Abstract: The PrepFiler? Forensic DNA Extraction Kit enables isolation of genomic DNA from a variety of biological samples. The kit facilitates reversible binding of DNA with magnetic particles resulting in high DNA recovery from samples with very low and high quantities of biological materials: 0.1 and 40 μL of human blood (donor 2) provided 14 and 2883 ng of DNA, respectively. Following the revised SWGDAM guidelines, performance of the developed method was investigated using different sample types including saliva on swabs, semen stains on cotton fabric, samples exposed to environment, samples with polymerase chain reaction (PCR) inhibitors, blood stains (on denim, cotton cloth, and FTA® paper), and touch evidence‐type samples. DNA yields for all samples tested were equal or better than those obtained by both phenol–chloroform extraction and commercial kits tested. DNA obtained from these samples was free of detectable PCR inhibitors. Short tandem repeat profiles were complete, conclusive, and devoid of PCR artifacts. 相似文献

Abstract: This paper presents how jewelry modeling waxes are used in the preparation of tool mark standards from exemplar tools. We have previously found that jewelry modeling waxes are ideal for preparing test tool marks from exemplar tools. In this study, simple methods and techniques are offered for the replication of accurate, highly detailed tool mark standards with jewelry modeling waxes. The techniques described here demonstrate the conditioning and proper use of jewelry modeling wax in the production of tool mark standards. The application of each test tool's working surface to a piece of the appropriate wax in a manner consistent with the tool's design is clearly illustrated. The resulting tool mark standards are exact, highly detailed, 1:1, negative impressions of the exemplar tool's working surface. These wax models have a long shelf life and are suitable for use in microscopic examination comparison of questioned and known tool marks. 相似文献

This review systematically searched UK academic and grey literature in relation to mother and child separation in prison. Attachment theory is referred to in current prison policy for mothers and could provide a framework linking policy and practice. Reviewing grey literature provided an opportunity to explore practice-based literature. Twenty-four academic papers and 51 grey documents were reviewed. Use of attachment theory in the academic literature varied according to discipline, ranging from extensive use to no use. There was greater use of attachment theory in the grey literature. Despite linguistic differences, all documents highlighted the detrimental impact of separation on imprisoned mothers. However, specificity was lacking regarding support for mothers, and staff needs were overlooked. Given its use across the sparse research and practice literature, and its basis for policy, attachment theory could underpin theoretically informed support for imprisoned mothers separated from their infants and staff who support them. 相似文献

Adolescents are constantly connected with each other and the digital landscape through a myriad of screen media devices. Unprecedented access to the wider world and hence a variety of activities, particularly since the introduction of mobile technology, has given rise to questions regarding the impact of this changing media environment on the mental health of young people. Depressive symptoms are one of the most common disabling health issues in adolescence and although research has examined associations between screen use and symptoms of depression, longitudinal investigations are rare and fewer still consider trajectories of change in symptoms. Given the plethora of devices and normalisation of their use, understanding potential longitudinal associations with mental health is crucial. A sample of 1,749 (47% female) adolescents (10–17 years) participated in six waves of data collection over two years. Symptoms of depression, time spent on screens, and on separate screen activities (social networking, gaming, web browsing, TV/passive) were self-reported. Latent growth curve modelling revealed three trajectories of depressive symptoms (low-stable, high-decreasing, and low-increasing) and there were important differences across these groups on screen use. Some small, positive associations were evident between depressive symptoms and later screen use, and between screen use and later depressive symptoms. However, a Random Intercept Cross Lagged Panel Model revealed no consistent support for a longitudinal association. The study highlights the importance of considering differential trajectories of depressive symptoms and specific forms of screen activity to understand these relationships. 相似文献

Establishing a sense of life meaning is a primary facet of well-being, yet is understudied in adolescent development. Using
data from 579 adolescents (53% female) from Latin American, Asian, and European backgrounds, demographic differences in meaning
in life, links with psychological and academic adjustment, and the role of meaning in explaining associations between ethnic
identity and adjustment were examined. Although no generational or gender differences were found, Asian Americans reported
higher search for meaning than Latin and European Americans. Presence of meaning was positively associated with self-esteem,
academic adjustment, daily well-being, and ethnic belonging and exploration, whereas search for meaning was related to lower
self-esteem and less stability in daily well-being. Presence of meaning mediated associations between ethnic identity and
adjustment, explaining 28–52% of ethnic identity’s protective effect on development. Ethnic identity thus appears to affect
adjustment, in part, through its role in fostering a positive sense of meaning in adolescents’ lives. 相似文献

Associations among neighborhood disadvantage, maternal acculturation, parenting and conduct problems were investigated in
a sample of 444 Chinese American adolescents. Adolescents (54% female, 46% male) ranged from 12 to 15 years of age (mean age = 13.0 years).
Multilevel modeling was employed to test the hypothesis that the association between maternal acculturation and adolescents’
conduct problems could be explained by differences in mothers’ reliance on monitoring and harsh discipline. In addition, guided
by segmented assimilation theory, measures of neighborhood disadvantage were expected not only to be related to differences
in parenting, but also to moderate the effects of maternal acculturation on parenting. Results indicated that increased maternal
acculturation was related to higher levels of maternal monitoring and lower levels of harsh discipline, which, in turn, were
related to lower levels of adolescents’ conduct problems. Hierarchical linear modeling results revealed that neighborhood
disadvantage was related to lower levels of maternal monitoring. However, neighborhood disadvantage did not moderate the link
between maternal acculturation and parenting practices.

Research on the academic adjustment of immigrant adolescents has been predominately conducted in large cities among established
migration areas. To broaden the field’s restricted focus, data from 172 (58% female) Asian American adolescents who reside
within a non-traditional or emerging immigrant community in the Southeastern US were used to examine gender differences in
academic adjustment as well as school, family, and cultural variables as potential mediators of gender differences found.
Results suggest that girls report significantly higher educational goals, intrinsic academic motivation, and utility value
of school compared to boys. These gender differences are statistically mediated by ethnic exploration and family processes,
most prominently, family respect. School connectedness and perceived discrimination are also associated with academic adjustment
at the bivariate level, suggesting that academic success may be best promoted if multiple domains of influence can be targeted. 相似文献

Parent and Peer Pathways to Adolescent Delinquency: Variations by Ethnicity and Neighborhood Context
Effects of ethnicity and neighborhood quality often are confounded in research on adolescent delinquent behavior. This study examined the pathways to delinquency among 2,277 African American and 5,973 European American youth residing in high-risk and low-risk neighborhoods. Using data from a national study of youth, a meditational model was tested in which parenting practices (parental control and maternal support) were hypothesized to influence adolescents' participation in delinquent behavior through their affiliation with deviant peers. The relationships of family and neighborhood risk to parenting practices and deviant peer affiliation were also examined. Results of multi-group structural equation models provided support for the core meditational model in both ethnic groups, as well as evidence of a direct effect of maternal support on delinquency. When a similar model was tested within each ethnic group to compare youths residing in high-risk and low-risk neighborhoods, few neighborhood differences were found. The results indicate that, for both African American and European American youth, low parental control influences delinquency indirectly through its effect on deviant peer affiliation, whereas maternal support has both direct and indirect effects. However, the contextual factors influencing parenting practices and deviant peer affiliation appear to vary somewhat across ethnic groups. Following an intentional or accidental bio-warfare agent (BWA) release, environmental sample analysis is absolutely critical to determine the extent of contamination. When dealing with nonspore forming BWA (e.g., Yersinia pestis), retention of cell viability is central to such analyses. Even though significant advances have been achieved in DNA sequencing technologies, a positive identification of BWAs in environmental samples must be made through the ability of cells to form colony-forming units upon culturing. Inability to revive the cells between collection and analysis renders such studies inconclusive. Commercial kits designed to preserve the viability of pathogens contained within clinical samples are available, but many of them have not been examined for their ability to preserve samples containing suspected BWAs. The study was initiated to examine the applicability of commercial solutions aiding in retention of Y. pestis viability in samples stored under nonpermissive temperatures, that is, 40 and 37°C. While none of the tested solutions sustained cell viability at 40°C, the results show five out of 17 tested preservatives were capable of supporting viability of Y. pestis at 37°C. 相似文献

Data were obtained from the National Longitudinal Study of Adolescent Health to examine the longitudinal association between
Asian and Pacific Islander (API) adolescents’ perceptions of maternal approval of their sexual activity and contraception
use, and four sexual outcomes during young adulthood. The study includes a nationally representative sample of 1,195 API adolescents.
Logistic regression analyses were used to estimate the associations between predictors (Wave I) and outcomes (Wave III), controlling
for covariates. API adolescents who perceived that their mothers approved of their sexual activities were more likely to have
engaged in sex before age 15, contracted HIV/Sexually Transmitted Diseases, had multiple sex partners, and paid money for
sex during young adulthood. Findings highlight the need for parent–adolescent communication and parental involvement in preventing
negative sexual health outcomes among API adolescents transitioning to young adulthood.
