of the good to be delivered is essential in determining whether the “market” or the “public” provides the best method of service delivery. Sommaire : L'émergence de la Nouvelle gestion publique (NGP) et sa foi dans les marchés ont conduit les gouvernements à rechercher des méthodes de rechange pour la prestation des services publics. L'une des méthodes les plus populaires est la privatisation. La justification du choix de la privatisation est fondée sur ce que Charles Wolf décrit comme un ≪“échec du non marché”≫. L'article fait valoir que le marché pourrait ne pas être aussi efficace que ses adeptes l'ont prétendu, en particulier lorsqu'il existe un monopole dans la prestation des services. Cela fut le cas dans de nombreuses municipalités de certains pays développés, où la privatisation de la prestation de leurs services en eau est repassée à la prestation publique. In a modern, plural society, there can be no settled agreement on the concrete legal content of a country's constitution. The idea of the constitution is nonetheless pivotal in contemporary, liberal-minded theories of political justification, such as the ones advanced by Jürgen Habermas and John Rawls. Justification in these theories depends finally on "constitutional patriotism," a consciously shared sentiment arising from an ethical assessment of their country by the country's people, according to which the country credibly pursues a certain regulative political ideal for which the constitution stands.  相似文献

The problem is simply stated, but resists solution. We begin with the classical historiographic images of Old Regime societies of South Asia and suggest that beneath the surface of abstract formulations for and against these images, unrecognised difficulties of assumption and method allow them to persist in the contemporary imagination. This not only hampers understanding of historic societies, taken individually, but prevents proper assessment of their comparative significance on an international plane. The relationship between these images of discontinuity and similar sociological images of European peasantry is not coincidental. The errors of the latter are demonstrable through an abundant historiography; the burden of this essay is directed towards a solution to the former. We provide an empirically‐based structural study of social relationships in the countryside of Maharashtra in the 150 years preceding European occupation: a) to begin the accumulation of examples that contradict the received images; b) to provide an analysis of one Old Regime society for its own comparative sake. By examining the social distribution of certain types of right and of access to control of land, we demonstrate the existence of powerful forms of social dominance transcending the frontiers of the village, and strongly influencing the organisation of social, economic and political life in the countryside. Whilst we focus on a specific structural problem (namely, the conception of social totality and its internal ordering), the conclusion and footnotes permit the actual dynamism of the forms of relationship studied to emerge in incipient statement on social change under the old order.?  相似文献
