Detection of a primer-binding site polymorphism for the STR locus D16S539 using the Powerplex 1.1 system and validation of a degenerate primer to correct for the polymorphism

Quality assurance samples submitted from the NCSBI as part of a contract with TBTG to outsource DNA Database samples showed unexpected discrepancies for the locus D16S539 when all other loci yielded identical results. Discrepancies observed included allele drop out and an imbalance in sister alleles with samples returned from TBTG. This led to a comprehensive review of the technical procedures used between the two laboratories to determine the cause of the discrepancies noted for the locus D16S539, since both laboratories were using the PowerPlex 1.1 typing kit from the Promega Corporation. The NCSBI and the TBTG utilize different extraction methods (organic extraction vs. FTA) and amplification conditions (AmpliTaq vs AmpliTaq Gold), respectively, so the exact cause of discrepancy observed was not immediately apparent. Experiments at the NCSBI associated the observed allele drop out and the imbalance of the sister alleles with the use of AmpliTaq Gold and a hot start procedure. Sequencing data revealed that a point mutation resides on the D16S539 primer-binding site that reaches polymorphic levels in African-American populations. This led to the development of a degenerate primer by the Promega Corporation to detect "missing" alleles when AmpliTaq Gold is used. The degenerate primer was then thoroughly tested to show its efficacy in detecting the "true" D16S539 profile when used.     

Primary Attachment to Parents and Peers during Adolescence: Differences by Attachment Style

This study examines the nature of adolescent attachment to parents and peers during adolescence. A projective measure was used to classify 99 11th and 12th grade students into secure, insecure dismissing, and insecure preoccupied attachment groups. Respondents identified their primary attachment figure by nomination and by rating the level of attachment support they received from mothers, fathers, best friends, and boy/girlfriends. On average, parents and peers were equally likely to be identified as primary attachment figures but individual preference was strongly tied to attachment style. Secure adolescents significantly favored mothers over best friends, boy/girlfriends, and fathers. Although secure adolescents with romantic partners rated mothers lower on attachment support, none of the adolescents from this group nominated a boy/girlfriend as their primary attachment figure. In contrast insecure adolescents indicated a strong preference for boy/girlfriends and best friends as their primary target for attachment and nearly a third of dismissing adolescents identified themselves as their primary attachment figure. Findings are discussed in terms of individual differences in attachment during adolescence.     

Halogenated solvent interactions with N,N-dimethyltryptamine: formation of quaternary ammonium salts and their artificially induced rearrangements during analysis

The psychoactive properties of N,N-dimethyltryptamine (DMT) 1a are known to induce altered states of consciousness in humans. This particular attribute attracts great interest from a variety of scientific and also clandestine communities. Our recent research has confirmed that DMT reacts with dichloromethane (DCM), either as a result of work-up or storage to give a quaternary N-chloromethyl ammonium salt 2a. Furthermore, this was observed to undergo rearrangement during analysis using gas chromatography-mass spectrometry (GC-MS) with products including 3-(2-chloroethyl)indole 3 and 2-methyltetrahydro-beta-carboline 4 (2-Me-THBC). This study further investigates this so far unexplored area of solvent interactions by the exposure of DMT to other halogenated solvents including dibromomethane and 1,2-dichloroethane (DCE). The N-bromomethyl- and N-chloroethyl quaternary ammonium derivatives were subsequently characterised by ion trap GC-MS in electron and chemical ionisation tandem MS mode and by NMR spectroscopy. The DCE-derived derivative formed at least six rearrangement products in the total ion chromatogram. Identification of mass spectrometry generated by-products was verified by conventional or microwave-accelerated synthesis. The use of deuterated DCM and deuterated DMT 1b provided insights into the mechanism of the rearrangements. The presence of potentially characteristic marker molecules may allow the identification of solvents used during the manufacture of controlled substances, which is often neglected since these are considered inert.     

Female and male sex offenders: a comparison of recidivism patterns and risk factors

Few studies have empirically validated the assertion that female and male sex offenders are vastly different. Therefore, utilizing a matched sample of 780 female and male sex offenders in New York State, the current study explored differences and similarities of recidivism patterns and risk factors for the two offender groups. Results suggested that male sex offenders were significantly more likely than female sex offenders to be rearrested for both sexual and nonsexual offenses. However, limited differences in terms of risk factors between female and male sex offenders were found.

Teaching technology decision making for product design and development: A university course on technology assessment and technology transfer

This paper explores how these supplemental, “contextual” skills may be taught and reports on an experimental, fairly successful upper-division course at Pennsylvania State University in the fall of 1995.     

The Sources of Terrorist Financing: Theory and Typology

Terrorism costs money. Although the costs of specific operations may be relatively inexpensive, terrorist organizations require much larger budgets in order to function. But how do terrorist groups acquire funding? What explains the variation in the particular sources used by different groups? This article develops a theory of terrorist financing that identifies criteria by which we can evaluate the different sources of terrorist funding, particularly in terms of their advantages and disadvantages to the terrorist group. These criteria are then applied across a typology of four primary types of terrorist financing: state sponsorship, illegal activity, legal activity, and popular support.     

State and Territory Government Strategic Plans: Exercises in Managing,Monitoring and Marketing

All but three of the Australian States and Territories have whole‐of‐government, jurisdiction‐wide strategic plans in place and the three exceptions had previously established a state plan in the decade after the first such plans were introduced by Tasmania and Victoria in 2001. Scholarly attention to date has been directed towards discrete aspects of state plans, such as the extent to which they can be seen as exercises in participative democracy or sustainability, rather than with examining state plans for what they explicitly purport to be – strategic plans originating in the core executive of government. We propose that there are three key strategic orientations for Australian state plans: to holistically manage, monitor and market government administration. These orientations may be competing, complementary or overlapping, and can vary over time. The article also proposes that state plans are a significant development in public management and warrant further and more detailed examination.
