We investigated the usefulness of a toothbrush as a source of DNA for an unidentified cadaver. Ten toothbrushes were obtained from ten individuals along with their peripheral blood. We recovered from 10 to 430 ng of DNA from all but one of the toothbrushes. All ten toothbrushes, including the one containing no detectable DNA by fluorometry, were typed correctly at all of the loci tested, including nine STRs. Three toothbrushes obtained in two actual deaths also identified two victims and one suspect. Therefore, toothbrushes seem to be useful as a source of evidential DNA for personal identification.  相似文献

Depression is a major cause of suicide among the elderly. Few previous community-based interventions against depression have reduced the suicide rate. This study aims to evaluate outcomes of a community-based program to prevent suicide among the elderly using a quasi-experimental design with a neighboring reference group. The program, including depression screening with follow-up and health education through primary care and public health nursing, was implemented for 10 years in Matsudai town, a rural area of Japan (population 6,015; suicide rate per 10(5) [65-year-olds] for males 290.6, and for females 361.3). Changes in the suicide risk were estimated by the incidence rate ratio (IRR). The female risk of completing suicide in the intervention area was reduced by 70% (age-adjusted IRR: 0.30; 95% CI: 0.14-0.67), while there was no change in the risk for males in the intervention area nor for males or females in the reference area (Kawanishi town: population 9,425; elderly suicide rate for males 212.2, females 151.9). A ratio of the female IRR in the intervention area to that in its prefecture was also estimated at 0.45 (95% CI: 0.19-0.97), showing that the reduction of suicide risk in the intervention area was greater than the historical trend. A community intervention against suicide using management of depression with nonpsychiatric, primary health care would be effective for elderly females, but not males.  相似文献

The denaturant gradient gel electrophoresis (DGGE) method was used in order to simultaneously estimate the genotypes of different factors in a gel plate consisting of one sheet. A genotype analysis of the blood groups (MN, Duffy, Kidd type) and serotype (Gc system) was carried out. DNA samples were extracted from trace deposits which were transferred on adhesive film from a blood trace obtained from a car tire after a fatal car accident. The reference DNA was prepared from the victim's blood. The PCR amplification fragments were amplified from the gene which controlled each blood group. The primers were designed in order to analyze the genotypes with one to three base substitutions in the amplification product. The denaturant concentration limit of the gel for the DGGE method to detect each genotype of the blood groups (MN, Duffy, Kidd type and Gc system) and other conditions of electrophoresis were performed according to previously methods.The each genotype of the blood groups and the Gc system were all simultaneously distinguished in one plate.  相似文献

Political scientists have long been concerned about the validity of survey measurements. Although many have studied classical measurement error in linear regression models where the error is assumed to arise completely at random, in a number of situations the error may be correlated with the outcome. We analyze the impact of differential measurement error on causal estimation. The proposed nonparametric identification analysis avoids arbitrary modeling decisions and formally characterizes the roles of different assumptions. We show the serious consequences of differential misclassification and offer a new sensitivity analysis that allows researchers to evaluate the robustness of their conclusions. Our methods are motivated by a field experiment on democratic deliberations, in which one set of estimates potentially suffers from differential misclassification. We show that an analysis ignoring differential measurement error may considerably overestimate the causal effects. This finding contrasts with the case of classical measurement error, which always yields attenuation bias.  相似文献

Acute toxicity of the combustion products from various fibers was evaluated in animal experiments. The materials used were cotton, gauze, acetate, rayon, polyester (PE), polychlal (PC), polyvinylidene chloride (PVDC), silk, wool, polyacrylonitrile (PAN), and modacryle (MA). Rats, mice, and rabbits were exposed to gases released from these materials, heated with an electric heater. In experiments with rabbits animals inhaled gases through a tracheal cannula under urethane narcosis. As indices for toxicity, the time at which animals were impaired severely, was used in rat experiments while the death time was used in experiments with rabbits and mice. The concentrations of O2 and CO in the exposure room were determined continously, blood COHb and cyanide values were also estimated. Gases from fibers containing nitrogen impaired severely and killed the animals earlier than any other materials. Blood analyses revealed the presence of high values of cyanide in PAN, MA, and silk experiments. HCN was considered to be responsible for the high toxicity of gases from these materials. In the case of wool, despite of high toxicity of its combustion products, blood cyanide and COHb values were not very high. Gases from cotton, gauze, and rayon impaired severely and killed the animals relatively early in the exposure period. Toxicity of combution products from these materials was attributable to CO on the basis of gas and blood analyses. Acute toxicity of PE and acetate gases, being less than that of cotton, gauze, rayon under the present experimental conditions, was explained mainly by CO. Neither severe impairment nor death occurred during exposure in PC and PVDC experiments.  相似文献

The estimation of chronological age has been performed by various methods in forensic science. Among these, racemization methods, which are based on the age-dependent non-enzymatic changes of L-form amino acids to D-form mainly using aspartic acid, are one of the most reliable and accurate methods to date. Separation of enantiomers is generally performed by gas chromatography or high performance liquid chromatography. Various tissues with low metabolic rates have been applied for this purpose. In addition, single proteins purified from these target tissues are also applicable. In this brief review we describe this method in detail, noting points of caution, as well as the advantages and disadvantages of the different target tissues. In addition, special attention is given to the correlation rates obtained between chronological age and enantiomer ratios. Currently, based on accuracy of estimated age, simplicity of the method, time required, and reproducibility, tooth dentin is considered one of the best target tissues. Alternatively, analysis of osteocalcin and elastin have also provided accurate and reproducible results.  相似文献

In this study, we investigated time-dependent expression of matrix metalloproteases (MMP)-2, MMP-9, chemokine CC motif ligand (CCL)-2, CCL-3, CCL-5, IL-1β, IL-6, and TNF-α mRNA at the skin injury site and sought their forensic potentials during the skin wound repair process. The tested wound ages in 42 mouse skin wounds were distributed at 0d, 1d, 3d, 5d, 7d, 10d, and 14d, respectively and then followed by real-time polymerase chain reaction. Ultimately, MMP-2 played an important role in the inflammation phase. On the contrary, MMP-9 became involved at a later phase during wound healing. Meanwhile, CCL-2 and CCL-3 were active throughout almost all of the process. However, CCL-5 mRNA had no significance. Collectively, an MMP-9/MMP-2 ratio of over 0.84 indicated that skin wound healing age was strongly 5 days or less. So elevated gene expressions of cytokines and chemokines in different phases of wound ages implied that combined exploration could make wound age determination more accurate and objective.  相似文献
